摘    要:建立检测黄连、黄柏中生物碱(小檗碱、巴马汀、药根碱)含量的MECC-AD测定方法。在自组装仪器上,以0.3 mm(i.d.)碳圆盘微电极为工作电极,在1.150 V(Ag/AgCl)的检测电位下,以含有0.008 mol·L-1的胆酸钠(SC)溶液与3%甲醇溶液的0.2mol·L-1H3BO3-0.05 mol·L-1Na2B4O7·10H2O缓冲液(pH值=8.45)为运行液;当毛细管长度65 cm(i.d.25μm),分离电压10 kV,三种待测物在15 min内完全分离。本方法测定三种生物碱的线性范围分别为(0.37~32.6、0.98~41.7、0.25~25.9μg·mL-1);检出限分别为(0.15、0.36、0.08μg·mL-1)。结论:该方法已成功应用于黄连、黄柏等中药材中3种生物碱含量的测定。

Abstract:Micellar electrokinetic capillary chromatography (MECC) with amperometricl detection (AD) was used to determine three alkaloids (berberine, palmatine and jatrorrhizine) in Phellodendri Cortex and Coptidis Rhizoma. At the laboratory -built system a carbon disk electrode was used as the working electrode and the optimal detection potential was 1. 150 V ( versus Ag/AgCl). At 10 kV of applied voltage and a 65cm ( i.d. 25 μm ) length capillary ,the three alkaloids were completely separated within 15 rain in 0.2 mol ·L^-1 HaBO3 - 0.05 mol ·L^-1 Na2B4O7 · 10H2O buffer ( pH = 8.45) including 0.008 mol ·L^-1 sodium cholate (SC) and 3% (v/v) methanol. The linear ranges for berberine, palmatine and jatrorrhizine are 0. 37 - 32.6, 0.98 - 41,7 and 0.25 - 25.9 μg·mL^-1; respectively, and the limit detections are 0.15, 0.36, 0. 08μg·mL^-11. This method has been successfully applied to the determination of the three alkaloids in Phellodendri Cortex and Coptidis Rhizoma.
