摘    要:目的:探讨胃癌细胞凋亡过程中细胞内游离Ca^2+(「Ca^2+」i)浓度和环磷酸腺苷(CAMP)浓度的变化特点及意义。方法:选择顺铂诱导胃癌细胞凋亡,采用光镜、电镜、琼脂糖凝胶电泳和流式细胞仪检测凋亡;Fura-2荧光负荷技术测「Ca^2+」i,竞争性蛋白结合法测cAMP浓度。结果:顺铂浓度为2μg/ml作用于胃癌细胞24小时出现典型的凋亡改变,即凋亡小体、细胞皱缩、染色质固缩和片段化,DNA梯状

Abstract:To study the change of intracellular-free calcium(2 ]i) and cAMP levels in the process of gastric carcinoma cellular apoptosis Apoptosis induced by cisplatin in gastric cancer cell was investigated by applying light microscopy, electron microscopy, agarose gel electrophoresis and flow cytometry i were determined by applying Fura-2 fluorescein load technic cAMP levels were determined by applying competitive protein binding method Gastric carcinoma cell treated with cisplatin (2ug/ml)for 24 hours expressed typical change of apoptosis, such as apoptosis body, cell shrinkage, condensation and fragmentation of chromosomes, DNA ladder bands, apoptosis peak in flow cytometry ]i in the process of gastric carcinoma cellular apoptosis was significantly higher than that in the control,especially at the beginning stage of apoptosis cAMP level was higher than that in control In conclusion, i and cAMP levels were significantly higher than those in the control in the process of gastric carcinoma cellular apoptos
