尼莫地平和丹参对豚鼠缺血-再灌注耳蜗损伤的影响

摘    要:目的探讨尼莫地平和丹参对缺血-再灌注豚鼠耳蜗损伤的改善机制.方法耳廓反射灵敏的健康豚鼠48只,随机分为正常组、缺血再灌注6 h组、缺血再灌注6 h用药组(于缺血前30 min腹腔注射尼莫通500 μg*kg-1,丹参5 g*kg-1)及缺血再灌注6 h用生理盐水组(腹腔注射等量生理盐水).各组动物于手术前及处死前分别测试ABR、耳蜗标本HE染色和电镜观察、耳蜗组织超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量测定、耳蜗组织iNOS免疫组化.结果与缺血再灌注6 h用生理盐水组相比,用药组ABR各波潜伏期缩短、阈值降低,毛细胞结构基本正常,耳蜗MDA含量及SOD活力、iNOS在耳蜗表达差异有统计学意义.结论尼莫地平和丹参合用可以有效改善缺血-再灌注听力损伤.

Abstract:Aim:Nimodipine combined with radix salviae miltiorrhizae were used in the ischemia reperfusion injury model of guinea pigs to study their protective effects on cochlea. Methods: Forty eight healthy guinea pigs with positive Preyer's reflx were allocated into 4 groups: the normal control group, the ischemia reperfusion group, the ischemia reperfusion group received nimodipine combined with radix salviae miltiorrhizae intraperitoneally before the onset of ischemia, and the group received normal saline intraperitoneally (NS group). All the changes of ABR, the morphological features of cochlea, the content of MDA,SOD activity, and iNOS immunoactivity were studied. Results:Compared with NS group, the latencies of all the waves were shorten and thresholds decreased for ABR of drug group ( P <0.05). Compared with NS group MDA contents decreased and SOD activity increased ( P <0.05), iNOS expression in cochlea increased ( P < 0.01 ). Conclusions: The combination of nimodipine and radix salviae miltiorrhizae can prevent the cochlea in guinea pigs from the ischemia reperfusion injury efficiently.
