摘    要:根据已知小麦1Bx14基因序列设计一对特异引物,对HMW -GS在Glu -Bx14位点已知的7个小麦品种进行了PCR扩增,结果表明,凡具有1Bx14的品种都能扩增出4 19bp的特异性带,与SDS -PAGE电泳结果一致。利用这一特异性标记对山东推广的2 0个小麦品种进行检测,发现7个品种带有1Bx14基因。研究表明,与SDS -PAGE相比,PCR标记更加快速、简便和准确。

Abstract:A pair of specific primers designed based on the known wheat 1Bx14 gene sequence were applied to amplify 7 wheat cultivars with known HMW-GS in Glu-Bx14 locus. The results indicated that 419bp specific bands were appeared in all wheat cultivars amplified with 1Bx14, which was in coincidence with the results of SDS-PAGE. 20 wheat cultivars in Shandong province were screened applying the specific marker, and 7 cultivars were found out with 1Bx14 gene. Compared with SDS-PAGE, PCR marker was more rapid, handy and exact.
