Abstract:A biomimetic method for the efficient extraction of ginsenoside was developed via the in vitro simulation of gastrointestinal tract environment.Two kinds of solvent,biomimetic gastric juice and biomimetic intestinal juice,were prepared to extract ginsenosides from ginseng.The extract conditions were investigated and optimized in detail.A quantitative method based on high performance liquid chromatography-triple quadrupole mass spectrometer in multiple reaction monitoring mode was established to detect the concentrations of extracted ginsenoside Re,Rg1,20(S)-Rf,Rb1,Ro,Rc,Rb2 and Rd.The extraction efficiency of the developed biomimetic method was further compared with that of an optimized ultrasonic extraction method.The quantitative results showed that the extraction concentration of ginsenosides in gastric juice was slightly higher than that in intestinal juice.The optimized conditions were as follows:refluxing at 37.0℃for 60 min with a ratio of liquid to solid of 15:1.The biomimetic method was validated and showed good repeatability,precision,and accuracy,and could extract ginsenoside from ginseng effectively.Under the optimized conditions,the concentration of ginsenosides extracted by biomimetic method was 10.4%-56.9%higher than that by ultrasonic method.Biomimetic method doesn’t require organic solvent and is suitable for the rapid extraction of ginsenoside.
