贝伐单抗抑制兔角膜碱烧伤新生血管的实验研究

摘    要:目的通过点眼和结膜下注射贝伐单抗两种不同的给药途径,抑制兔角膜碱烧伤后的新生血管,探讨该药的这两种给药方式的疗效。方法普通白色家兔50只,取2只为正常对照组,其余48只右眼碱烧伤后随机分为三组,每组16只:A组为碱烧伤对照组,B组为点眼组,用贝伐单抗(5mg/ml)点眼,3次/d,C组为结膜下注射组,用贝伐单抗(2.5mg/0.1ml)结膜下注射。在不同的时间点(第1、4、7、14d)测量角膜新生血管的生长情况及免疫组化检测角膜血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白的表达。结果 B、C组的CNV相对于A组得到明显抑制,CNV的长度和面积及VEGF蛋白的表达差异有统计学意义,而B组和C组之间没有明显差异。结论两种用药方法对于抑制角膜新生血管没有明显差异。

Curative effect of bevacizumab on neovascularization in alkali burn cornea of rabbit

Abstract:Objective Through eyedrop application and subconjuctival injection of bevacizumab administered in two different ways to inhibit cornea neovascalarization in alkali-burned rabbit cornea to explore different methods of drug efficacy.Method 50 normal white rabbits,taking two for the normal control group,the remaining 48 were divided into 3 groups after alkali burn:A group of alkali burn control group,B group were instilled with bevacizumab(5mg/ml) for three times daily,C group were subconjunctival injected with bevacizumab(2.5mg/0.1ml).At different time points(1,4,7,14 days) measured growth of CNV and immunohistochemical detection of corneal vascular endothelial growth factor(VEGF) protein.Results B,C group compared with A group,CNV was significantly curbed,CNV area and the length and the expression of VEGF protein were significantly different statistically,while the B group and C group no significant difference between.Conclusion Two application methods for inhibition of corneal neovascularization was no significant difference.
