双光气急性中毒家兔肺神经内分泌细胞的改变

摘    要:首次观察到家兔急性双光气中毒性肺水肿时,肺神经内分泌细胞内的颗粒减少。应用银染技术和图象分析方法,在石蜡切片上检测出中毒动物肺神经内分泌细胞数量下降,吸光度降低,细胞内颗粒面积与所在细胞的面积比也减少。提示肺神经内分泌细胞可能在双光气中毒性肺水肿的发生和发展中起重要作用。

Changes of pulmonary neuroendocrine cells following exposure to diphosgene in rabbits

Abstract:This paper dealt with the relation of pulmonary neuroendocrine cells (NE cells) to diphosgene induced pulmonary edema for the first time. Twenty young New Zealand rabbits, weighing around 0.6 kg, were exposed to diphosgene for 20 min at the dose of 650-720 μg / L. Another 5 rabbits were used as controls. Five animals were killed each time at 1, 2, 4 and 8 h, after the intoxication of diphosgene. Five cubes of lung tissue were taken from the right lower lobe of each animal. The specimens were fixed in Bouin's fluid, prepared by routine paraffin section and stained with the Grimelius and Pascual's silver nitrate method. The sections were observed under a light microscope and treated with an automatic image-analysis instrument. In the experiment group, the number of silver stained positive NE cells was markedly declined with a very significant difference as compared with the controls (p<0.001). The optical density of positivecells was prominently declined (p< 0.001), and granule area / cell area of positive NE cells was also distinctly reduced (p < 0.05).The above findings reveal that the pulmonary NE cells could directly be affected by diphosgene and, then, release their intracytoplasmic granules, a peptide which may induce the reactivity of local blood vessels and regulate the bronchial function. Thus, the NE cells can be regarded as the organ possessing double functions of both receptor and effector.This experiment points out that the pulmonary NE cells may be a clue for reviewing the mechanism of pulmonary edema after exposure to diphosgene and providing the available means in the clinical treatment.
