胃癌组织中环氧合酶-2、核转录因子-κB蛋白的表达及相关性

目的 通过检测环氧合酶-2(COX-2)与核转录因子-κB(NF-κB)在慢性胃炎、不典型增生及胃癌组织中的表达情况,探讨两者间相关性及其与胃癌发生、发展的关系.方法 采用免疫组织化学染色法对28例慢性浅表性胃炎(CSG)、23例慢性萎缩性胃炎伴肠化(CAG+IM)、19例不典型增生(AH)及53例胃癌(GC)组织标本进行检测,观察COX-2与NF-κB蛋白的表达情况.结果 胃癌组织中COX-2及NF-κB蛋白表达的阳性率显著高于慢性浅表性胃炎和慢性萎缩性胃炎伴肠化两组,差异有统计学意义(P<0.05).COX-2和NF-κB在胃癌中的高表达与浸润深度及有无淋巴结转移有关(P<0.05),且两者间存在显著正相关性(γs=0.437,P<0 01).结论 COX-2及NF-κB两者相互协同参与胃癌形成的全过程,不仅是其早期事件,而且在胃癌发展晚期亦起重要作用. Abstract： Objective To detect the expression of cyclooxygenase-2, nuclear factor-kappaB in human chronic gastritis, atypical hyperplasia, gastric carinoma and to identify the correlation between COX-2 and NF-κB in the development and progression of human gastric carcinoma. Methods Expression of COX-2 and iNOS protein was examined by immunohistochemistry in 28 cases chronic superficial gastritis(CSG), 23 cases chronic atrophic gastritis with intestinal metaplasia(CAG+IM), 19 cases atypical hyperplasia(AH) and 53 cases gastric carcinoma(GC) tissue. Results There were significant differences (P<0.05) when GC group compared with CSG, CAG+IM,separately. The expression of COX-2 and NF-κB protein in gastric carcinoma tissue was correlated with invasion deepth and lymph node metastasis(P<0.05). The expression of COX-2 was positively correlated with that of NF-κB(γs=0.437,P<0.01). Conclusions The overexpression of COX-2 and NF-κB might contribute to the development and progression of gastric carcinoma in coordination.
