压力性尿失禁大鼠泌尿生殖道组织及耻尾肌一氧化氮、神经肽Y的表达及意义

摘    要:目的探讨神经递质NO、NPY在压力性尿失禁大鼠泌尿生殖道组织及耻尾肌中的表达及其与压力性尿失禁发生的关系。方法建立压力性尿失禁动物模型,取建模成功的SD雌性大鼠15只作为实验组,未经任何处理的正常大鼠20只作为对照组。处死后取尿道、阴道前壁及耻尾肌,HE染色,观察上述标本的组织形态学差异,并行免疫组化染色方法检测NO与NPY的分布及其表达差异。结果HE染色显示,实验组大鼠的尿道、阴道前壁、耻尾肌与对照组相比,组织萎缩变薄,肌纤维排列疏松、紊乱。免疫组化染色显示,NO与NPY在大鼠尿道、阴道前壁、耻尾肌表达,与对照组相比,实验组表达减弱,两组相比差异有显著性(P〈0.05)。结论NO、NPY可能与压力性尿失禁发生有关。

Abstract: Objective ] To investigate the expression of neurotransmitter NO and NPY in the pubocoecygeus and genitourinary tract of rats with stress urinary incontinence and the relationship of the occurrence of stress urinary incontinence. Methods ] Established animal model of stress urinary incontinence and choosing 15 SUI SD rats as experimental group and the other 20 normal SD rats as control group , both groups were killed and taken away thole pelvic tissues including the urethra , anterior vaginal wall and pubococeygeus . The samples were examined by HE staining to observe the histologieal and morphological differences and to detect the expression levels of NO and NPY by immunohistochemieal methods. Results ] HE staining showed that urethra , anterior vaginal wall and puboeoceygeus of the experimental group have the atrophy thinning, muscle fiber arranged loosely and disordered compared with the control group. Immunohistochemistry showed that NO and NPY expressed in the urethra, anterior vaginal wall and pubococeygeus. Expression in rats model group decreased significantly compared with control group (P〈0.05). Conclusion ] NO and NPY may be related with the occurrence of SUI.
