摘    要:研究了大孔吸附树脂精制中药栀子中栀子苷的工艺条件,以栀子苷含量为考察指标,比较了不同吸附树脂、吸附流速、洗脱流速等因素对精制栀子苷的影响,表明HPD-100树脂精制栀子苷的最佳工艺条件:吸附流速为2.0 mL/min,洗脱流速为1.0 mL/min,梯度洗脱,收集30%乙醇水溶液洗下来的洗脱液,干燥后,得到纯度为90.75%的栀子苷产品。此工艺可较好地精制中药栀子中的重要有效成分栀子苷,为工业化生产提供了方法依据。

Abstract:To screen the optimum macroporous resin and technical conditions for the adsorption and purification of geniposide from the Chinese native medicine gardenia,the static and dynamic adsorption/desorption methods were used,and the separation and purification process was evaluated by measuring the concentration of geniposide by UV spectrophoto-meter.The HPD-100 macroporous resin was the most effective compared with other macroporous resins.The optimum conditions were screened as follows,The sample containing geniposide was passed through a column packed witi HPD-100 resin at the flow rate of 2.0 mL/min.After adsorp-tion,the resin was gradient-eluted by using aqueous ethanol solution at the flow rate of 1.0 mL/min.The eluate with 30% ethanol was collected and vacuum-distilled to remove ethanol and water and then was vacuum-dried to obtain a solid product.Under these conditions,a product with 90.75% of geniposide was obtained.It can be feasible to be utilized in industrial production of geniposide.
