黄芪饮片质量评价和分级标准探讨

摘    要:目的:通过分析黄芪饮片外观指标与内在成分关系,得出黄芪饮片质量评价和制订其等级标准的参考依据。方法:采用Agilent ZORBAX SB-C18色谱柱,流动相为乙腈-水(32∶68),蒸发光散射检测器检测,氮气流速为2.7 L·min^-1;漂移管温度为105℃;增益为1,测定黄芪甲苷成分含量。采用Waters XBridge C18色谱柱,流动相为乙腈-0.2%甲酸水溶液(梯度洗脱),二极管阵列检测器检测,检测波长为260 nm,测定4种黄酮类成分含量。利用SPSS分析软件对饮片外观指标和各有效成分内在指标进行数理统计分析。结果:黄芪饮片外观指标与黄芪甲苷含量呈显著的负相关,即黄芪饮片直径和质量越大,黄芪甲苷含量越小;与毛蕊异黄酮葡萄糖苷、毛蕊异黄酮、芒柄花苷和芒柄花素4种黄酮类成分含量不呈相关性,但各黄酮类成分之间呈显著的、不同的正负相关性。结论:本实验结果为评价黄芪饮片质量和制订其等级标准提供参考。

Abstract:Objective:To acquire the reference basis for evaluating the quality of slices of Astragali Radix and formulating its grading standards by analyzing the relationship between the appearance index and the internal composition of slices of Astragali Radix.Methods:The content of astragaloside IV was determined by Agilent ZORBAX SB-C18 column,mobile phase was acetonitrile-water(32∶68)with evaporative light scattering detector(ELSD).The nitrogen flow rate was 2.7 L·min^-1,the drift tube temperature was 105℃,the gain was 1.Waters XBridge C18 column was used to determine the contents of calycosin glucoside,calycosin,formononetin and ononin with mobile phase of acetonitrile-0.2%formic acid aqueous solution(gradient elution),and the wavelength of DAD was 260 nm.SPSS software was used to analyze the correlation between appearance indexes and active ingredients of slices of Astragali Radix.Results:The content of astragalosideⅣshowed a significant negative correlation with the diameter and quality of medicinal slices.While the content of four flavonoids ingredients showed no correlation with their diameter and quality.But there were significant positive and negative correlations between four flavonoids.Conclusion:The results of this experiment provide a reference basis for evaluating the quality of slices of Astragali Radix and formulating its grading standards.
