| 摘 要： | 目的探讨端粒酶11-聚体正义寡核苷酸(S0DN)、反义寡核苷酸(ASODN)对肺腺癌细胞株(SPC-A1)的作用。方法脂质体介导的11-聚体正、反义寡核苷酸与肺腺癌低分化SPC-A1细胞株共培养后，MTT法检测细胞生长抑制率；瑞氏染色后观察细胞形态学变化；流式细胞仪检测细胞周期分布及细胞凋亡。结果①ASODN抑制肺腺癌细胞SPC-A1生长，诱导细胞凋亡。②SODN抑制肺腺癌细胞SPC-A1生长，可能通过诱导细胞分化途径。③无义寡核苷酸(NODN)对肺腺癌细胞SPC-A1生长无抑制。结论ASODN和SODN分别通过诱导细胞凋亡和诱导细胞分化途径抑制肺腺癌细胞SPC-A，生长。

| Abstract: | Objective To investigate the effects and mecha nism of sense oligodeo xynucleotides (SODN) and antisense oligodeoxynucleotides (ASODN) on adenocarcino ma cell line (SPC-A 1).Methods After transfecting telomerase 11-base SODN and 11-base ASODN into SPC-A 1 cell line by lipofection, cell growth in hibition rate, cell cycle, and apoptasis rate were detected with MTT and FCM. Th e morphological changes of cells were observed too. Results 1. ASODN inhibited the growth of SPC-A 1 cell and induced cells apoptasis. 2. SODN inhibited the g ro wth of SPC-A 1 cells and induced SPC-A 1 cells to differentiate. 3. NODN didn' t in hibit the growth of SPC-A 1 cells. Conclusions ASODN and SODN could inhibit th e growth of SPC-A 1 cells by inducing cells apoptasis and differentiation respect ively. |
