鼠疫菌对不同剂量和pH值的糖(醇)培养基发酵观察

摘    要:目的观察并探讨鼠疫菌对不同剂量和pH值的糖(醇)培养基发酵结果,以便在工作中找出鼠疫菌对糖(醇)类发酵的理想选择值。方法选取对鼠疫菌有典型鉴定特征的5种糖(醇)类:阿胶糖、鼠李糖、麦芽糖、蜜二糖、甘油,用鼠疫菌生化实验常规制备方法配制糖(醇)基础培养液,按不同剂量(0.5~1.0 g)、不同pH(7.2、7.4、7.6)制成的糖(醇)培养基,接种鼠疫菌,观察其对鼠疫菌发酵影响。结果 1%和pH7.6糖(醇)培养基对鼠疫菌正常发酵结果优于0.5%和pH7.2~7.4的糖(醇)培养基,特别是对阿胶糖和甘油。结论制备糖(醇)培养基时选取剂量1%和pH7.6为鼠疫菌发酵的理想选择值。

Abstract:Objective Observing and discussing the fermentive effect on Yersini pestis in different culture medium and different doses and PH value so that we can find out the ideal value of Yersini pestis in sugar(alcohol) culture medium.Methods Five kinds of sugar(alcohol) with typical morphological feature of Yersini pestis were selected: Donkey-hide gelatine,rhamnose,maltose,melibiose and glycerol.Use conventional biochemical method to prepare sugar(alcohol) culture medium which is suitable for the growth of plague.According to different doses(0.5 ~1.0g) of sugar(alcohol) and pH(7.2,7.4,7.6),preparing different culture medium and vaccinate plague bacteria,then observe the effect of plague bacteria fermentation.Results Culture medium with 1% sugar(alcohol) and pH value 7.2~7.4 is superior to 0.5% sugar(alcohol) and pH value 7.6,especially for Donkey-hide gelatine and glycerol in fermentive effect on Yersini pestis.Conclusion The ideal value of sugar alcohol medium for Yersini pestis is 1% sugar(alcohol) and pH7.6.
