当归多糖硫酸酯抗小鼠L6565逆转录病毒的作用

摘    要:目的 观察当归多糖硫酸酯(APS)抗小鼠L65 65逆转录病毒的作用,以探讨其治疗艾滋病的可能性。方法 用小鼠L65 65病毒感染出生2 4h内的乳鼠建立模型;以RT PCR法测定血浆病毒载量;流式细胞术测定CD4 +,CD8+细胞的百分数,计算CD4 +/CD8+细胞的比值;镜检计数外周血白细胞、红细胞和骨髓有核细胞数;测定脾脏、胸腺的重量并计算脏体系数。结果 与正常对照组相比,模型组小鼠血浆病毒载量较高( 4 1 65±1 98) ,CD4 +细胞百分率( 1 8.2±4 .9)、红细胞、骨髓有核细胞数及胸腺系数显著降低,而白细胞数和脾脏系数显著升高(P <0 .0 5或P <0 .0 1 ,n =1 0 ) ;与模型组相比,APS在1 0和3 0mg·kg- 1 剂量下血浆病毒载量显著降低( 3 3 2 8±1 0 3和3 3 94±1 71 ) ,CD4 +细胞百分率( 5 2 .6±5 .8和5 7.0±5 .5 ) ,CD4 +/CD8+比值( 3 .6±0 .6和3 .4±0 .5 )、红细胞、骨髓有核细胞数及胸腺系数显著升高,白细胞数和脾脏系数显著降低(P <0 .0 5或P <0 .0 1 ,n =1 0 )。结论 APS不仅具有一定的抗病毒作用,而且对免疫功能有一定的调节作用,提示其对艾滋病可能有一定的治疗作用

Abstract:AIM To investigate the antivirus effects of Angelica polysaccharide sulfate (APS) on L6565 murine leukemia virus and discuss the possibility of APS on AIDS therapy. METHODS model was set up by infecting infant mice with L6565 murine leukemia virus. Quantitative RT-PCR method was used to measure viral load in plasma. The percentage of CD4+ and CD8+ lymphacytes was estimated by flowcytometric method and the ratio of CD4+/CD8+ was calculated. The number of WBC, RBC and marrow karyote was counted, index of immune organs was weighed and calculated. RESULTS Compared with control, the viral load in plasma in the mice infected with the virus was at high level (4165±198). The percentage of CD4+ lymphocyte(18.2±4.9), the ratio of CD4+/CD8+, the number of RBC and marrow karyote, and thymus index were significantly decreased in the model mice, while the number of WBC and spleen index were remarkably increased (P<0.05 or P<0.01, n=10). But at 10 and 30 mg·kg-1, compared with model, the viral load in plasma (3328±103 and 3394±171), WBC and spleen index were significantly decreased by APS, while the percentage of CD4+ lymphocyte(52.6±5.8 and 57.0±5.5), the ratio of CD4+/CD8+(3.6±0.6 and 3.4±0.5), the number of RBC and marrow karyote was remarkably increased by APS(P<0.05 or P<0.01, n=10), some of the items even recovered to normal level. CONCLUSION These results suggest that APS have not only antivirus effects, but also immunomodulative effect. APS has a potential therapy effect on AIDS.
