The infectivity and development of four populations of Meloidogyne hapla were compared, at three temperatures, on tomato and two varieties of cucumber. A population from Canada produced few root-galls on cucumber and, except at 24 C, no larvae developed into adult females and produced egg masses. In contrast, a population with 45 chromosomes from America produced many galls on cucumber and small proportions of larvae became females and produced egg masses at 20 and 24 C. At 18 C this population produced no egg masses on cucumber, but a population from Britain and one from America with 17 chromosomes produced more egg masses at this temperature than at 20 or 24 C. Genotypical differences among graminaceous species in release of phytosiderophores and uptake of iron phytosiderophores 总被引:1,自引:0,他引:1
Graminaceous species can enhance iron (Fe) acquisition from sparingly soluble inorganic Fe(III) compounds by release of phytosiderophores (PS) which mobilize Fe(III) by chelation. In most graminaceous species Fe deficiency increases the rate of PS release from roots by a factor of 10–20, but in some species, for example sorghum, this increase is much less. The chemical nature of PS can differ between species and even cultivars.The various PS are similarly effective as the microbial siderophore Desferal (ferrioxamine B methane sulfonate) in mobilizing Fe(III) from a calcareous soil. Under the same conditions the synthetic chelator DTPA (diaethylenetriamine pentaacetic acid) is ineffective.The rate of Fe(III)PS uptake by roots of graminaceous species increases by a factor of about 5 under Fe deficiency. In contrast, uptake of Fe from both synthetic and microbial Fe(III) chelates is much lower and not affected by the Fe nutritional status of the plants. This indicates that in graminaceous species under Fe deficiency a specific uptake system for FePS is activated. In contrast, the specific uptake system for FePS is absent in dicots. In a given graminaceous species the uptake rates of the various FePS are similar, but vary between species by a factor of upto 3. In sorghum, despite the low rate of PS release, the rate of FePS uptake is particularly high.The results indicate that release of PS and subsequent uptake of FePS are under different genetic control. The high susceptibility of sorghum to Fe deficiency (lime-chlorosis) is most probably caused by low rates of PS release in the early seedling stage. Therefore in sorghum, and presumably other graminaceous species also, an increase in resistance to lime chlorosis could be best achieved by breeding for cultivars with high rates of PS release. Benzyladenine-induced changes in the translatable mRNA population in excised cucumber cotyledons were studied. Poly (A)+ RNA was prepared from etiolated cotyledons incubated with or without benzyladenine (BA) for various periods in the dark. Using nonequilibrium pH gradient electrophoresis-SDS polyacrylamide gel electrophoresis and isoelectric focusing-SDS polyacrylamide gel electrophoresis, both basic and neutral proteins translated in vitro were separated. The mass production of juvenile seeds of the sea cucumber,Stichopus japonicus has recently developed by the SeaFarming Center of Saga Prefecture. Methods for the culture ofperiphicic diatoms have been improved. There are three importantsteps in propagating the diatoms. The first step is theenrichment, with the addition of the nutrient salts, undercontrolled light intensity. The second step is washing withhigh pressure seawater and reversal of the plates. The laststep is elimination of copepods, which feed on diatoms, usinga pesticide. Small periphitic diatoms such as Navicula,Amphora, Achnanthes, and Nitzschia are easily culturedat a density of more than one million cells cm–2, andthese diatoms are able to induce larval metamorphosis andserve as a food source for juvenile sea cucumbers. 相似文献

Cotyledons excised from dark-grown seedlings of cucumber (Cucumis sativus L.) were cultured in vitro under UV radiation at different wavelengths, obtained by passage of light through cut-off filters with different transmittance properties. Growth and the synthesis of chlorophyll (Chl) in cotyledons were inhibited and malondialdehyde was accumulated upon irradiation at wavelengths below 320 nm. Exogenous application of scavengers of free radicals reversed the growth inhibition induced by UV-B. Measurement of the fluorescence of Chl a suggested that electron transfer in photosystems was affected by UV-B irradiation. On the basis of these results, the involvement is postulated of active species of oxygen in damages to thylakoid membranes and the growth inhibition that are induced by UV-B irradiation.Abbreviations Chl

Abstract. The patterns of growth in organs curving under the influence of gravity were analysed by time-lapse photography of cress and cucumber hypocotyls which were delimited into 1 mm zones by ion-exchange beads. Geotropic curvature resulted from changes in growth rate on both sides of the organ. Growth inhibition of varying degrees of intensity occurred in all the previously growing zones of the upper (concave) side. An absolute reduction in length due to compression frequently occurred in some zones. Also, in both species growth stimulation was observed on the lower (convex) side. The disparity in growth rate between the upper and lower surfaces varied with time, being more apparent in the subapical region in the first hour of curvature. A later promotion of growth rate on the lower surface subsequently increased the curvature of the more basal zones. Autotropic straightening occurred as a consequence of growth changes, both inhibitory and stimulatory, in the apical zones. These events indicate a polarity of response in which apical zones have precedence over basal zones. 相似文献

The behavioural response of the predatory mite Phytoseiulus persimilis to volatiles from several host plants of its prey, spider mites in the genus Tetranychus, was investigated in a Y-tube olfactometer. A positive response to volatiles from tomato leaves and Lima bean leaves was recorded, whereas no response was observed to volatiles from cucumber leaves, or leaves of Solanum luteum and Solanum dulcamara.Different results were obtained for predators that differed in rearing history. Predators that were reared on spider mites (Tetranychus urticae) on Lima bean leaves did respond to volatiles from Lima bean leaves, while predators that had been reared on the same spider mite species but with cucumber as host plant did not respond to Lima bean leaf volatiles. This effect is compared with the effect of rearing history on the response of P. persimilis to volatile allelochemicals of prey-infested plant leaves. 相似文献

The subcellular localization of lipoxygenase (LOX) from cucumber fruit has been studied. Two methods have been employed to obtain organelles; (1) maceration of the tissue, followed by separation on a linear sucrose gradient and (2) release from protoplasts by osmotic shock, followed by a discontinuous Ficoll gradient. It was possible to obtain high LOX activity in the intact protoplasts from both peel and flesh tissue. However, fewer intact vacuoles were obtained following osmotic rupture than from macerated tissue. Both methods produced more particulate LOX activity from the peel than from flesh tissue, and both showed that this activity was associated with the vacuoles. Cell wall carbohydrate composition and 1-aminocyclopropane-1-carboxylic acid (ACC) content have been determined in chilled (2.5°) and non-chilled (12.5°) cucumber fruit. The major compositional change that accompanied the increased capability for ACC synthesis during chilling was a diminished loss of galactose residues, relative to the loss which occurred at 12.5°. However, the loss of galactose residues increased markedly when fruit were transferred from 2.5° to 20°, and wall galactose levels eventually declined to similar levels in both chilled and non-chilled fruit. Rhamnose, arabinose, xylose, mannose and cellulose content of walls was similar in chilled and non-chilled fruit and did not change substantially upon transfer of fruit to 20°. Upon transfer of chilled fruit from 2.5° to 20°, an increase in the relative amount of galacturonic acid in cell walls occurred; this change did not occur in non-chilled fruit. Thus, chilling stress results in a rapid change in the neutral sugar and galacturonic acid composition of cell wall pectic polysaccharides upon warming. 相似文献
