Microsclerotium formation by six isolates of Verticillium dahliae was studied at different temperatures both in vitro and in Arabidopsis thaliana . In vitro mycelial growth was optimal at 25°C, but microsclerotium formation was greatest at 20°C (two isolates) or 15–20°C (one isolate). Seedlings of A. thaliana were root-dipped in a conidial suspension, planted, and either placed at 5, 10, 15, or 25°C, or left at 20°C until the onset of senescence, after which some of the plants were placed at 5, 10, 15, or 25°C. The amount of microsclerotia per unit of shoot weight was assessed in relation to isolate and temperature. The optimal temperature for production of microsclerotia was 15–25°C. Two isolates each produced about 10 times more microsclerotia than each of the other four isolates. For these isolates, high R 2adj.-values of 0.77 and 0.66 were obtained, with temperature and its square as highly significant (P   < 0.001) independent variables. R 2adj.-values for the other isolates varied between 0.28 and 0.39. Moving plants to different temperatures at the onset of senescence led to microsclerotial densities that were intermediate between densities on plants that had grown at constantly 20°C and plants grown at other temperatures. This suggests that vascular colonization rate and rate of microsclerotium formation are similarly affected by temperature. The senescence rate of plants appeared unimportant except for plants grown at 25°C, which showed the highest amounts of microsclerotia per unit of plant weight in the most rapidly senescing plants.  相似文献

Verticillium lecanii (Fungi: Deuteromycete) blastospores were applied to a chrysanthemum crop by an ULV electrostatically charged rotary atomiser (APE-80). The deposition of spores and subsequent control ofAphis gossypii were compared to high volume hydraulic application. A full rate treatment (2×1013 blastospores per ha.) was applied by the APE-80 at week 1 and reduced spore rates of 1/6th and 1/12th applied by both the APE-80 and the hydraulic sprayer once and twice a week respectively for weeks 1 to 6. Untreated plots served as controls. Initial deposits of spores were higher with the electrostatic sprayer and better distributed with respect to the position of the target aphids. Significantly lower aphid populations were recorded on the electrostatically treated plots in week 4. The single full rate treatment had significantly fewer aphids than the untreated plots from week 3 and all treatments had significantly fewer aphids than the untreated plots from week 5 onwards. The proportion of the aphid population killed byV. lecanii was higher on the electrostatically treated plots until week 6.   相似文献

A wild type strain ofVerticillium lecanii and a mutant strain with increased tolerance to the fungicide benomyl were evaluated in greenhouse experiments for effects on Heterodera glycines populations. Nematodes were applied at 300 eggs and juveniles per 4,550-cm³ pot (two soybean plants in 4,990 g loamy sand per pot) and at both 300 and 10,000 eggs and juveniles per 1,720-cm³ pot (one soybean plant in 2,060 g sand per pot). With 300 nematodes added per pot, both V. lecanii strains significantly reduced nematode populations in loamy sand (fungus applied at 0.02% dry weight per dry weight loamy sand) and sand (0.006% and 0.06% fungus application rates). The mutant strain applied at 0.002% to sand also significantly reduced cyst numbers. When 10,000 nematodes were added per pot, only the mutant strain at 0.06% significantly decreased population. Various media were tested for isolation of the fungus strains from prills, loamy sand, and sand, but the fungi were recovered from few of the greenhouse pots.  相似文献

The pathogenicity of several isolates of the hyphomycete fungi Verticillium lecanii and Metarhizium anisopliae to Frankliniella occidentalis was investigated. Treatment of adult thrips with M. anisopliae resulted in at least 94% mortality at 7 days post-inoculation. In contrast, V. lecanii isolates only gave mortalities of between 20 and 70%. Detailed studies were made on the most virulent isolate of M. anisopliae (275) to determine its efficacy at different doses and temperatures. At 23 C the LC was ca. 3 105 conidia ml-1 after 5 days and the LTs were 50 50 3 and 4.5 days at 10 7 and 106 conidia ml-1 respectively. Temperature influenced fungal virulence to adult thrips; the LT at 18 and 20 C was ca. 4 days and at 23 or 26 C it was ca. 3 days. 50 Larvae were less susceptible to infection than adult thrips (27% versus 100% mortality), presumably due to the inoculum being shed with the exuvium during ecdysis. Conidia of M. anisopliae isolate 275 germinated rapidly on the surfaces of larvae, pupae and adults, with most germlings producing appressoria within 24 h post-inoculation. Fungal elements were present in significant amounts in the body 3 days after treatment.  相似文献

Summary In vitro study showed thatVerticillium dahliae Kleb. grew well in a wide range of acid and alkaline media (viz. pH 3.5 to 10.5). The best growth of the fungus was observed in pH 5.5. Soil pH 3.5 was toxic for growing antirrhinum seedlings. Development of Verticillium wilt of antirrhinum was affected by soil pH. The severity of the disease was greater in alkaline soil conditions compared with acid conditions. Soil of pH 3.5 gave very good control of the symptom expression by the infected plants. Rhizosphere analysis results showed that fungal population with the exception ofPenicillium spp., was drastically reduced in the rhizosphere of the plants grown in acid soil. Although the overall population of fungi was reduced in theV. dahliae infected antirrhinum rhizosphere in acid soil, the population ofPenicillium spp. markedly increased. The antagonistic activity of thePenicillium spp. in the rhizosphere might also have reduced the disease severity. Since the seedlings did not grow properly in very dry and very wet soil, rhizosphere analysis of these soils was not possible. Disease severity was much less in wet soil compared with plants grown in medium moisture level and dry soil, but the plant growth was very poor. Dedicated to the memory of the late Prof. H. K. The arms race between fungal pathogens and plant hosts involves recognition of fungal effectors to induce host immunity. Although various fungal effectors have been identified, the effector functions of ribonucleases are largely unknown. Herein, we identified a ribonuclease secreted by Verticillium dahliae (VdRTX1) that translocates into the plant nucleus to modulate immunity. The activity of VdRTX1 causes hypersensitive response (HR)‐related cell death in Nicotiana benthamiana and cotton. VdRTX1 possesses a signal peptide but is unlikely to be an apoplastic effector because its nuclear localization in the plant is necessary for cell death induction. Knockout of VdRTX1 significantly enhanced V. dahliae virulence on tobacco while V. dahliae employs the known suppressor VdCBM1 to escape the immunity induced by VdRTX1. VdRTX1 homologs are widely distributed in fungi but transient expression of 24 homologs from other fungi did not yield cell death induction, suggesting that this function is specific to the VdRTX1 in Vdahliae. Expression of site‐directed mutants of VdRTX1 in N. benthamiana leaves revealed conserved ligand‐binding sites that are important for VdRTX1 function in inducing cell death. Thus, VdRTX1 functions as a unique HR‐inducing effector in V. dahliae that contributes to the activation of plant immunity.  相似文献

【背景】棉花黄萎病是由大丽轮枝菌(Verticillium dahliae Kleb.)引起的一种世界性病害,近年来对该病害的生物防治因具有环境友好和人畜安全的特性而倍受关注。【目的】筛选棉花黄萎病高效拮抗细菌并对其进行鉴定,为棉花黄萎病的生物防治扩充菌种资源。【方法】采用稀释涂布平板法分离细菌,并进行拮抗细菌的初筛和复筛,通过形态特征、生理生化特征和16S rRNA基因序列分析对筛选到的细菌进行鉴定,确定其分类地位。【结果】初筛分离到535株对病原菌具有拮抗作用的细菌,并选取了108株拮抗细菌进行复筛,最终筛选到了4株优势拮抗细菌。通过形态观察、生理生化特征和16SrRNA基因序列分析,将菌株BHZ-29、SHT-15、SHZ-24和SMT-24分别鉴定为贝莱斯芽孢杆菌(Bacillusvelezensis)、枯草芽孢杆菌斯皮兹仁亚种(Bacillus subtilis subsp. spizizenii)、萎缩芽孢杆菌(Bacillus atrophaeus)和香草芽孢杆菌(Bacillus vanillea)。【结论】获得了4株高效拮抗细菌,并且首次报道了香草芽孢杆菌对棉花黄萎病菌具有抑制作用。  相似文献

对我国自育的108个陆地棉品种的抗黄萎病性进行了研究。在黄萎病发病期内,对黄萎病发病情况进行连续调查,测定产量、考查产量因素并检测纤维品质。利用因子分析法对陆地棉抗黄萎病反应规律进行分析,得出不同时期的黄萎病病指主要与前后3~5个阶段抗病性有关。病情发展主要由4个主因子决定,且第1、2主因子具有较大的方差贡献率。第1主因子(F1)主要与品种7月26日至8月9日的黄萎病病指有关,第2主因子(F2)主要与品种8月20日至9月4日的黄萎病病指有关。利用因子分析结果将108个品种划分为4个类型,前期抗病性较好而后期发病较快的第Ⅰ类品种,其产量较低,单株结铃数、单铃重、衣分均低于其他3类;纤维品质均较差,纤维长度、整齐度、比强度和马克隆值均较其他3类差。前期和后期病指均较低、发病缓慢的第Ⅱ类则小区产量最高,纤维品质处于平均水平。第Ⅲ类品种前期发病较慢,中期发病较快,具有较高的小区产量,单铃重最高;纤维整齐度、比强度和伸长率好于其他3类品种;前期发病较快,中期发病平缓,后期仍具有较高病指的为第Ⅳ类品种,小区产量较低,单株产量、单株结铃数和衣分较高;其他性状处于中等水平。但研究表明,某一阶段具有的抗病性并不能完全代表品种的抗病性。  相似文献

1980—1981年先后自北京、昆明、福州等地双孢蘑菇[Agaricus bisporus (Lange)Sing.]上分离到一种真菌,在PDA上的培养性状与蘑菇轮枝菌(Vertieillium psalliotae Tresch.)和蛛网丝枝霉中国变种[Aphanocladium aranearum(Petch) Gams var sincase J.D.Chen]近 似。镜检可见其兼具上述两种菌的形态。经反复多次纯化,并挑取单孢子培养,肯定了这种真菌是纯菌。在培养基上已保存4年,形态稳定。它具典型蘑菇轮枝菌形态,而其丝枝霉型瓶梗的形成和瘪缩时间、长度、原瓶梗数量、末端瓶梗数及排列密度等,都与蛛网丝枝霉中国变种有一定差异,与丝枝霉属其它种亦不同,故定为两型丝枝霉新种(Aphanocladium dimorphum sp-nov.)。本文详细描述了该新种的形态,井对文甲所提的五种真菌及其与Engyodontium属间的关系进行了讨论。  相似文献
