An integrated genetic linkage map for E. nitens was constructed in an outbred three-generation pedigree. Analysis of 210 RFLP, 125 RAPD and 4 isozyme loci resulted in 330 markers linked in 12 linkage groups covering 1462 cM (n=11 in eucalypts). The 12th linkage group is comprised of only 5 markers and will probably coalesce with another linkage group when further linked loci are located. Co-dominant RFLP loci segregating in both parents were used to integrate linkages identified in the male and female parents. Differences in recombination frequencies in the two parents were observed for a number of pairs of loci, and duplication of sequences was identified both within and between linkage groups. The markers were distributed randomly across the genome except for the RFLPs in linkage group 10 and for some loci showing segregation distortion, which were clustered into three regions of the map. The use of a large number of co-dominant RFLP loci in this map enables it to be used in other pedigrees of E. nitens and forms a basis for the detection and location of QTL in E. nitens and other eucalypt species.  相似文献

Eucalyptus trees are among the most important species for industrial forestry worldwide. However, as with most forest trees, flowering does not begin for one to several years after planting which can limit the rate of conventional and molecular breeding. To speed flowering, we transformed a Eucalyptus grandis × urophylla hybrid (SP7) with a variety of constructs that enable overexpression of FLOWERING LOCUS T (FT). We found that FT expression led to very early flowering, with events showing floral buds within 1–5 months of transplanting to the glasshouse. The most rapid flowering was observed when the cauliflower mosaic virus 35S promoter was used to drive the Arabidopsis thaliana FT gene (AtFT). Early flowering was also observed with AtFT overexpression from a 409S ubiquitin promoter and under heat induction conditions with Populus trichocarpa FT1 (PtFT1) under control of a heat‐shock promoter. Early flowering trees grew robustly, but exhibited a highly branched phenotype compared to the strong apical dominance of nonflowering transgenic and control trees. AtFT‐induced flowers were morphologically normal and produced viable pollen grains and viable self‐ and cross‐pollinated seeds. Many self‐seedlings inherited AtFT and flowered early. FT overexpression‐induced flowering in Eucalyptus may be a valuable means for accelerating breeding and genetic studies as the transgene can be easily segregated away in progeny, restoring normal growth and form.  相似文献

While the pharmacological and toxicological properties of eucalypts are well known in indigenous Australian medicinal practice, investigations of the bioactivity of eucalypt extracts against high mortality diseases such as pancreatic cancer in Western medicine have to date been limited, particularly amongst the genera Corymbia and Angophora. Four Angophora and Corymbia species were evaluated for their phytochemical profile and efficacy against both primary and secondary pancreatic cancer cell lines. The aqueous leaf extract of Angophora hispida exhibited statistically higher total phenolic content (107.85 ± 1.46 mg of gallic acid equiv. per g) and total flavonoid content (57.96 ± 1.93 mg rutin equiv. per g) and antioxidant capacity compared to the other tested eucalypts (< 0.05). Both Ahispida and Afloribunda aqueous extracts showed statistically similar saponin contents. Angophora floribunda extract exerted significantly greater cell growth inhibition of 77.91 ± 4.93% followed by Ahispida with 62.04 ± 7.47% (< 0.05) at 100 μg/ml in MIA PaCa‐2 cells with IC50 values of 75.58 and 87.28 μg/ml, respectively. More studies are required to isolate and identify the bioactive compounds from these two Angophora species and to determine their mode of action against pancreatic malignancies.  相似文献

以雷州半岛2年生桉树无性系DH201(Eucalyptus grandis  E. tereticornis)能源林为研究对象,对6种不同密度配置下的林分生长量、生物量以及热值等进行了分析,探讨密度对桉树生物质能源林生长的影响。结果表明,密度对桉树胸径和树高生长量影响极显著,在林分不同发育阶段对林分蓄积量的影响不同;单株生物量随密度增大而减小。1年生的林分蓄积量和生物量均以密度为10000 ind. hm-2的最大,分别为52.57 m3 hm-2和32.02 t hm-2;2年生的林分蓄积量和生物量均以密度为2500 ind. hm-2的最大,分别为75.85 m3 hm-2和45.66 t hm-2;密度对热值及灰分含量的影响不显著。因此,1年生和2年生的桉树能源林的最优密度分别为10000 ind. hm-2和2500 ind. hm-2。  相似文献

Photoinhibition of photosynthesis at low temperatures was investigated in two species of subalpine eucalypt, Eucalypts nitens (Deane and Maiden) Maiden and E. pauciflora Sieb. ex Spreng. Imposition of an artificial cold-hardening treatment increased the frost tolerance of leaf tissue and increased tolerance to excess light. Cold-hardened seedlings of both species had a higher photosynthetic capacity than non-hardened seedlings at 6 and 16°C and lower levels of non-photochemical quenching (NPQ) at 20 and 5°C. Furthermore, hardened seedlings had faster rates of NPQ development at 5 and −3.5°C. An increase in minimal fluorescence, which indicates slowly reversible photoinhibition, was evident in all seedlings at −1.5 and −3.5°C but was less pronounced in hardened seedlings, with a threefold faster rate of development of NPQ, at −3.5°C than non-hardened seedlings. Hardened seedlings also recovered faster from photoinhibition at −3.5°C. Thus cold hardening increased tolerance to high light in these species. Differences between E. nitens and E. pauciflora in their response to excess light were small and significant only at −3.5°C. Faster recovery from photoinhibition of E. pauciflora was consistent with its occurrence in colder habitats than E. nitens. Received: 27 April 1997 / Accepted: 9 September 1997  相似文献

Intra-specific hybrids within Eucalyptus nitens and E. globulus were compared directly with inter-specific E. nitens × globulus using common parents. Diameter (age 2, 4, 6, 10 years) and Pilodyn (age 6 years) were used as indirect measures of growth and wood density, respectively. Genetic parameters were estimated for all cross types and traits. A direct comparison of the general combining ability with the general hybridising ability was made. Heterosis was estimated for intra- and inter-specific hybrids. The intra-specific hybrids in both species exhibit intermediate heritabilities and levels of additive genetic variance. The inter-specific F1 E. nitens × globulus exhibited high incidence of abnormalities at young ages and high levels of later age mortality. The mean performance of surviving inter-specific F1 hybrids was generally intermediate for all traits, to varying degrees, between the parental intra-specific crosses, and there is little evidence for significant heterosis. Different provenances of E. globulus may exhibit different responses to hybridisation. In this case, the inter-specific F1 hybrids, using Taranna E. globulus, appear to perform worse on average than those using King Island provenance for growth traits. There is evidence that specific hybrid families are produced that outperform most of the pure species families for one or other of the traits examined; however, there is no reliable quantitative genetic method of predicting which parents should be used.  相似文献

 Regions of the genome influencing height and leaf area in seedlings of a three-generation outbred pedigree of Eucalyptus nitens have been identified. Three QTLs affecting height and two QTLs affecting leaf area were located using single-factor analysis of variance. The three QTLs affecting height each explained between 10.3 and 14.7% of the phenotypic variance, while the two QTLs for leaf area each explained between 9.8 and 11.6% of the phenotypic variation. Analysis of fully informative marker loci linked to the QTLs enabled the mode of action of the QTLs to be investigated. For three loci the QTL effect segregated from only one parent, while for two loci the QTL showed multiple alleles and the effect segregated from both parents in the pedigree. The two QTLs affecting leaf area were located in the same regions as two of the QTLs affecting height. Analysis of these regions with fully informative markers showed that both QTLs were linked to the same markers, but one had a similar size of effects and a similar mode of action for both height and leaf area, whilst the other showed a different mode of action for the two traits. These regions may contain two closely linked genes or may involve a single gene with a pleiotrophic effect on both height and leaf area. The QTL with the greatest effect showed multiple alleles and an intra-locus interaction that reduced the size of the effect. Assessment for two of the QTLs in a second related family did not show an effect associated with the marker loci; however, this was consistent with the mode of action of these QTLs and the pattern of inheritance in the second family. Adaptation to aridity is considered a major factor in the evolution of the genus Eucalyptus. For the first time, targeted metabolite profiling has uncovered a quantitative yet discrete phytochemical link with eucalypt taxonomy. The distribution of cyclitols among Eucalyptus species, and a range of other Australian tree genera, support their proposed functions in plant tissues and provide putative links with the acclimation of trees to arid environments.  相似文献

Native plants rarely occur in cogongrass (Imperata cylindrica) dominated phosphate-mine clay settling areas (CSA) in central Florida. This is primarily due to the allelopathic nature and strong competitiveness of cogongrass and frequent fires in these grasslands. This study examined the performance of fast-growing 2.5-year-old cottonwood (Populus deltoides) and 2–3.5-year-old eucalypts (Eucalyptus grandis and Eucalyptus amplifolia) in suppressing cogongrass in an old CSA near Lakeland, Florida. Understory vegetation was studied in two-row cottonwood and one-row, two-row, and four-row E. grandis cultures in the commercial planting and a clone-configuration-fertilizer study (SRWC-90). Cogongrass was still dominant in two-row cottonwood and the first four treatments of study SRWC-90. A total of 57 herbs and 26 shrubs, mostly native, were present in the understory. More herbs and shrubs occurred in the commercial planting than in the cogongrass-dominated study SRWC-90. Stand age and proximity to natural areas positively affected species recruitment. Cogongrass importance value index (IVI) decreased with increasing stand density, cottonwood basal area in the commercial planting and with both E. grandis and E. amplifolia basal area in study SRWC-90. Fast-growing trees with good survival on intensively prepared CSAs produce early and permanent canopy closure and appear to suppress cogongrass.  相似文献

依据永安市巨桉人工林调查的基础,对巨桉碳含量空间结构特征进行了研究,结果表明:巨桉不同胸径个体地上部分各器官的碳含量在42.30%~55.07%之间,地上部分各器官碳含量大小顺序为:叶>枝>干,方差分析表明不同胸径标准木之间碳含量没有达到显著水平,而地上部分各器官碳含量差异显著,采用邓肯新复全距法和q检验法对其进行多重比较,两种方法检验结果均表明巨桉枝和干的碳含量之间没有显著差异,叶和枝以及叶和干之间差异显著;不同高度层叶碳含量之间不存在明显差异,把巨桉单株林木的树冠分为上下两部位,方差分析表明上下两部分叶碳含量也不存在明显差异;不同大小根的碳含量大小顺序为:细根>中根>粗根>根兜;巨桉不同粗细枝的碳含量存在一定的差异,枝越细碳含量的值越高;各器官碳含量中,叶的碳含量较高,为51.86%,果的碳含量最低,为46.62%,排列顺序为:叶>根>枝>干>果。  相似文献
