目的:研究面神经撞击伤后面神经核内生长相关蛋白(GAP-43)的变化及高压氧(HBO)治疗的影响。方法:利用BIM-Ⅱ型生物撞击机致家兔右侧面神经干损伤,应用SP免疫组化染色法观察损伤后1、3、7、14和21天(分别用HBO治疗及不用HBO为对照)面神经核内GAP-43的变化,并用电镜观察面神经干超微结构变化。结果:面神经损伤第1天,伤侧面神经核区GAP-43样免疫反应性神经元(GLIN)开始增多,第14天其增多达到高峰,第21天逐渐下降。HBO治疗组伤后不同时期GLIN的增多明显高于对照组。伤后第21天,与对照组相比HBO组面神经干电镜观察可见较多的新生髓鞘。结论:HBO治疗可使伤后面神经核内GLIN增多,因而有助于面神经损伤后的修复与再生。  相似文献

The density of substance P (SP)-, calcitonin gene-related peptide (CGRP)- and vasoactive intestinal polypeptide (VIP)-immunoreactive (ir) nerve endings was quantitatively evaluated in intact and inflamed gastrocnemius-soleus muscle of the rat. In persistently inflamed muscle (12 days after a single injection of Freund’s adjuvant into the muscle), the density of SP-ir fibres was significantly increased. CGRP- and VIP-ir fibres displayed an insignificant increase in density. The density of fibres ir for nerve growth factor (NGF) and for growth-associated protein 43 (GAP-43/B-50), a marker for axonal sprouting, regeneration and synaptic reorganisation, increased significantly in persistently inflamed muscle. The data are consistent with the established contribution of NGF on the expression of SP and GAP-43 in afferent neurones under the influence of a persistent inflammation. 目的研究支气管哮喘(简称哮喘)过敏性刺激诱发气道感觉神经敏化机制。方法成年雄性豚鼠39只,按随机数字表法分为生理盐水致敏/激发对照组(A组,9只)、卵白蛋白(OVA)致敏/生理盐水激发对照组(B组,9只)、OVA致敏/激发实验组(C组,21只)。A组以生理盐水(0.5ml/只)致敏,B、C组以10%OVA(0.5ml/只)致敏,第10天开始雾化吸入生理盐水(A、B组)或1%OVA(C组)进行激发,每天1次,每次30min,根据实验需要又将C组21只豚鼠分为激发1d组(C1组,6只)、连续激发3d组(C2组,6只)、连续激发5d组(C3组,9只)。利用免疫荧光双标技术结合激光共聚焦扫描显微观察与Westernblot技术,研究生长相关蛋白43(GAP43)在气道神经以及结状神经节、颈静脉神经节内分布与水平及与P物质(SP)和胶质源神经生长因子(GDNF)受体c RET表达神经元关系。结果免疫荧光结果显示,C3组豚鼠气道内GAP43免疫反应阳性神经呈网状分布于大、中支气管内,以黏膜下层为主,部分GAP43阳性神经纤维向黏膜层内延伸;在结状神经节和颈静脉神经节内有大量GAP43免疫阳性神经胞体,在结状神经节内主要与SP免疫阳性胞体共存,在颈静脉神经节内主要与c RET免疫阳性胞体共存。Westernblot结果显示,A、B、C1、C2、C3组GAP43蛋白表达水平吸光度(A)值分别为0.38±0.04、0.41±0.03、0.49±0.05、0.79±0.08、0.76±0.04。C1、C2、C3组分别与A、B组比较差异均有统计学意义(P均0.05);C2组GAP43蛋白表达与C1组比较差异有统计学意义(P<0.01),但与C3组GAP43蛋白表达比较差异无统计学意义(P>0.05)。结论哮喘过敏性刺激能诱发气道感觉神经———SP肽能神经、GDNF敏感性神经纤维与胞体表达GAP43蛋白。  相似文献

Cardiac hamartomas are very rare and are demarcated masses of enlarged, hypertrophied, mature myocytes and collagen tissue. Cardiac hamartomas are generally circumscribed in the right ventricle or atrium, but not reported in the crista terminalis (CRT). The CRT is crucial in electrophysiology, is related to arrhythmogenesis, and is targeted by radiofrequency catheter procedures. Previous works only described the benign natures of prominent CRT using non-invasive methods. This study describes an unusual cardiac hamartoma originating from the CRT and extending toward the tricuspid valve. Microscopically, this hamartoma comprised dense collagen and adipose tissue, mixed with hypertrophy, but with disarrayed cardiomyocytes. An irregular gap junction, connexin43, was demonstrated in this cardiac hamartoma.  相似文献

The canine's olfactory acuity is legendary, but neither its main olfactory system nor its vomeronasal system has been described in much detail. We used immunohistochemistry on paraffin-embedded sections of male and female adult dog vomeronasal organ (VNO) to characterize the expression of proteins known to be expressed in the VNO of several other mammals. Basal cell bodies were more apparent in each section than in rodent VNO and expressed immunoreactivity to anticytokeratin and antiepidermal growth factor receptor antibodies. The thin layer of neurone cell bodies in the sensory epithelium and axon fascicles in the lamina propria expressed immunoreactivity to neurone cell adhesion molecule, neurone-specific beta tubulin and protein gene product 9.5. Some neurones expressed growth-associated protein 43 (GAP43): and a number of those also expressed neurone-specific beta tubulin-immunoreactivity. Some axon fascicles were double labelled for those two proteins. The G-protein alpha subunits Gi and Go, involved in the signal transduction pathway, showed immunoreactivity in the sensory cell layer. Our results demonstrate that the canine vomeronasal organ contains a population of cells that expresses several neuronal markers. Furthermore, GAP43 immunoreactivity suggests that the sensory epithelium is neurogenic in adult dogs.  相似文献

目的:采用条件性基因敲除技术构建造血系统间隙连接蛋白43(Cx43)基因敲除(Cx43~(-/-))小鼠模型,并探讨Cx43在维持造血细胞自我更新及功能稳定中的作用。方法:将引进的2对转基因小鼠Cx43 loxP/loxP和Lyz-Cre/+杂交,选取F1雌性子代Cx43 loxP/-_Lyz-Cre/+与雄性Cx43 loxP/loxP合笼回配,提取所获得子代小鼠鼠尾组织基因组DNA,采用PCR方法鉴定小鼠基因型,RT-PCR方法筛选Cx43~(-/-)小鼠,同时分析小鼠不同器官中Cx43基因的表达差异;该类小鼠经5-氟尿嘧啶(5-FU;125 mg/kg)处理,在化疗前及化疗后第5、10和15天经眼球取血分析其血象变化。Cx43~(-/-)及Cx43~(+/+)小鼠予7.5 Gy(~(60)Co-γ)的致死量照射,剂量率1 Gy/min,照射后6 h分别给予事先准备就序的骨髓细胞,每只3×10~6细胞于尾静脉注入,2周后处死小鼠检测造血是否重建:分离股骨切片后,收集骨髓细胞进行细胞表型分析(选用的单抗为CD45R、Gr-1、CD4、 CD8a、TCRαβ、Mac-1、抗sIgM、TER119、Sca-1及CD117);同时进行体外造血细胞集落实验观察造血细胞的体外增殖能力。结果:本研究通过2种转基因小鼠间杂交和回交,成功获得造血系统选择性Cx43基因敲除小鼠;该类小鼠骨髓及外周血细胞无Cx43表达,参与造血的组织,如肝脏和脾脏中Cx43表达也显著下调(P0.01),而心脏和肾脏的Cx43表达则无影响,小鼠成年后外周血象分析并无明显异常,但应急代偿能力下降,经5-FU处理后,其造血功能恢复显著减缓,处理15 d后,Cx43~(+/+)小鼠造血功能已接近正常水平,而Cx43~(-/-)小鼠仍无明显的恢复迹象,血红蛋白、白细胞及血小板仍处低位,2者差别有统计学显著性(P0.01);体外集落试验也证实Cx43~(-/-)小鼠造血干/祖细胞的增殖能力下降,其CFU-GM或CFU-E集落数均明显少于Cx43~(+/+)小鼠(P0.01),但流式细胞术结果显示,Cx43~(-/-)小鼠骨髓中Lin~-/c-Kit~+/Sca-1~+细胞亚群数量与Cx43~(+/+)小鼠相比差异并无统计学显著性;Cx43~(-/-)小鼠在化疗或移植后其骨髓造血功能重建均延迟,且化疗15 d后骨髓切片及涂片均证实其骨髓中造血细胞增生程度明显降低,脂肪组织显著增多,而且T、B细胞发育也有异常。此外,其外周血中CD4~+CD8~+细胞比例比野生型小鼠增多(P0.05),但CD4~+T细胞显著减少(P0.01),尤其是TCRαβ亚群细胞减少最为明显(P0.01)。同样,Cx43~(-/-)小鼠外周血中CD45R~+sIgM~-细胞亚群比例与野生型小鼠相比显著减少(P0.01)。结论:骨髓中Cx43基因表达在造血干/祖细胞发育(尤其是应急状态时)具重要作用,敲除Cx43基因后造血干/祖细胞增殖减缓,造血及免疫重建功能受损。  相似文献

We report on the expression of growth associated protein (GAP)43 and neural cell adhesion molecule (NCAM) in congenital fibre type disproportion (CFTD) with myopathological additional signs of interstitial myositis. We assume that sarcolemmal GAP43 in developmental disordered myocytes plays a role in maintenance of growth morphology. In muscular dystrophy light microscopical evaluation reveals no GAP43 immunoreactivity in regenerating fibres. The expression of GAP43 seems to be a characteristic feature of CFTD. The expression of NCAM, particularly in the sarcolemma of small muscle fibres of CFTD, indicates a functional state of permanent partial denervation. Whether the steroid-responsive interstitial myositis is pathogenetically related to CFTD or a coincidental inflammation is not known. Because of the clinical and myopathological data the differential diagnosis of Emery-Dreifuss muscular dystrophy is considered.  相似文献
