Synthesis and chiroptical properties of two new planar-chiral macrocycles

Could simple intraannular-arm macrocyclic systems exist in enantiopure stable forms? The effective synthesis of two representative compounds of such a class, their resolution into enantiomers, and experiments justifying their stability toward racemization are presented.     

Novel antibiotics: second generation macrocyclic peptides designed to trap Holliday junctions

Described are the syntheses of 15 macrocyclic peptides designed to trap Holliday junctions (HJs) in bacteria during site-specific and homologous recombination. This leads to inhibiting bacterial growth. These second generation macrocycles were based on the C-2 symmetrical HJ. They were synthesized using a strategy that permits elucidation of the amino acid role in binding HJs. The syntheses of these macrocycles are an important step in the development of a new class of antibiotics.     

Novel expanded porphyrinoids with multiple-inner-ring-fusion and/or tunable aromaticity

Novel expanded porphyrinoids with advanced structure features have a wide range of benefits (such as multi-metal coordination and facile tunable aromaticity) not offered by their normal porphyrin analogues. Considering research efforts have been devoted to address their limited synthetic accessibility issue. This review highlights some of these recent synthetic progresses towards these novel expanded porphyrinoids.     

Tandem reactions of α-diazo ketones with macrocyclic olefins: diastereoselective synthesis of oxanorbornane fused macrocyclic lactones

Tandem cyclization-cycloaddition reactions of α-diazo ketones with macrocyclic olefins in the presence of rhodium(II) acetate catalyst led to the oxanorbornane fused macrocyclic di- or tetralactone ring systems in moderate yield. This forms the first example of 1,3-dipolar cycloaddition reactions with a macrocyclic olefin as a dipolarophile, affording a variety of new oxanorbornane fused macrocycles with diastereoselectivity.     

Efficient syntheses of new chiral peptidomimetic macrocycles through a configurationally driven preorganization

A new family of 32-membered ring peptidomimetic macrocycles has been efficiently obtained in a simple one-pot two-step reductive amination reaction, from easily prepared precursors. The structural and stereochemical variables have been explored in order to rationalize the obtained selectivity. The formation of the [2A+2B] tetraimine intermediate has been explained in terms of a very favorable configurationally driven preorganization as detected by NMR, CD and molecular modeling.     

Syntheses of macrocyclic bisbibenzyls on solid support

We describe a route for the polymer supported total synthesis of the cyclic bisbibenzyls of the isoplagiochin type found in liverworts. TentaGel^® resins were used as solid support for a sequence involving Suzuki, Wittig and hydrogenation protocols. The polymer linked intermediates could be characterized by HR-MAS NMR. This route is to be extended to the synthesis of small libraries of differently halogenated derivatives.

Molecular modeling of 14 bicyclophosphoranic macrocycles I, II and III has been carried out using Biosym V95 program based on the ESFF forcefield and including the modules Insight II and Discover 3.     

Biological applications of amide and amino acid containing synthetic macrocycles

ABSTRACT

Amino acid derived macrocycles with elaborate well-defined stereochemistry are a unique class of compounds that have been isolated from natural sources. Macrocycles like cyclosporine, octreotide, and valinomycin have been used in multiple applications, like drugs or ion sensors. Chemists have long been fascinated by the unique molecular recognition capabilities of these macrocycles and tried to design synthetic analogs with similar functions. This article is focused on reviewing current research on amide and amino acid containing macrocycles that have been developed in research laboratories for biological recognition, specifically for anion sensing, ion transport, carbohydrate sensing, and peptide sensing.     

Design,Synthesis and Structural Analysis of New Macrocycles Containing Dispiro-1,3-dioxane Units

The synthesis and the structure of new macrocycles containing semiflexible dispiro-1,3-dioxane units is reported. The structural analysis of the compounds is performed by high field NMR spectra, mass spectrometry investigations (MALDI, ESI-MS) and the solid state molecular structure obtained for two compounds by single crystal X-ray diffractometry. The dynamics of the macrocycles promoted by the flipping of the middle cyclohexane ring of the dispirane units is investigated using low temperature NMR experiments.

Supramolecular Chemistry In Silico

Computational protocols capable of modelling supramolecular complexes have been evaluated. The complexation of cations by crown ethers and quaternary ammonium ions by an oxacalix[3]arene are presented as examples. In the latter case reliable qualitative results were obtained using the semi-empirical PM3 method where guest LUMO and electrostatic potential energies have been shown to correlate with experimental binding data. The optimal method for more accurate results combines semi-empirical equilibrium geometry and property calculations with single point energy calculations at the HF/6–31G* or BP/6–31G* quantum mechanical level.
