In this work the interaction of a non-steroidal anti-inflammatory drug (NSAID), diclofenac, with egg yolk phosphatidylcoline (EPC) liposomes, used as cell-membrane models, was quantified by determination of the partition coefficient. The liposome/aqueous phase partition coefficient was determined by derivative spectrophotometry, fluorescence quenching, and measurement of zeta-potential. Theoretical models based on simple partition of the diclofenac between two different media, were used to fit the experimental data, enabling the determination of Kp. The three techniques used yielded similar results. The effects of the interaction on the membranes characteristics were further evaluated, either by studying membrane potential changes or by effects on membrane fluidity. The liposome membrane potential and the size and size-homogeneity of liposomes were measured by light scattering. The effects of diclofenac on the internal viscosity or fluidity of the membrane were determined by use of spectroscopic probes—a series of n-(9-anthroyloxy) fatty acids in which the carboxyl terminal group is located at the interfacial region of the membrane and the fluorescent anthracene group is attached at different positions along the fatty acid chain. The location of the diclofenac on the membrane was also evaluated, by fluorescence quenching using the same series of fluorescent probes. Because the fluorescent anthracene group is attached at different positions along the fatty acid chain, it is possible to label at a graded series of depths in the bilayer. The interactions between the drug and the probe are a means of predicting the location of the drug on the membrane.  相似文献

The characteristics,performance and application of membrane electrode based on ion associate of diclofenac with base dye Safranine T are described.The electrode response to diclofenac has the sensitivity of 47±1.0 mV decade~(-1)over the range of 5×10~(-5)to 5×10~(-2)mol/L at pH 6-12,and the detection limit of 3.2×10~(-5)mol/L.The electrode is easy assembled at a relatively low cost has fast response time(2-4 s)and can be used for a period up to 3.5 months without any considerable divergence in potential.The proposed sensor displayed good selectivity for diclofenac in the presence of different substances.It was used to determine diclofenac in pharmaceuticals by means of the standard addition method.  相似文献

A new, repeatable, and rapid method has been developed for resolution of binary mixtures of acetaminophen and diclofenac with minimum sample pretreatment and without separation of the analytes. The method, based on the PLS1 processing of absorbance data in the UV region, was successfully used for quantification of the drug content of three tablet preparations. The results obtained were in good agreement with HPLC recovery data. The method also enabled determination of drug-dissolution profiles of these commercial tablets, by simultaneous determination of both analytes during the dissolution test.  相似文献

The subject of this work is the definition of a simple model based on general ITP theory that allows describing and predicting the behavior of ITP systems compatible with ESI‐MS detection. The model is exemplified by anionic ITP of weak acids that represent an interesting potential application field of ITP‐ESI‐MS. Suitable ESI‐compatible electrolyte systems of very simple composition are proposed including a special free‐acid ITP arrangement. The properties of these systems are discussed using illustrative diagrams of their stacking windows. The use of anionic ITP‐ESI‐MS in negative‐ion ESI mode is reported for the first time and its suitability for sensitive trace analysis is demonstrated. The presented ITP‐ESI‐MS application example comprises a free‐acid ITP system formed of formic and propionic acids and direct injection analysis of ibuprofen and diclofenac in waters with quantitation limits of the order 10?10 M.  相似文献

This work describes the electrochemical behavior of diclofenac on the surface of a carbonceramic electrode (CCE) modified with multi-walled carbon nanotubes (MWCNT) and an ionic liquid (IL) composite. The MWCNT-IL composite showed an enhancement effect in the electro-oxidation of diclofenac with respect to a bare carbon ceramic electrode. Based on the experimental outcomes, a possible mechanism for the electro-oxidation of diclofenac is proposed and discussed. Under the optimized experimental conditions, the MWCNT-IL CCE showed a linear response to diclofenac over the concentration range 50 nM–20 µM with a detection limit of 27 nM. The developed diclofenac sensor showed good stability, sensitivity, and reproducibility in the measurement of diclofenac in human blood plasma samples.  相似文献

In this study, a non-cytotoxic and pH-sensitive poly(acrylamidoglycolic acid) based nanocomposite (PAGA-NC) hydrogels reinforced with cellulose nanocrystals (CNCs) was synthesized using redox free radical polymerization. The successful formation and crystalline behaviour of PAGA-NC hydrogels was verified by fourier transform infrared (FTIR) spectroscopy and X-ray diffraction (XRD) analyses. The results showed that morphological, rheological and mechanical properties of the PAGA-NC hydrogels were strongly influenced by the CNCs content. Moreover, swelling properties were investigated, and the results suggested that they behaved as pH sensitive manner. The in vitro MTT assay showed that the PAGA-NC hydrogels are cytocompatibile to NIH-3T3 fibroblast cells. In addition, diclofenac sodium (DCF) model drug was successfully encapsulated into these PAGA-NC hydrogels via equilibrium swelling method. The in vitro release of DCF from PAGA-NC hydrogels was retained at pH 1.2 and maximum release was observed at 7.4, revealing as potential candidates for controlled release carriers for oral drug delivery applications.  相似文献

研究了磁化多壁碳纳米管(MWCNTs)对于水中非甾体抗炎药双氯芬酸的吸附过程.结果表明,双氯芬酸的吸附量随磁性MWCNTs投加量的增加而增大,而且吸附剂量增加到一定阶段后,双氯芬酸的吸附量达到平衡.在磁性MWCNTs的量为0.7g·L-1时,水溶液中双氯芬酸被磁性MWCNTs吸附的量达到最大,为33.37mg·g-1,对应的双氯芬酸去除率为98.1%.双氯芬酸的去除率随溶液pH的增加先增大后下降,随温度的升高而下降.用准一级、准二级模型进行了动力学分析.回归结果表明,准二级模型更准确地反映了吸附动力学.通过实验确定了Langmuir和Freundlich等温线的线性相关系数与标准偏差,结果揭示出Langmuir等温线与实验数据有很好的拟合度.对热力学参数的计算表明,ΔG00,意味着磁性MWCNTs对双氯芬酸的吸附是自发的;ΔH00,指明吸附是一个放热的物理吸附过程,温度低对吸附有利;ΔS00,代表该吸附是熵增过程.  相似文献

We report a new in-tube solid phase microextraction approach named electrochemically controlled in-tube solid phase microextraction (EC in-tube SPME). This approach, which combined electrochemistry and in-tube SPME, led to decrease in total analysis time and increase in sensitivity. At first, pyrrole was elctropolymerized on the inner surface of a stainless steel tube. Then, the polypyrrole (PPy)-coated in-tube SPME was coupled on-line to liquid chromatography (HPLC) to achieve automated in-tube SPME–HPLC analysis. After the completion of EC-in-tube SPME–HPLC setup, the PPy-coated tube was used as working electrode for uptake of diclofenac as target analyte. Extraction ability of the tube in presence and in absence of applied electrical field was investigated. It was found that, under the same extraction conditions, the extraction efficiency could be greatly enhanced by using the constant potential. Important factors are also optimized. The detection limit (S/N = 3) and precision were 0.1 μg L−1 and 4.4%, respectively.  相似文献

双氯芬酸钠是一种非甾体抗炎药,抗炎症效果较好,抗炎化学药物双氯芬酸钠有可能违禁添加到缓解炎症保健食品中。目前已有一些文献研究水、肉及牛奶等中的药物双氯芬酸钠,主要利用高效液相色谱、表面增强拉曼及电化学等方法,需要相对复杂的预处理过程,操作步骤相对复杂,耗时。为了快速、无损检测保健食品中违禁添加物的成分,探索建立采用1 000~2 524 nm波段或特征最优波段的光谱作为自变量,将近红外高光谱成像技术结合化学计量学分析的方法定量分析缓解炎症保健食品中可能添加的不同浓度的双氯芬酸钠。研究主要运用了8种光谱预处理方法,基于全光谱波长建立了偏最小二乘回归及主成分回归模型,同时,为了提高模型的准确性和稳定性,利用β系数方法选择最优波段1 130~1 147,1 412~1 468,1 658~1 709,2 010~2 055,2 122~2 178和2 395~2 423 nm为自变量建立与双氯芬酸钠浓度的多元线性回归模型。比较了偏最小二乘回归、主成分回归及多元线性回归三种模型。分析结果显示,经标准正态变量预处理方法建立多元线性回归模型,该模型的准确性和和预测能力较好,其预测最低限为0.05%,预测值与实测值的R2为0.992 5,预测均方根误差为0.004 9,标准预测偏差为0.004 9,说明近红外高光谱成像技术为快速定量分析缓解炎症保健食品中违禁添加抗炎药物双氯芬酸钠提供了理论基础。有望进一步开发拓展到其他违禁添加药物的快速定量应用。  相似文献
