The colony-forming efficiency (CFE) of primary human tumor cells cultured in the adhesive-tumor-cell culture system (ATCCS) using Ham's F12 (F12) or Eagle's minimum essential medium, alpha modification (alphaMEM) and culture medium supplemented with either swine, equine or bovine sera were compared. AlphaMEM supplemented with equine serum provided the highest CFE of the combinations. The CFE increase due to the change from F12 to alphaMEM was approximately 5-fold, and the increase due to the change in serum from swine to equine was approximately 2-fold. Cytokeratin staining showed that this increase was not due to fibroblast growth. The high-average CFE with alphaMEM, approximately 3%, means that an inoculum of only 2 X 10(3) cells is needed to achieve formation of approximately 65 colonies in control cultures, thereby increasing the performance of this system when used in a chemosensitivity assay.  相似文献

Arachidonic acid metabolites are mediators of various pathophysiologic events following endotoxin administration. However, their role in the endotoxin-induced increase in glucose metabolism has not been examined. Rats were administered either saline or BW755C (an inhibitor of both the cyclooxygenase and lipoxygenase pathways) 30 min prior to injection of E. coli endotoxin and whole body glucose kinetics assessed using a constant iv infusion of [6-3H] glucose. Treatment with BW755C prevented the endotoxin-induced hypotension and tachycardia. Endotoxin produced characteristic increases in the plasma glucose (23-70%) and lactate (2- to 9-fold) concentrations, as well as elevations in the rate of glucose appearance (34-63%) and metabolic clearance (40-92%). In contrast to the amelioration in hemodynamics, pretreatment with BW755C did not prevent these alterations in glucose metabolism normally seen after endotoxin. BW755C markedly reduced the endotoxin-induced increase in plasma catecholamine concentrations, but levels were still elevated 2- to 4-fold compared to control values. The results suggest that arachidonic acid metabolites mediate the early hypotensive response following endotoxin, but are not by themselves responsible for the elevated rates of glucose production and utilization.  相似文献

The role of ketone bodies in myocardial substrate oxidation was examined using freshly isolated Ca2+-tolerant heart myocytes, beta-hydroxybutyrate (beta OHB) inhibited lactate oxidation by the myocytes by 30-60%, and the inhibition was concentration dependent. Palmitate oxidation was also markedly decreased, whereas octanoate oxidation was only minimally affected by the presence of beta OHB. Lactate, octanoate, or palmitate had little, if any, effect on beta OHB oxidation. beta OHB oxidation was reduced by 22-28% in myocytes isolated from chronically diabetic rats, whereas the oxidation of palmitate remained similar to the controls. However, beta OHB still inhibited palmitate oxidation to the same extent as in the control cells. Our data support the role of beta OHB as a physiologic regulator of myocardial substrate metabolism.  相似文献

Using whole cottonseed as a lipid source, silage-based diets that were isocaloric and isonitrogenous yet varied in lipid level were fed to multiparous cows. In Experiment 1, 48 cows (n = 12 per treatment) were allotted to 1 of 4 treatments where diets were formulated to supply 3.9, 4.3, 5.3 and 6.3% of total lipid. In Experiment 2, 66 cows (n = 22 per treatment) were allotted to 1 of 3 treatments where diets were formulated to supply 3.1, 5.5 and 8.3% of total lipid. Length of the first ovarian cycle, length of the first normal estrous cycle, postpartum intervals to onset of ovarian luteal activity and to first estrus were not affected by diet (P>0.10) in either experiment. Mean progesterone (P(4)) concentrations for first normal estrous cycles were not different (P>0.10) in either experiment. Anestrous periods were divided into 3 phases for analyses: Phase I) parturition to onset of ovarian luteal activity, Phase II) first ovarian luteal activity and Phase III) first normal estrous cycle. No differences were observed in P(4) concentrations during any phase of the postpartum period. In conclusion, isocaloric and isonitrogenous diets with increasing levels of lipid had no effect on reproductive performance in suckled beef in these experiments.  相似文献

David Marr and others have hypothesized that the visual system processes complex scene information in stages, the first of which involves the detection of light intensity edges or zero-crossings (Marr, 1982). Ideal zero-crossing detector mechanisms have been described and modeled in terms of their possible physiological implementation (Marr and Hildreth, 1980; Poggio, 1983). We now present evidence of visual cortical receptive fields which resemble in spatial organizational terms the requirements of zero-crossing analysis. The linear and nonlinear summation within and between the receptive field subunits are described and compared with predicted processes. The relative subunit sizes and separations are analyzed in these terms. Our findings support the notion that receptive fields may correspond with zero-crossing filters rather than zero-crossing detector gates.  相似文献

Germfree fat cells released significantly less FFA and glycerol under basal conditions (i.e. in the absence of hormonal stimulation) than conventional cells. The lipolytic response to norepinephrine stimulation (0.2 μg/ml) was not different in the two cell populations.E. coli endotoxin increased basal lipolysis and norepinephrine stimulated (0.2 μg/ml) FFA release in adipocytes from conventional dogs, while having no consistent influence on lipolysis of adipocytes from germfree dogs. The endotoxin effect was not dose dependent (0.2–2.0 μg/0.5 ml cell suspension).Indomethacin (5.0 μg/ml) significantly increased basal FFA and glycerol release from cells of germfree origin, and FFA efflux from cells of conventional dogs. Endotoxin obviated the influence of indomethacin on basal lipolysis of germfree cells.Endotoxin by itself did not alter cAMP concentrations in adipocytes from germfree dogs. The combination of indomethacin and endotoxin, however, significantly increased intracellular cyclic nucleotide concentrations.Compared to conventional fat cells, germfree fat cells are characterized by significantly reduced basal lipolysis, lack of a consistent lipolytic response to endotoxin stimulation and dissociation of the lipolytic response and cAMP levels by the combined influence of endotoxin and indomethacin.  相似文献

The delayed rectifier current of embryonic Xenopus spinal neurons plays the central role in developmental conversion of calcium-dependent action potentials to sodium-dependent spikes. During its maturation, this potassium current undergoes a pronounced increase in rate of activation. The mechanism underlying the change in kinetics was analyzed with whole-cell voltage clamp of neurons cultured under various conditions. Calcium is necessary at an early stage of development, to permit influx that triggers subsequent release of calcium from intracellular stores. Its action is prevented by depletion of protein kinase C and mimicked by stimulation of the kinase. Calcium influx through voltage-dependent channels at early stages of development regulates the differentiation of potassium current kinetics and modulation of the ionic dependence of action potentials.  相似文献
