Summary Production of extracellular hydrogen peroxide by fungal oxidases is been investigated as a requirement for lignin degradation. Aryl-alcohol oxidase activity is described in extracellular liquid and mycelium ofPleurotus eryngii and studied under non-limiting nitrogen conditions. This aryl-alcohol oxidase catalyses conversion of primary aromatic alcohols to the corresponding aldehydes and H2O2, showing no activity with aliphatic and secondary aromatic alcohols. The enzyme is stable at pH 4.0–9.0, has maximal activity at 45°–50°C and pH 6.0–6.5, is inhibited by Ag+, Pb2+ and NaN3, and has aK m of 1.2 mM using veratryl alcohol as substrate. A single protein band with aryl-alcohol oxidase activity was found in zymograms of extracellular and intracellular crude enzyme preparations fromP. eryngii.  相似文献

The antibody titer was followed in a group of patients, clinically diagnosed with toxocariasis, during a 5 year period. We observed that larvae can survive for at least 5 years in humans. Antigenic stimulation was enough to keep high levels of immunoglobulins over this period. Antibody levels decreased slowly and this pattern is similar to that shown by animal models.  相似文献

The production in a 5-1 fermenter of the extracellular enzymes laccase and aryl-alcohol oxidase by the fungus Pleurotus eryngii was studied. The latter enzyme has been purified 50-fold by Sephacryl S-200 and Mono Q chromatography. Purified aryl-alcohol oxidase is a unique flavoprotein with 15% carbohydrate content, a molecular mass of 72.6 kDa (SDS/PAGE) and a pI of 3.9. The enzyme presents wide specificity, showing activity on benzyl, cinnamyl, naphthyl and aliphatic unsaturated alcohols. Neither activity nor inhibition of veratryl alcohol oxidation was found with saturated alcohols, but competitive inhibition was produced by aromatic compounds which were not aryl-alcohol oxidase substrates, such as phenol or 3-phenyl-1-propanol. From these results, it was apparent that a double bond conjugated with a primary alcohol is necessary for substrate recognition by aryl-alcohol oxidase, and that activity is increased by the presence of additional conjugated double bonds and electron donor groups. Both affinity and maximal velocity during enzymic oxidation of methoxybenzyl alcohols were affected in a similar way by ring substituents, increasing from benzyl alcohol (Km = 0.84 mM, Vmax = 52 U/mg) to 4-methoxybenzyl alcohol (Km = 0.04 mM, Vmax = 208 U/mg). Aryl-alcohol oxidase presents also a low oxidase activity with aromatic aldehydes, but the highest activity was found in the presence of electron-withdrawing groups.  相似文献

ObjectiveRisk factors for differentiated thyroid carcinoma (DTC) are poorly understood, but serum TSH levels, thyroid nodularity, and presence of autoimmunity are well-recognized factors that modulate DTC prevalence. TSH stimulates proliferation of both normal and neoplastic follicular cells. Consequently, thyroid-stimulating immunoglobulins (TSI), because of its TSH-like action, should induce DTC progression in patients with Graves’ disease (GD). The study objective was to compare the prevalence of incidental DTC in patients undergoing thyroidectomy for benign thyroid disease.MethodsThe pathology reports of 372 patients with preoperative diagnosis of euthyroid multinodular goiter (EMG) or hyperthyroidism were reviewed. Scintigraphy results and serum TSI levels were used to diagnosed either GD or hyperactive MG (HMG) to hyperthyroid subjects. Prevalence of DTC in each category was calculated using a Chi-square test.ResultsEMG, GD, and HMG were diagnosed in 221, 125, and 26 patients. There were 58 DTCs, distributed as follows [n (%)]: EMG, 49 (22.2%); GD, 8 (6.4%), and HMG, 1 (3.8%). Difference in prevalence of incidental DTC between the groups was statistically significant (p < 0.001). After adjustment for age, patients with EMG had a greater DTC prevalence than GD patients, with an OR of 4.17 (p < 0.001). Tumor size (mm, mean ± SD) was 6.92 ± 11.26, 1.97 ± 1.85, and 9.0 for EMG, GD and HMG respectively (p = 0.017).ConclusionsIncidental DTC was less prevalent in GD as compared to EMG irrespective of age. This finding may suggest a predisposition to develop DTC in patients with thyroid nodular disease and/or a potential effect of autoimmunity to protect against development of neoplastic disease.  相似文献

The stability of ecological communities depends strongly on quantitative characteristics of population interactions (type‐II vs. type‐III functional responses) and the distribution of body masses across species. Until now, these two aspects have almost exclusively been treated separately leaving a substantial gap in our general understanding of food webs. We analysed a large data set of arthropod feeding rates and found that all functional‐response parameters depend on the body masses of predator and prey. Thus, we propose generalised functional responses which predict gradual shifts from type‐II predation of small predators on equally sized prey to type‐III functional‐responses of large predators on small prey. Models including these generalised functional responses predict population dynamics and persistence only depending on predator and prey body masses, and we show that these predictions are strongly supported by empirical data on forest soil food webs. These results help unravelling systematic relationships between quantitative population interactions and large‐scale community patterns.  相似文献

Purpose

The main goal of any life cycle assessment (LCA) study is to identify solutions leading to environmental savings. In conventional LCA studies, practitioners select from some alternatives the one which better matches their preferences. This task is sometimes simplified by ranking these alternatives using an aggregated indicator defined by attaching weights to impacts. We address here the inverse problem. That is, given an alternative, we aim to determine the weights for which that solution becomes optimal.

Methods

We propose a method based on linear programming (LP) that determines, for a given alternative, the ranges within which the weights attached to a set of impact metrics must lie so that when a weighting combination of these impacts is optimized, the alternative can be optimal, while if the weights fall outside this range, it is guaranteed that the solution will be suboptimal. A large weight value implies that the corresponding LCA impact is given more importance, while a low value implies the converse. Furthermore, we provide a rigorous mathematical analysis on the implications of using weighting schemes in LCA, showing that this practice guides decision-making towards the adoption of some specific alternatives (those lying on the convex envelope of the resulting trade-off curve).

Results and discussion

A case study based on the design of hydrogen infrastructures is taken as a test bed to illustrate the capabilities of the approach presented. Given are a set of production and storage technologies available to produce and deliver hydrogen, a final demand, and cost and environmental data. A set of designs, each achieving a unique combination of cost and LCA impact, is considered. For each of them, we calculate the minimum and maximum weight to be given to every LCA impact so that the alternative can be optimal among all the candidate designs. Numerical results show that solutions with lower impact are selected when decision makers are willing to pay larger monetary penalties for the environmental damage caused.

Conclusions

LP can be used in LCA to translate the decision makers’ preferences into weights. This information is rather valuable, particularly when these weights represent economic penalties, as it allows screening and ranking alternatives on the basis of a common economic basis. Our framework is aimed at facilitating decision making in LCA studies and defines a general framework for comparing alternatives that show different performance in a wide variety of impact metrics.  相似文献

The fact that pests are the most abundant species in agricultural settings has broadly precluded the attention to non-pest species and the study of temporal dynamics of diversity in agroecosystems. Because, agroecosystems hold increasingly important portions of biological diversity, understanding of non-pest species dynamics in such systems will contribute significantly to their conservation. In addition, deep understanding of both pest and non-pest population dynamics in a community context necessarily requires a long-term approach. By means of the analysis of weekly fruit fly sampling sessions across 12 years, in three tropical fruit orchards, we describe the temporal dynamics of species richness and turnover, structure and composition of Anastrepha fruit fly ensembles considering pest and non-pest species. Furthermore, we ask if time series of non-pest species covariate with time series of pest species, as a way to evaluate the best management scheme to minimize negative impacts of pest control on non-pest species. Among 18 Anastrepha fruit fly species detected over 12 years, five were considered as pest species. Fruit fly ensembles were characterized by strong seasonal dynamics composed of annual cycles. Sapodilla was the most diverse orchard. Overall, fruit fly ensembles appeared stable throughout time. The temporal dynamics of non-pest species covaried positively with temporal dynamics of pest abundance, with consequent management implications. Results suggest that in mango and grapefruit orchards, pest control could be focused during time periods with low potential impact on non-pest species; while in sapodilla orchards other approaches should be developed. The approach described here could be used in agroecosystems to minimize the impact of pest management on non-pest species particularly in highly anthropized landscapes and human-managed ecosystems were biodiversity conservation is a high priority.  相似文献

The phylogenetic relationships of genus Passer (Old World sparrows) have been studied with species covering their complete world living range. Mitochondrial (mt) cyt b genes and pseudogenes have been analyzed, the latter being strikingly abundant in genus Passer compared with other studied songbirds. The significance of these Passer pseudogenes is presently unclear. The mechanisms by which mt cyt b genes become pseudogenes after nuclear translocation are discussed together with their mode of evolution, i.e., transition/transversion mitochondrial ratio is decreased in the nucleus, as is the constraint for variability at the three codon positions. However, the skewed base composition according to codon position (in 1st position the percentage is very similar for the four bases, in 2nd position there are fewer percentage of A and G and more percentage of T, and in 3rd codon position fewer percentage of G and T and is very rich in A and C) is maintained in the translocated nuclear pseudogenes. Different nuclear internal mechanisms and/or selective pressures must exist for explaining this nuclear/mitochondrial differential DNA base evolutive variability. Also, the phylogenetic usefulness of pseudogenes for defining relationships between closely related lineages is stressed. The analyses suggest that the primitive genus Passer species comes from Africa, the Cape sparrow being the oldest: P. hispaniolensis italiae is more likely conspecific to P. domesticus than to P. hispaniolensis. Also, Passer species are not included within weavers or Estrildinae or Emberizinae, as previously suggested. European and American Emberizinae sparrows are closely related to each other and seem to be the earliest species that radiated among the studied songbirds (all in the Miocene Epoch). In this work, a murine experimental model of toxocariasis has been developed in BALB/c, C57BL/10 and C3H murine strains orally inoculated with 4,000 Toxocara canis embryonated eggs, in order to investigate the isotype-specific immune responses against excretory-secretory antigens from larvae. T. canis specific IgG+M, IgM, IgG, IgA, IgG1, IgG2a and IgG3 were tested by ELISA. The dynamics of the specific immunoglobulins (IgG+IgM) production showed a contrasting profile regarding the murine strain. Conversely to the results obtained with the IgM isotype, the IgG antibody class showed similar patterns to those obtained with IgG+IgM antibodies, only in the case of the BALB/c strain, being different and much higher than the obtained with IgG+IgM antibodies, when the C3H murine strain was used. The antibodies IgG+IgM tested in BALB/c and C57BL/10 were both of the IgM and IgG isotypes. Conversely, in the C3H strain only IgG specific antibody levels were detected. The IgG1 subclass responses showed a similar profile in the three murine strains studied, with high values in BALB/c, as in the case of the IgG responses.  相似文献
