Two hundred twenty-eight patients with successful coronary stent implantation were randomized either to protamine administration and femoral sheath removal (group I, n = 117) or no heparin neutralization and delayed sheath removal (group II, n = 111). The hospital stay after treatment was shorter in patients receiving protamine; therefore, protamine use for neutralizing circulating heparin may be safely administered immediately after stent implantation.  相似文献

We have identified and cloned a new member of the papain family of cysteine proteinases from a human brain cDNA library. The isolated cDNA codes for a polypeptide of 334 amino acids that exhibits all of the structural features characteristic of cysteine proteinases, including the active site cysteine residue essential for peptide hydrolysis. Pairwise comparisons of this amino acid sequence with the remaining human cysteine proteinases identified to date showed a high percentage of identity (78%) with cathepsin L; the percentage of identity with all other members of the family was much lower (<40%). On the basis of these structural characteristics, we have tentatively called this novel protein cathepsin L2. The cDNA encoding the mature cathepsin L2 was expressed in Escherichia coli, and after purification, the recombinant protein was able to degrade the synthetic peptide benzyloxycarbonyl-L-phenylalanyl-L-arginine-7-amido-4-methylcoumarin, which is commonly used as a substrate for cysteine proteinases. Cathepsin L2 proteolytic activity on this substrate was abolished by trans-epoxysuccinyl-L-leucylamido-(4-guanidino)butane, an inhibitor of cysteine proteinases, thus providing additional evidence that the isolated cDNA encodes a functional cysteine proteinase. Northern blot analysis of polyadenylated RNAs isolated from a variety of human tissues demonstrated that cathepsin L2 is predominantly expressed in the thymus and testis. This finding is in marked contrast with the wide tissue distribution of most cysteine proteinases characterized to date, including cathepsin L, and suggests that cathepsin L2 may play a specialized role in the thymus and testis. Expression analysis of cathepsin L2 in human tumors revealed a widespread expression in colorectal and breast carcinomas but not in normal colon or mammary gland or in peritumoral tissues. Cathepsin L2 was also expressed by colorectal and breast cancer cell lines as well as by some tumors of diverse origin, including ovarian and renal carcinomas. These results open the possibility that this novel enzyme may be involved in tumor processes, as already reported for other cysteine proteinases, including cathepsin L.  相似文献

Poly(1‐trimethylsilyl‐1‐propyne) (PTMSP) membranes have been used to separate ethanol–water mixtures by pervaporation. This polyacetylene is known to present high affinity toward ethanol, showing high selectivity and ethanol permeation flux. The performance of this polymer in the separation of alcohol–water solutions has been evaluated over long periods (572 h) at a high temperature (75°C) to examine the deterioration of the transport properties in the separation of 50 wt % ethanol–water solutions. Although PTMSP membranes present good characteristics for the separation of gases and liquid mixtures, their organic selectivity decrease with the operating time because of the relaxation processes of the polymeric chains, which affect the free volume of the polymer, the deterioration being more evident for concentrated solutions. The effects of the operation temperature on the characteristic parameters of pervaporation have also been studied to establish how this variable affects the performance of PTMSP membranes. The selectivity increases slightly with the operation temperature, but the effect of the temperature on the separation factor decreases as membranes are degraded with the operation time. © 2006 Wiley Periodicals, Inc. Seeds from 22 accessions of the yam bean species Pachyrhizus ahipa (14 accessions), P. erosus (5), and P. tuberosus (3) were investigated for oil and protein contents, fatty acid composition of the seed oil, and the total tocopherol content and composition. Plants from the accessions were grown under greenhouse conditions during one (P. erosus and P. tuberosus) or two years (P. ahipa). The pattern of the investigated seed quality traits was very similar in the three species. Yam bean seeds were characterized by high oil (from about 20 to 28% in one environment) and protein contents (from about 23 to 34%). Seed oil contained high concentrations of palmitic (from about 25 to 30% of the total fatty acids), oleic (21 to 29%), and linoleic acids (35 to 40%). Levels of linolenic acid were very low, from about 1.0 to 2.5%. Total tocopherol content was relatively low in P. erosus (from 249 to 585 mg kg−1 oil) and P. tuberosus (from 260 to 312 mg kg−1 oil) compared with the levels found in P. ahipa grown under identical conditions (508 to 858 mg kg−1 oil). In all the samples, γ-tocopherol was predominant, accounting for more than 90% of the total tocopherol content. The combination of high oil and protein contents, together with high palmitic acid, low linolenic acid, and high γ-tocopherol concentration, makes these crops an interesting alternative as sources of high palmitic acid oil for the food industry.  相似文献

The advances in the field of nutritional support have made certain nutrients very relevant, which, although they have been known for a long time, at present represent an important chapter in nutrition, entering into what is known as "nutritional pharmacology". Among these nutrients is glutamine, an amino acid classified as non-essential, but which in certain circumstances may become to be considered as an "essential nutrient". In the present review, a review is made of its metabolic role, synthesis and degradation, metabolic routes and functions under normal conditions as well as under critical conditions. It is known that glutamine stimulates the synthesis and inhibits the degradation of proteins, it is an important vehicle for the transport of nitrogen and carbon within the tissues, it stimulates the synthesis of hepatic glycogen, it is an energy source for cell division, for the growth of different cells of rapid replication, such as enterocytes, colonocytes, and fibroblasts, as well as for other cells of the immune system, such as lymphocytes and macrophages. Thus its role in the maintenance of structure, in metabolism and function of the intestinal mucosa, and in dysfunctions of the immune system. In parenteral nutrition, at present there are no preparations which include it, given the stability problems which it presents, although attempts have been made to resolve this, using different possibilities, such as di-peptides. However, in enteral nutrition, the diets tend to include it, although in a small proportion. Nevertheless, having recognized its beneficial role in a certain type of patients, at present there are diets which contain glutamine in higher doses, with the object of attempting to cover the increased demands of glutamine which shall arise in these situations. The inclusion of glutamine in nutritional therapy is supported by multiple studies which reflect the beneficial effect of this nutrient, both in enteral nutrition as in parenteral nutrition.  相似文献

The multi-compartment vehicle routing problem (MC-VRP) consists of designing transportation routes to satisfy the demands of a set of customers for several products that, because of incompatibility constraints, must be loaded in independent vehicle compartments. Despite its wide practical applicability the MC-VRP has not received much attention in the literature, and the few existing methods assume perfect knowledge of the customer demands, regardless of their stochastic nature. This paper extends the MC-VRP by introducing uncertainty on what it is known as the MC-VRP with stochastic demands (MC-VRPSD). The MC-VRPSD is modeled as a stochastic program with recourse and solved by means of a memetic algorithm. The proposed memetic algorithm couples genetic operators and local search procedures proven to be effective on deterministic routing problems with a novel individual evaluation and reparation strategy that accounts for the stochastic nature of the problem. The algorithm was tested on instances of up to 484 customers, and its results were compared to those obtained by a savings-based heuristic and a memetic algorithm (MA/SCS) for the MC-VRP that uses a spare capacity strategy to handle demand fluctuations. In addition to effectively solve the MC-VRPSD, the proposed MA/SCS also improved 14 best known solutions in a 40-problem testbed for the MC-VRP.  相似文献

In this paper, Bayesian network (BN) and ant colony optimization (ACO) techniques are combined in order to find the best path through a graph representing all available itineraries to acquire a professional competence. The combination of these methods allows us to design a dynamic learning path, useful in a rapidly changing world. One of the most important advances in this work, apart from the variable amount of pheromones, is the automatic processing of the learning graph. This processing is carried out by the learning management system and helps towards understanding the learning process as a competence-oriented itinerary instead of a stand-alone course. The amount of pheromones is calculated by taking into account the results acquired in the last completed course in relation to the minimum score required and by feeding this into the learning tree in order to obtain a relative impact on the path taken by the student. A BN is used to predict the probability of success, by taking historical data and student profiles into account. Usually, these profiles are defined beforehand; however, in our approach, some characteristics of these profiles, such as the level of knowledge, are classified automatically through supervised and/or unsupervised learning. By using ACO and BN, a fitness function, responsible for automatically selecting the next course in the learning graph, is defined. This is done by generating a path which maximizes the probability of each user??s success on the course. Therefore, the path can change in order to adapt itself to learners?? preferences and needs, by taking into account the pedagogical weight of each learning unit and the social behaviour of the system.  相似文献

Service Oriented Architectures, which allow for the integration of different subsystems and of applications running on different devices and platforms, may be very suitable to solve the problem of service personalization in large smart environments like cities, where the number of potential users and potentially available services is rapidly growing. Taking this into account, we have designed an agent-based service oriented architecture for smart spaces. Two of the greatest challenges in the design of such a solution are providing effective device, service and context federation and composition mechanisms and handling user mobility. For the first challenge, we have designed a hierarchical architecture and developed a set of inheritance, aggregation and access mechanisms for devices, services and context. To handle user mobility, we have followed three different strategies, using stationary, mobile or nomadic agents. In this paper, we describe the main aspects of our architecture and perform an experimental evaluation to determine the advantages and drawbacks of the different strategies.  相似文献

Recycled PET/organoclay nanocomposites were prepared by melt intercalation process with several amounts (1, 3, and 5 wt %) of clay modified with quaternary ammonium salt (DELLITE 67G) dispersed in a recycled poly(ethylene terephthalate) (rPET) matrix. The resultant mechanical properties (modulus and yield strength) of the nanocomposites were found to be different from those of rPET. Wide angle X‐ray scattering (WAXS) and Transmission Electron Microscopy (TEM) measurements have shown that although complete exfoliation was not achieved, delaminated clay platelets could be observed. Thermal analysis did not show significant changes in the thermal properties from those of recycled PET. Mechanical testing showed that nanocomposite properties were superior to the recycled PET in terms of strength and elasticity modulus. This improvement was attributed to nanoscale effects and strong interaction between the rPET matrix and the clay interface, as revealed by WAXS and TEM. © 2007 Wiley Periodicals, Inc.
