Primer walking of cloned DNA is a standard research tool. It has been used in the past to determine the sequence of individual clones of interest. With the expansion of DNA sequencing capacity the need to be able to walk larger numbers of clones has become necessary. Our laboratory is a mid-sized genomics facility. In conjunction with the Advanced Biomedical Computing Center (ABCC) we have developed methods for automating the primer selection, DNA sequencing, contig assembly and sequence analysis for clones arrayed in microtiter format. This approach has allowed us to walk 475 clones (five microtiter plates) selected from a cDNA library. 相似文献

An analysis method, based on the numerical inversion of the Laplace transform, is described for the evaluation of the time domain sensitivity of networks that include lossy coupled transmission lines. The sensitivity can be calculated with respect to network components and parameters of the transmission lines. Sensitivity analysis is useful for waveform shaping and optimization. Electrode pitting was investigated in resistance spot welding of 1.5-mm-thick sheet aluminum alloy 5182 using a medium-frequency
direct-current welder and electrodes with a tip face curvature radius of 50 mm and tip face diameter of 10 mm. Detailed investigation
of the metallurgical interactions between the copper electrode and aluminum alloy sheet was carried out using scanning electron
microscopy/energy-dispersive X-ray spectroscopy (SEM/EDX) and X-ray diffraction (XRD). The results indicated that electrode
degradation, which eventually leads to weld failure, proceeded in four basic steps: aluminum pickup, electrode alloying with
aluminum, electrode tip face pitting, and cavitation. Since pitting and cavitation result from Al pickup and alloying, periodic
electrode cleaning could extend electrode tip life by limiting the buildup of Al on the tip face. This work is part of the

STUDY OBJECTIVE: To validate the utility of a previously reported 3-point limited sampling model (LSM) for determining etoposide area under the curve to infinity (AUC(infinity)). DESIGN: Secondary analysis of data from two clinical trials of etoposide. SETTING: University medical center clinical research center. PATIENTS: Thirty-four patients with different malignancies. INTERVENTIONS: Etoposide was administered as a 2-hour infusion to 34 patients. Serial plasma samples were drawn over 24 hours after the infusion and analyzed for etoposide by high-performance liquid chromatography. MEASUREMENTS AND MAIN RESULTS: The 3-point LSM AUC was compared with a 14-point actual AUC calculated by the linear trapezoidal rule. Actual and predicted AUC(infinity) by the LSM were highly correlated (r=0.97, p<0.0001). The LSM predictions had a mean absolute error of 10.9% (95% CI -14.1, -5.3) and a mean error of -9.7% (95% CI 6.9, 14.9). Nine patients with poor AUC(infinity) estimations by the LSM (error > 12%) tended to have abnormally low or high peak concentrations. CONCLUSION: Our findings suggest the development of more robust LSM using other techniques, such as pharmacostatistical models, that can accommodate a greater degree of pharmacokinetic variability. 相似文献

A multiplanar positron camera is proposed, made of six MWPC modules, arranged to form the lateral surface of a hexagonal prism. Each 50 x 50 cm(2) module has a single MWPC sandwiched by two 2-cm thick lead glass tube converters. The experimental results for a 15 x 15 cm(2) test module are reported. For 511 keV gamma-rays incident almost perpendicular onto a 1.0-cm thick converter, a detection efficiency of 4.3%, a time resolution of 130 ns (FWHM) and a spatial resolution of 2.8 mm (FWHM) have been measured with a standard Argon-Methane (70-30) mixture at 1.2 atm. The chamber may also be operated in high resolution mode: 1.2-mm (FWHM) spatial resolution has been measured at a 50% lower efficiency. The use of fast delay lines (specific delay 8 ns/cm) for the position read-out ensures a high rate capability. The expected performance of the six-module MWPC camera is discussed and compared with that of a BGO crystal ring camera. We address the challenges of building a good content adaptation service for mobile devices and propose a decision engine that is user-centric with QoS awareness, which can automatically negotiate for the appropriate adaptation decision to use in the synthesis of an optimal adapted version. The QoS-sensitive approach complements the lossy nature of the transcoding operations. The decision engine will look for the best trade off among various parameters in order to reduce the loss of quality in various domains. Quantitative methods are suggested to measure the QoS of the content versions in various quality domains. Based on the particular user perception and other contextual information on the client capability, the network connection, and the requested content, the proposed negotiation algorithm will determine a content version with a good aggregate score. Titania precipitation on cotton has been achieved using a commercially available antimicrobial polymer, Reputex 20. The cotton swatches precipitated with titania retain antimicrobial activity, and we have also shown the ability to encapsulate diisopropylfluorophosphatase (DFPase), an enzyme capable of breaking down organofluorophosphates. Cotton swatches are easily prepared and precipitation occurs at room temperature in aqueous solutions at neutral pH. Both the antimicrobial properties of Reputex 20 and the hydrolytic activity of the DFPase enzyme are retained after titania precipitation, generating a cotton material exhibiting multifunctional properties. © 2015 Wiley Periodicals, Inc.
