Vacuum skin packaging (VSP) is a new and promising technology that includes the instantaneous heating of the upper packaging film immediately before its descent over the meat surface. In the present paper we evaluated this technology for the packaging of a wide variety of beef retail cuts and compared this with traditional vacuum packaging (TVP) performed in parallel. Shelf-life and quality evolution was estimated by analysing three spoilage bacterial groups, an ubiquitous psychrotrophic food pathogen, and three biochemical and physico-chemical attributes throughout 40 days of storage at 4 °C. Statistically-significant lower microbial counts were obtained for all microbial parameters and meat types. The VSP lot exhibited lower pH. This, however, involved a lower water holding capacity and a higher degree of firmness, the latter indicating a limited tenderisation process in the VSP batch. Extended aging times in the carcasses were studied to improving the tenderness of VSP samples. The extension of the aging process prior to VSP may be useful for improving the tenderness of certain meat types and to improve the sensory quality of meat cuts processed under this system. 相似文献

Groundwater resources are in many parts of the world the only source for private domestic, agricultural and public water supply.
Subsurface has also become major recipient of wastewater and solid waters especially in the developing countries. Overexploitation
of coastal aquifers and pollution are among the main problems related to groundwater resources assessment and management in
Santiago Island (Cabo Verde). Brackish groundwater is the only available water type in the region that is being provided to
numerous parts of the Island such as Praia Baixo, Montenegro, and Charco for agriculture and human supply. Solute and isotope
data obtained in different groundwater systems were used in the identification of groundwater resources degradation. In order
to understand the influence of the anthropogenic activities on the water quality and the main origin of the salts in groundwater,
a statistical approach (Principal Components Analyses—PCA) was performed on the physico-chemical data. The results obtained
indicate water–rock interaction mechanisms as the major process responsible for the groundwater quality (mainly calcium-bicarbonate
type), reflecting the lithological composition of the subsurface soil. Also, anthropogenic contamination was identified, in
several points of the island. Isotopic techniques (δ2H, δ18O and 3H content) combined with geochemistry provided comprehensive information on groundwater recharge, as well as on the identification
of salinization mechanisms (e.g. seawater intrusion, salt dissolution, and marine aerosols) of the groundwater systems, at

Enolase, a glycolytic enzyme that catalyzes the dehydration of 2-phospho-d-glycerate (PGA) to form phosphoenolpyruvate (PEP), is a homodimer in all eukaryotes and many prokaryotes. Here, we report the crystal structure of a complex between yeast enolase and an equilibrium mixture of PGA and PEP. The structure has been refined using 29 854 reflections with an F/sigma(F) of >/=3 to an R of 0.137 with average deviations of bond lengths and bond angles from ideal values of 0.013 A and 3.1 degrees , respectively. In this structure, the dimer constitutes the crystallographic asymmetric unit. The two subunits are similar, and their superposition gives a rms distance between Calpha atoms of 0.91 A. The exceptions to this are the catalytic loop Val153-Phe169 where the atomic positions in the two subunits differ by up to 4 A and the loop Ser250-Gln277, which follows the catalytic loop Val153-Phe169. In the first subunit, the imidazole side chain of His159 is in contact with the phosphate group of the substrate/product molecule; in the other it is separated by water molecules. A series of hydrogen bonds leading to a neighboring enolase dimer can be identified as being responsible for ordering and stabilization of the conformationally different subunits in the crystal lattice. The electron density present in the active site suggests that in the active site with the direct ligand-His159 hydrogen bond PGA is predominantly bound while in the active site where water molecules separate His159 from the ligand the binding of PEP dominates. The structure indicates that the water molecule hydrating carbon-3 of PEP in the PEP --> PGA reaction is activated by the carboxylates of Glu168 and Glu211. The crystals are unique because they have resolved two intermediates on the opposite sides of the transition state. 相似文献

We have previously characterized the release of the signal peptide sequence-less fibroblast growth factor (FGF) prototype, FGF-1, in vitro as a stress-induced pathway in which FGF-1 is released as a latent homodimer with the p40 extravesicular domain of p65 synaptotagmin (Syn)-1. To determine the biologic relevance of the FGF-1 release pathway in vivo, we sought to resolve and characterize from ovine brain a purified fraction that contained both FGF-1 and p40 Syn-1 and report that the brain-derived FGF-1:p40 Syn-1 aggregate is associated with the calcium-binding protein, S100A13. Since S100A13 binds the anti-inflammatory compound amlexanox and FGF-1 is involved in inflammation, we examined the effects of amlexanox on the release of FGF-1 and p40 Syn-1 in response to stress in vitro. We report that while amlexanox was able to repress the heat shock-induced release of FGF-1 and p40 Syn-1 in a concentration-dependent manner, it had no effect on the constitutive release of p40 Syn-1 from p40 Syn-1 NIH 3T3 cell transfectants. These data suggest the following: (i) FGF-1 is associated with Syn-1 and S100A13 in vivo; (ii) S100A13 may be involved in the regulation of FGF-1 and p40 Syn-1 release in response to temperature stress in vitro; and (iii) the FGF-1 release pathway may be accessible to pharmacologic regulation. 相似文献

Can computer science learn from the social sciences? We focus our discussions on an undesired phenomenon that commonly affects software developers. Known as the Pygmalion effect, this phenomenon has been studied intensely by social scientists but almost entirely ignored by computer scientists. Pygmalion was a sculptor who fell in love with his statue, Galatea, brought to life for him by Aphrodite. In the 1960s, R. Rosenthal and L. Jacobson, two American psychologists, used this myth to name an observation of theirs: whenever someone evaluates something, the evaluator's expectations concerning the evaluated object influence the evaluation, in a way that tends to prove the evaluator's initial hypothesis. Intensive studies since then have confirmed this initial observation: virtually every evaluation process in which humans intervene is prone to the Pygmalion effect, and computer science is no exception: the expectations of computer systems evaluators can strongly bias the outcome of the evaluation 相似文献

The aim of this study was to evaluate our results in treatment and management of symptomatic hepatic venous malformations using transcatheter embolization therapy. From 1991 to 1997 five venous malformations were embolized in one man and four women ranging in age from 31 to 50 years. All patients presented nonspecific abdominal pain and were assessed in the general surgery unit. Percutaneous embolization was recommended. In all cases polyvinyl alcohol was used to embolize the afferent arterioles. Clinical and echographic control follow-up was carried out on an outpatient basis in our Vascular and Interventional Radiology Unit. Embolization was possible in all cases. No complications developed during the procedure, except in one case where the subject suffered a spasm of the hepatic artery. Mean follow-up time was 42 months (6-73 months). Four patients remained asymptomatic during the entire follow-up period, whereas one patient required reembolization. In our experience, transcatheter embolization of hepatic venous malformations is a noninvasive technique which requires few admissions and presents few complications. With further use, it could become the treatment of choice in symptomatic hepatic venous malformations as an alternative to surgery. 相似文献

The heparin-binding fibroblast growth factor (FGF) prototypes lack a classical signal sequence, yet their presence is required in the extracellular compartment for the activation of cell-surface receptor-dependent signaling. Early studies with FGF-1 demonstrated its presence in bovine brain as a novel high molecular weight complex, and subsequent studies identified a second heparin-binding protein that co-purified with FGF-1. Polypeptide sequence analysis revealed that this heparin-binding protein corresponded to the extravesicular domain of bovine synaptotagmin (Syn)-1, a transmembrane component of synaptic vesicles involved in the regulation of organelle traffic. Since FGF-1 is released in response to heat shock as a mitogenically inactive Cys-30 homodimer, we sought to determine whether this heparin-binding protein was involved in the release of FGF-1. We report that a proteolytic fragment of the extravesicular domain of Syn-1 is associated with FGF-1 in the extracellular compartment of FGF-1-transfected NIH 3T3 cells following temperature stress. By using heparin-Sepharose affinity to discriminate between the monomer and homodimer forms of FGF-1 and resolution by conventional and limited denaturant gel shift immunoblot analysis, it was possible to identify FGF-1 and Syn-1 as potential components of a denaturant- and reducing agent-sensitive extracellular complex. It was also possible to demonstrate that the expression of an antisense-Syn-1 gene represses the release of FGF-1 in response to heat shock. These data indicate that FGF-1 may be able to utilize the cytosolic face of conventional exocytotic vesicles to traffic to the inner surface of the plasma membrane where it may gain access to the extracellular compartment as a complex with Syn-1. 相似文献
