The results of combined epidemiological, clinical, serological, bacteriological and histopathological studies following an outbreak of disease caused by L. pomona on a farm stocked with cattle, sheep, pigs, goats and horses maintained for experimental purposes, are reported.
The incidence of infection was high in horses, cattle and pigs. A few low titres were seen in sheep. The goats were not infected. Apart from a single bovine abortion all the clinical symptoms observed occurred in pregnant sows. Seven of these aborted or gave birth to stillborn pigs within a six week period.
Fifteen species of wildlife were trapped or shot on the farm during the year following the outbreak. L. pomona was isolated from four skunks and a porcupine. Epidemiological studies indicated that wildlife reservoir hosts were the primary source of infection for the domestic livestock.
Leptospiruria and the serological response were studied in a group of eight infected sows. Microscopic agglutination titres of 102 or less could not be associated with leptospiruria and the duration of leptospiruria was found to range from a few weeks to over two years in individual sows. Direct dark-field examination of urine proved superior to guinea-pig inoculation as a method of detecting leptospiruria and it is suggested that the former technique could be adopted with advantage as a routine aid to diagnosis.
Isolation of transmissible gastroenteritis virus was attempted from segments of jejunum collected from piglets submitted for diagnosis of transmissible gastroenteritis. The virus was isolated more frequently in susceptible piglets than in pig kidney or pig thyroid cells. Practically, both cell systems were equally capable of demonstrating the virus when the tissue suspensions were sonicated. The pig thyroid cells prepared with glands collected from minimal disease pigs were preferred to the pig kidney cells for initial virus isolation because of their ability to respond to transmissible gastroenteritis virus with a progressive cytopathic effect. However, the pig thyroid cells, prepared from pool of glands collected in abattoirs, were often contaminated with parvoviruses and could not be used for diagnostic work. A Canadian isolate of porcine parvovirus, isolated from cultured pig thyroid cells, was shown to be antigenically indistinguishable from a British (59e/63) and a German (G10/1) strain when treated by the modified direct complement-fixation, the hemagglutination-inhibition and the fluorescent antibody tests. These tests also revealed that antibodies to parvoviruses were detectable in a large proportion of the conventionally raised pigs in the provinces of Quebec and Ontario. Cell cultures, prepared from tissues collected in a slaughterhouse, were often found to be infected with parvovirus. In cell cultures the infection was demonstrated more effectively by immunofluorescence than by the hemagglutination test. 相似文献
Class I major histocompatibility complex (class I MHC) molecules, known to be important for immune responses to antigen, are expressed also by neurons that undergo activity-dependent, long-term structural and synaptic modifications. Here, we show that in mice genetically deficient for cell surface class I MHC or for a class I MHC receptor component, CD3zeta, refinement of connections between retina and central targets during development is incomplete. In the hippocampus of adult mutants, N-methyl-D-aspartate receptor-dependent long-term potentiation (LTP) is enhanced, and long-term depression (LTD) is absent. Specific class I MHC messenger RNAs are expressed by distinct mosaics of neurons, reflecting a potential for diverse neuronal functions. These results demonstrate an important role for these molecules in the activity-dependent remodeling and plasticity of connections in the developing and mature mammalian central nervous system (CNS). 相似文献
As a result of diverse management policies, densities of red and roe deer have varied considerably for the last few decades in Europe. Their selective browsing on woody species can modify the interactions between competing species and thus change overall patterns of plant diversity in forests. However, no classification of woody species according to browsing by deer yet exists in Europe. The selectivity index, which measures the balance between availability and consumption of plant species by herbivores, could be used for such a classification. The aim of this study was, first, to rank woody species according to this selectivity index, then to test whether their rank changed when browsing pressure varied in space or time. We also explored morphological and ecological life traits (Ellenberg indicator values) that might explain differences in selectivity.In 2006, we re-sampled a network of 217 coupled floristic and browsing survey plots, which had been previously described twice (1976, 1981). The plots were located in the forest of Arc-en-Barrois (France), a large woodland area divided into a North and a South Forest which have had different histories of browsing pressure.For the three samples and the two forests, we calculated a selectivity index for 19 woody species. Dogwood (Cornus sp.) and field rose (Rosa arvensis) appeared to be the most selected species whereas beech (Fagus sylvatica), common mezereon (Daphne mezereum) and wild-service tree (Sorbus torminalis) were always avoided. The rank order of species according to their selectivity index remained stable over the three sample years and between the two forests (Spearman's rank correlations: ρ ≈ 0.8 spatially, and range from 0.3 to 0.8 temporally). This indicates that variations in browsing pressure did not strongly modify deer selectivity patterns. Selectivity was positively correlated with wood density and negatively correlated with maximum height of the species, but unrelated to other plant traits. Finally, selectivity can be seen as an intrinsic characteristic of those woody species, and could be used as an additional life trait when analyzing vegetation communities. 相似文献
Clinical field cases of equine infectious anemia were studied and the disease was reproduced experimentally in horses. Attempts were made to adapt the complement-fixation test to the detection of antibodies in the serum of infected animals and to the demonstration of antigens in tissue extracts.
A moderate complement-fixing antibody response was demonstrated in the serum of horses shortly after primary exposure to the infectious agent. However, this reactivity was of short duration and occurred with normal as well as with infected saline tissue extracts. It was therefore concluded that this reaction was not specific for equine infectious anemia. Possibly it is due to the appearance of auto-tissue antibodies. The value of this reaction in the diagnosis of the infection was limited because of its short duration and absence in chronic infection and following re-exposure to the infectious agent.
Sera of sheep and calves infected with the California type 10 and Cyprus type 3 viruses of bluetongue were tested by the regular and modified direct complement-fixation tests. To obtain satisfactory complement fixation it was necessary to use the latter test. Cross reactivity was found, therefore, the California type 10 antigen could be used in testing sera of animals infected with the Cyprus virus. 相似文献
