Nonionic microemulsions containing triglycerides and fatty acid esters as lipophilic components have been studied. The phase inversion temperature (PIT) of the systems was determined by a conductometric method. Partial phase diagrams were constructed in the phase inversion temperature range. Water solubilization capacity of the nonionic surfactant systems studied was dependent on surfactant and oil types in analogy to ordinary hydrocarbon systems. The PIT:s increased with increased molecular weight for both esters and triglycerides.  相似文献

Inclusion distribution was studied in commercial aluminum DC-cast billets and slabs using a newly developed deep-etching method. Analyses revealed a nonuniform distribution of nonmetallic inclusions across billet diameters and lengths, and also across slab thicknesses and widths. In as-cast billets, more inclusions were found at the beginning and end of the billet length; more were present near the cross-section center than near the surface. In slabs, inclusions were located mostly within 13 mm of the surface and in a band between the centerline and the surface. Few inclusions were found 60 to 100 mm from the slab surface or at the centerline. In addition, comparing slab quality after casting using three types of ceramic foam filters (CFFs; i.e., 30 ppi, 50 ppi, and 50 ppi + HF) revealed significant differences in inclusion size, number, and distribution. Casting slabs using a finer pore-size filter (50 ppi) reduced the number of non-metallic inclusions greatly. The inclusion distribution patterns observed in the solidified slabs are discussed in terms of melt flow during casting.  相似文献

We have created protein domains with extreme surface charge.These mutated domains allow for ion-exchange chromatographyunder conditions favourable for selective and efficient capture,using Escherichia coli as a host organism. The staphylococcalprotein A-derived domain Z (Zwt) was used as a scaffold whenconstructing two mutants, Zbasic1 and Zbasic2, with high positivesurface charge. Far-ultraviolet circular dichroism measurementsshowed that they have a secondary structure content comparableto the parental molecule Zwt. Although melting temperatures(Tm) of the engineered domains were lower than that of the wild-typeZ domain, both mutants could be produced successfully as intracellularfull-length products in E.coli and purified to homogeneity byion-exchange chromatography. Further studies performed on Zbasic1and Zbasic2 showed that they were able to bind to a cation exchangereven at pH values in the 9 to 11 range. A gene fusion betweenZbasic2 and the acidic human serum albumin binding domain (ABD),derived from streptococcal protein G, was also constructed.The gene product Zbasic2–ABD could be purified using cation-exchangechromatography from a whole cell lysate to more than 90% purity.  相似文献

Nickel sulfide precipitates produced by the AMAX Acid Leach Process for oxide nickel ores were leached in hydrochloric acid. The effects of process variables such as temperature, acid concentration, stoichiometric excess of HCl, gas sparging and heat treatment of feed were investigated. The nickel leachability was found to be in the 60–80% range. Chemical and mineralogical examination of the leach residues indicated the presence of NiS2. This higher nickel sulfide is insoluble in hydrochloric acid, and its presence hinders the leaching of NiS. Several methods are suggested to reduce the sulfur content in order to attain complete dissolution. The thermodynamics and kinetics of nickel sulfide leaching are briefly discussed.  相似文献

The macrophage colony-stimulating factor receptor and several other hematopoietic growth factor receptors induce the tyrosine phosphorylation of a 145- to 150-kD protein in murine cells. We have previously cloned a cDNA for the murine 150-kD protein, SHIP, and found that it encodes a unique signaling intermediate that binds the SHC PTB domain through at least one tyrosine phosphorylated (NPXY) site in the carboxyl-terminal region. SHIP also contains several potential SH3 domain-binding sites, an SH2 domain for binding other tyrosine phosphorylated proteins, and an enzymatic activity that removes the phosphate from the 5 position of phosphatidylinositol 3,4,5-phosphate or from inositol 1,3,4,5-phosphate. SHIP has a negative effect on cell growth and therefore loss or modification may have profound effects on hematopoietic cell development. In this study, we have cloned a cDNA for human SHIP and examined mRNA and protein expression of SHIP and related species in bone marrow and blood cells. Flow cytometry indicates that at least 74% of immature CD34+ cells express SHIP cross-reacting protein species, whereas within the more mature population of CD33+ cells, only 10% of cells have similar expression. The majority of T cells react positively with the anti-SHIP antibodies, but significantly fewer B cells are positive. Immunoblotting detects up to seven different cross-reacting SHIP species, with peripheral blood mononuclear cells exhibiting primarily a 100-kD protein and a CD34+ acute myeloblastic leukemia expressing mainly 130-kD and 145-kD forms of SHIP. Overall, these results indicate that there is an enormous diversity in the size of SHIP or SHIP-related mRNA and protein species. Furthermore, the expression of these protein species changes according to both the developmental stage and differentiated lineage of the mature blood cell.  相似文献

Reverberation chambers can be used to measure the absorption cross section of a dielectric object. The absorption cross section of a dielectric object depends on its size, shape, and electrical material parameters. By comparing with a theoretical model of the absorption cross section, material parameters can be extracted from measurements. A model based on a plane wave approach of incident fields is used here, valid for electrically large material samples in an isotropic environment such as that in a reverberation chamber. Which material parameter can be extracted depends on the properties of the material sample. The presented method combines the accuracy of cavity methods with the flexibility of being able to measure samples of arbitrary size and shape. Because both the reverberation chamber and the material sample are electrically large, the method is particularly useful at millimeter-wave frequencies.  相似文献

A numerical model of wires in rectangular metal cavities is introduced for computing antennas in a simplified theoretical reverberation chamber. The code is based on the method of moments, and it uses the Ewald summation for efficient calculation of the cavity Green's function. The Q-value of the chamber is accounted for in the model by a homogeneous material filling the chamber. The S-parameters of two dipoles placed in many random positions in the cavity are computed and averaged to provide the average transfer function of the chamber. This represents mode stirring by moving the antennas, referred to as position stirring. The computed results are compared with theoretical statistical values as well as experimental results. The discrepancies are discussed.  相似文献

A sensitive short-term mutagenicity test, the microscale fluctuation test has been coupled with a concentration method based on adsorption on Sep-PakR C18 cartridges as a method for screening drinking water mutagens. Comparison with XAD-2 concentration method showed that Sep-Pak adsorbed 5 times higher quantity of organics but was slightly less efficient for adsorbing TOX.Microscale fluctuation test was found to be more sensitive than Ames test by testing known direct-acting mutagens and concentrates of drinking water. Samples derived from conventional treatment including chlorination from eight surface water supplies in Norway were concentrated at pH 2 by adsorption on the disposable columns. The adsorbates were tested at different doses by the microscale fluctuation assay. The mutagenic properties of drinking water samples were also related to total organic carbon (TOC), total organic halogen (TOX) and trihalomethanes (THM) concentrations. Dose-related mutagenic responses were found for all the samples with S. typhimurium TA 100 and TA98 strains without metabolic activation. Good relationship was found between mutagenicity data and TOX and THM results. The method showed to be simple, rapid and suitable for routine screening of mutagens in drinking water.  相似文献
