This paper describes an experimental and analytical study on fracture and damage behavior of GFRP woven laminates at cryogenic temperatures. CT (compact tension) tests were carried out at room temperature, liquid nitrogen temperature (77 K) and liquid helium temperature (4 K) to evaluate the critical values of the fracture mechanics parameters. During the CT tests, AE (acoustic emission) method was implemented. AE signals can identify the critical load at which gross failure occurs. A FEA (finite element analysis) was also applied to calculate the fracture mechanics parameters. The failure criteria (Hoffman criterion and maximum strain criterion) or the damage variable based on the continuum damage mechanics was incorporated into the model to interpret the experimental measurements and to study the damage distributions within the specimen. Several methods of calculating J-integral are discussed.  相似文献

Hereditary argininemia manifests as neurological disturbance and mental retardation, features not observed in other amino acidemias. The cytotoxic effect of a high concentration of L-arginine (L-Arg) was investigated using NB9 human neuroblastoma cells (NB9), which express neuronal nitric oxide synthase (nNOS). When the concentration of L-Arg in the medium increased from 50 microM to 2 mM after incubation for 48 hr, the intracellular concentration of L-Arg increased from 68.0 +/- 1 pmol/10(6) cells to 1310.0 +/- 5 pmol/10(6) cells and that of L-citrulline (L-Cit) from undetectable levels to 47.1 +/- 0.2 pmol/10(6) cells (mean +/- SD of three independent analyses). This increase in intracellular L-Arg levels caused a decrease in NOS activity by approximately 71%. Flow cytometric analysis showed that reactive oxygen species (ROS) are produced in NB9 exposed to 2 mM L-Arg. The production of ROS was abolished by a NOS inhibitor, NG-nitro-L arginine-methylester. Production of ROS was also observed when NB9 were treated with L-Cit for 48 hr. To investigate the effect of L-Cit on the activity of NOS, a kinetic study on nNOS was conducted using cellular extracts from NB9. The apparent Km value of nNOS for L-Arg was 8.4 microM, with a Vmax value of 8.2 pmol/min/mg protein. L-Cit competitively inhibited NOS activity, as indicated by an apparent Ki value of 65 nM. These results suggest that L-Cit formed by nNOS in L-Arg-loaded neuronal cells inhibits NOS activity and nNOS in these L-Arg-loaded cells functions as a NADPH oxidase to produce ROS, which may cause neurotoxicity in argininemia.  相似文献

The contribution of hydrogen bonds to the conformational stability of human lysozyme was investigated by the combination of calorimetric and X-ray analyses of six Tyr --> Phe mutants. Unfolding Delta G and unfolding Delta H values of the Tyr --> Phe mutant proteins were changed by from +0.3 to -4.0 kJ/mol and from 0 to -16 kJ/mol, respectively, compared to those of the wild-type protein. The net contribution of a hydrogen bond at a specific site to stability (Delta Gwild/HB), considering factors affected by substitutions, was evaluated on the basis of X-ray structures of the mutant proteins. In the present study, one of six mutant proteins was suitable for evaluating the strength of the hydrogen bond. Delta Gwild/HB for the intramolecular hydrogen bond at Tyr124 was evaluated to be 7.5 kJ/mol. Results of the analysis of other mutants also suggest that hydrogen bonds of the hydroxyl group of Tyr, including the hydrogen bond with a water molecule, contribute to the stabilization of the human lysozyme.  相似文献

Previously we reported than BO-IMI, a reagent which contains a BODIPY fluorophore linked to an imidazole group, can be used to covalently label a phosphomonoester in a single step under aqueous conditions [P. Wang, R.W. Giese, Anal. Chem. 65 (1993) 3518]. The reaction was conducted in the presence of a water-soluble carbodiimide 1-ethyl-3-(3'-N,N'-dimethylaminopropyl) carbodiimide [EDC] to activate the phosphomonoester, and the coupling took place onto both the N1 and N3 imidazole nitrogens of BO-IMI. Whether the two BO-IMI-phosphomonoester regioisomers migrated separately or together during capillary electrophoresis depended on the pH, due to a difference in their pKa values. Since then, we have studied the reaction in more detail leading to the information reported here. First, we have learned that the regioisomer ratio changes during the course of the reaction, and found that the mechanism involves both spontaneous and BO-IMI-catalyzed hydrolysis of the less stable isomer. Second, there is a background reaction in which BO-IMI becomes attached to EDC. Third, the BO-IMI-phosphomonoester product (a mixture of two isomers), that is observed by capillary electrophoresis at an alkaline pH, is found to no longer contain the two fluorine atoms present in the starting BO-IMI reagent. This is because they are placed by hydroxy groups at high pH. Finally, an event was discovered which complicates the detection of less than about 60 fmol of a phosphomonoester with BO-IMI: hydrolysis of a tiny fraction of the BO-IMI takes place during the coupling reaction, which leads to chemical noise in the capillary electropherogram.  相似文献

STUDY OBJECTIVES: To evaluate renal function during and after hypotensive anesthesia with sevoflurane compared with isoflurane in the clinical setting. DESIGN: Randomized, prospective study. SETTING: Inpatient surgery at Rosai Hospital. PATIENTS: 26 ASA physical status I and II patients scheduled for orthopedic surgery. INTERVENTIONS: Patients received isoflurane, nitrous oxide (N2O), and fentanyl (Group I = isoflurane group; n = 13) or sevoflurane, N2O, and fentanyl (Group S = sevoflurane group; n = 13). Controlled hypotension was induced with either isoflurane or sevoflurane to maintain mean arterial pressure at 60 mmHg for 120 minutes. MEASUREMENTS AND MAIN RESULTS: Measurements included serum inorganic fluoride (previously speculated to influence renal function), creatinine clearance (CCr; to assess renal glomerular function), urinary N-acetyl-beta-D-glucosaminidase (NAG; to assess renal tubular function), blood urea nitrogen (BUN), and serum creatinine (as clinical renal function indices). Serum fluoride, CCr, and NAG were measured before hypotension, 60 minutes, and 120 minutes after the start of hypotension, 30 minutes after recovery of normotension, and on the first postoperative day. BUN and serum creatinine were measured preoperatively and on the third and seventh postoperative days. Minimum alveolar concentration times hour was 3.6 +/- 1.8 in Group I and 4.0 +/- 0.7 in Group S. In both groups, BUN and serum creatinine did not change, and CCr significantly decreased after the start of hypotension. In Group I, serum fluoride and NAG did not change. In Group S, serum fluoride significantly increased after the start of hypotension compared with prehypotension values and compared with Group I values. In addition, NAG significantly increased at 120 minutes after the start of hypotension and at 30 minutes after recovery of normotension, but returned to prehypotension values on the first postoperative day. CONCLUSIONS: Two hours of hypotensive anesthesia with sevoflurane under 5 L/min total gas flow in patients having no preoperative renal dysfunction transiently increased NAG, which is consistent with a temporary, reversible disturbance of renal tubular function.  相似文献

To explain contact failure phenomena and their development in the gas-insulated switchgear (GIS). tests were conducted by simulating abnormal current-carrying conditions using an actual 300-kV, 2000-A GIS. This paper describes the process observed. ranging from contact failures to ground faults, deteriorative phenomena and some detection methods for initial contact failure conditions. A summary of this study is as follows: (1) The contact resistances are greatly dependent on the materials and construction of contacts. (2) The relation between the initial contact resistance and the carried current resulting in arcing is derived with approximately constant Joule-heat generation. (3) Ground faults result from sagging of a shield melted by arcing or a combination of two causes: (i) scattering of molten metal caused by arcing; and (ii) sagging of a conductor caused by bolt melt down. (4) To detect initial contact failures, it is recommended to observe the contact resistance, the temperature rise on the tank surface, or the electrical partial discharges.  相似文献

The chip size package (CSP) is being used in various portable electronic products recently. Further evaluation of the reliability of its soldered joints is required all the more now because those soldered joints are invisible. This study focused on the thermal fatigue life of soldered joints in the CSP. CSPs were mounted on printed circuit boards (PCBs) in various configurations and mounting conditions, and underwent thermal cycle testing. Then, the fatigue lives of their soldered joints were compared. As a result, the following two facts became apparent. First, reflowing at a 210°C peak tends to result in failures that may be derived from poor wetting between solder and pad, in cases where the CSP is mounted on a nickel and gold plated pad. And second, the size of the soldered joint has a great influence on its fatigue life. The larger the soldered joints that we made, the longer fatigue life they indicated. A finite element method (FEM) analysis of those mounted structures was also executed. Viscoplastic (creep and plastic) property of solder was evaluated to compute equivalent inelastic strain occurring in the joints. A parameter in the Coffin-Manson equation is obtained from the computed inelastic strain amplitudes and the experimented actual fatigue lives. This result will enable estimation of the fatigue life of soldered joints of the CSP without actual tests  相似文献

The effects of arachidonic acid on ACh-gated channel currents were examined using Torpedo nicotinic ACh receptors expressed in Xenopus oocytes. Arachidonic acid decreased ACh-evoked currents during treatment, to a greater extent in Ca(2+)-free extracellular solution. The currents were enhanced for more than 30 min after washing, reaching 150 and 170% in Ca(2+)-containing and -free extracellular solutions, respectively. The current enhancement was inhibited by the selective protein kinase C (PKC) inhibitor, GF109203X, whereas the current depression was not affected. Furthermore, arachidonic acid-evoked current depression was blocked in mutant ACh receptors with PKC phosphorylation site deletions on the alpha and delta subunits, but the long-lasting potentiation effect remained. These results indicate that arachidonic acid may decrease ACh receptor currents by a direct binding to PKC phosphorylation sites of the ACh receptors and may potentiate the currents via a novel pathway related to arachidonic acid-regulated PKC activation, but not via PKC phosphorylation of the ACh receptor itself.  相似文献

Studies on the structure and substrate specificity of purified rat kidney nuclear (RKN) lysozyme are reported. The carboxyl and amino terminal residues of RKN-lysozyme were found to be leucine and alanine respectively. The amino acid composition indicated similarities and differences as compared with that of hen egg white (HEW) lysozyme. There were alterations in the nine amino acid residues, Lys, His, Arg, Asp, Glu, Pro, 1/2 Cys, Tyr and Trp. The other nine residues were present in identical proportions to those of HEW-lysozyme. The decrease in the arginine and aspartic acid residues was found to be compensated by the increase in the number of lysine, histidine and glutamic acid residues. The overall ratio of the acidic to basic amino acids has thus been conserved in the mammalian enzyme. In addition, RKN-lysozyme contained decreased numbers of Trp, Tyr and 1/2 Cys, and increased numbers of proline residues as found in HEW-lysozyme. RKN-lysozyme did not cross react with heterologous antibodies produced against HEW-lysozyme, and vice versa. RKN-lysozyme showed distinct specificity towards the lysis of M. luteus. Against this substrate, it was three times more efficient than HEW-lysozyme. It also cleaved E. coli B, but its efficiency was half as much as with M. luteus. However, it cleaved P. septica and B. subtilis at a rate similar to HEW-lysozyme under identical conditions.  相似文献
