This article is the second of two papers that review the field of spatially sensitive social scientific research into the links between social status and transport disadvantage. The first paper undertook a comprehensive review of the social scientific and transport planning literature to mark the level of development in the field and identify conceptual and methodological issues and constraints in this field of inquiry. The present article supports the advancement of socially and geographically sensitive transport research by opportunities for the development of more sophisticated spatial analytical methodologies. The approach we present is able to account for factors not previously addressed in either social or transport planning research, in particular the temporal dimensions of transport service accessibility. The article articulates the methodology through an empirical case study of socio-spatial transport disadvantage within the Gold Coast City. The article demonstrates that there are important theoretical and practical lessons to be gained for researchers and policy makers in addressing the social dimensions of transport and infrastructure provision. Further, the article argues that an attentiveness to new ways of combining and representing social and transport data-sets can promote policy relevant empirical social inquiry. The article also contributes in a productive way to the empirical knowledge of Australia's sixth-largest metropolitan area, which is often overlooked by urban scholars.  相似文献

OBJECTIVES: Transitional cell carcinomas of upper urinary tract (uttTCC) constitute 5% to 6% of all urothelial tumors. Ureteropyeloscopy has become the standard for clinical evaluation of uutTCC. Moreover, endoscopic treatments have been advocated as a conservative approach for low grade tumors or patients with intermediate grade tumors whose renal function is compromised. Therefore, grading has become the most predictive variable in defining therapeutic approach. In addition to morphologic evaluation, a series of biologic markers may be used to increase the accuracy of grading such as DNA analysis and p53 protein expression. In this study, we have evaluated these markers by means of cell image analysis with the SAMBA 400 system. METHODS: Thirteen cases of uttTCC were studied with cytologic smear, cell block, and histologic confirmation. DNA analysis was performed on cytologic smear. Immunostaining was performed on cell blocks. A grade was assigned on the basis of DNA evaluation and p53 expression quantitation. These grades were combined for each case and compared with the initial cytologic grading and the final histologic grading. RESULTS: Cytology alone diagnosed TCC in all but 1 case that was diagnosed atypical. Discrepancies were found in primary grading: cytologic grading concurred with histologic grading in 6 of the 13 cases. CONCLUSIONS: These results, although in a limited but selected number of cases, show the potential of computerized evaluation of biologic markers as parameters for a more objective grading of tumors.  相似文献

Recombinant porcine myoglobin has been produced in Escherichiacoli using the cII fusion expression system of Nagai and Th?gersen[Nature, 309, 810–812 (1984)]. After processing and reconstitutionwith haem, the protein is gel-electrophoretically and spectrophotometricallyindistinguishable from native pig myoglobin. Large crystalsof both native and recombinant porcine myoglobin were grownfrom 50 mM sodium phosphate, pH 7.1, 80% ammonium sulphate.The crystals belong to space group C2 (a = 156.9 ?, b = 42.0?, c = 92.2 ?, ß = 127.9?) and diffract to a nominal2.5 ? resolution. We plan to explore apomyoglobin as a bindingsurface in studies combining site-directed mutagenesis and X-rayanalysis. These experiments will be extended by studying thebinding of haem analogues to the mutant apoproteins.  相似文献

The aim of this study was to investigate the treatment efficiency of passive vertical‐flow wetland filters containing Phragmites australis and/or Typha latifolia and granular media of different adsorption capacities. A cost–benefit analysis was performed on the characteristics of constructed wetlands. Cheap gravel and sand as well as expensive granular activated carbon, charcoal and Filtralite (light expanded clay) were used as filter media. Different concentrations of lead and copper sulfate were added to polluted urban beck inflow water in order to simulate pre‐treated mine wastewater, landfill leachate or highway runoff. The relationships between growth media, microbial and plant communities as well as the reduction of lead, copper, five‐day biochemical oxygen demand (BOD) in particular, and potentially pathogenic bacteria were investigated. A breakthrough of copper within the first 9 weeks was only recorded for filters containing media with limited adsorption capacities. The breakthrough was independent of the presence of plants. However, after maturation of the biomass, which dominated the schmutzdecke and litter zone, lead and copper concentrations were reduced by 97–99% and BOD by 41–60% in all wetlands. There appears to be no additional benefit in using macrophytes and expensive adsorption media in constructed wetlands to enhance metal reduction during the set‐up period of 5 months. © 2001 Society of Chemical Industry  相似文献

Orthotopic liver transplantation is now a very well-established technique for treating patients with end-stage liver disease. Since 1967, more than 26,000 liver transplants have been performed, including 15,000 in the United States. The overall 1-year survival rate is approximately 80% and 5-year survival is 70%. Nuclear imaging plays an important role in the management of liver transplant recipients before and after liver transplantation. The evaluation of candidates potentially includes liver-spleen scan for liver volume, multiple gated acquisition scan, adenosine or stress thallium study, bone scan, and quantitative ventilation perfusion scan for hepatopulmonary syndrome. In the post-transplant phase, the deconvolution analysis (which corrects for the problem of recirculation) is a promising tool for diagnosing rejection, although its role in the transplant population has to be established. A variety of nuclear medicine techniques are helpful in the postoperative diagnosis of biliary complications. By performing a semiquantitative analysis to discriminate hepatocyte dysfunction from biliary disease and measuring hepatocyte extraction fraction by deconvolution analysis and excretion, (T1/2 values measured by the nonlinear list squares technique) have been very promising.  相似文献
