目的:研究嗅鞘细胞(OECs)的免疫原性及移植实验性免疫性脑脊髓炎(EAE)后颈淋巴系统的反应。方法:用新生ICR小鼠嗅球培养出OECs, 流式细胞术(FCM)检测其表面主要组织相容性抗原I、II类(MHC-I、MHC-II)表达情况。将经荧光染料CFSE标记OECs悬液或PBS注入EAE大鼠或正常大鼠的侧脑室,收集移植7天后颈部淋细胞,FCM检测CFSE+、抗原提呈细胞标志物CD83+及CFSE+CD83+的表达率,混合淋巴细胞培养检测淋巴细胞增殖率(PI)。结果:OECs高表达MHC-I (78.4±4.45%),中表达MHC-II(43.2±2.98%)。移植实验显示移植OECs的EAE大鼠颈部淋巴结中CD83+(3.83±0.03%)、CFSE+(2.47±0.05%)及CD83+CFSE+(1.98±0.04%)检出率及OECs诱导的PI值(1.32±0.06)均高于对照组或正常大鼠(P<0.05)。同时,PBS注射的EAE大鼠,较正常大鼠亦有较高的CD83+检出率(3.12±0.04%)及PI值(1.20±0.03)(P<0.05)。结论: OECs高表达的MHC分子提示OECs有免疫原性,移植后局部淋巴系统可出现针对OECs的抗原引流、提呈及T细胞的活化,进一步分析提示这种免疫反应的启动依赖于EAE的炎性环境,而OECs移植则加重这一过程。 相似文献

背景:嗅鞘细胞作为一种可再生且自体取材的移植细胞,在脊髓疾病移植治疗中受到广泛关注,关于脑出血的移植治疗目前有待实验结果的进一步积累。目的:观察嗅鞘细胞移植后脑出血大鼠神经前体细胞的增殖,评估嗅鞘细胞移植治疗大鼠脑出血的疗效。设计:完全随机对照实验。单位:华中科技大学同济医学院附属同济医院神经内科。材料:实验于2002-03/2003-03在华中科技大学同济医学院临床神经研究中心完成。选择健康雄性Wistar大鼠32只随机分为2组,每组16只。嗅鞘细胞移植组在制作大鼠尾状核出血模型第3天时,取10μL嗅鞘细胞悬液向大鼠脑内匀速注射(1μL/min)。对照组注射生理盐水10μL。方法:大鼠在移植前,移植后第3,7,14,30天分别进行神经功能评分。模型制作后第1天在两组中各取1只大鼠,制备脑组织切片,神经元轴突髓鞘观察采用髓鞘固蓝染色,神经纤维观察采用神经纤维嗜银染色。各组移植后第3,7,14,30天各取1只大鼠,制作石蜡切片,观察嗅鞘细胞移植后存活、迁徙及神经前体细胞的增殖情况,并进行神经前体细胞计数。主要观察指标:①两组大鼠脑出血后神经元轴突髓鞘和神经纤维观察。②两组大鼠脑出血后第3,7,14,30天神经前体细胞计数。③两组大鼠脑出血后第3,7,14,30天时神经功能缺损评分。结果:32只大鼠均进入实验分析。①脑出血后第30天时血肿周边及血肿灶中嗅鞘细胞计数:移植组的髓鞘化数量和神经纤维数量明显多于对照组。②两组大鼠脑出血后神经前体细胞计数:脑出血后第7,14,30天,嗅鞘细胞移植组的神经前体细胞数明显多于对照组[(41.1&;#177;2.4)个/视野,(34.5&;#177;1.2)个/视野;(43.6&;#177;1.2)个/视野,(37.2&;#177;2.0)个/视野;(19.3&;#177;1.0)个/视野,(14.2&;#177;0.4)个/视野,(t=2.42-4.02,P〈0.05)]。③两组大鼠脑出血后神经功能缺损评分:嗅鞘细胞移植组在脑出血后第14,30天时明显低于第3天[(2.21&;#177;0.20)分,(1.50&;#177;0.21)分,(2.74&;#177;0.21)分,0=2.06,3.27,P〈0.05)],对照组只在脑出血后第30天时低于第3天[(1.96&;#177;0.12)分,(2.76&;#177;0.20)分,(t=2.47,P〈0.05)]。结论:①嗅鞘细胞有增加内源性神经前体细胞数量的作用。②嗅鞘细胞可促进脑内神经细胞轴突再生,使其重新髓鞘化并建立突触联系,恢复其运动功能,进而加快损伤组织的修复。 相似文献

脑室穿刺引流联合脑脊液置换术治疗重型脑室出血的疗效评价 总被引:3,自引:0,他引:3
对90例重型脑室出血患者随机进行开颅清除血肿(A组),脑室穿刺引流(B组)和脑室穿刺引流联合脑脊液置换术治疗(C组),分析评价3种手术方法的疗效。结果:3组治疗后临床神经功能缺损程度评分较治疗前均有显著降低(P〈0.05)IB、C组的总有效率和基本痊愈率高于A组,脑脊液正常时间、住院天数较A组明显缩短,病死率低于A组(P〈0.05),而B、C组间无显著性差异(P〉0.05)IC组的基本痊愈率高于A、B组(P〈0.05)。认为脑室穿刺引流联合脑脊液置换术治疗重型脑室出血可以有效降低病死率,提高治愈率,是治疗重型脑室出血的有效方法。 相似文献

目的:探讨早期康复治疗对脑出血患者神经功能和日常生活能力的影响。方法:68例脑出血患者在发病24h内行颅内血肿微创清除术后,随机分为康复组和对照组,康复组患者在生命体征基本平稳后行早期康复治疗,对照组接受常规神经外科治疗。结果:治疗后康复组临床神经功能缺损评分、Barthel指数评分均优于对照组(P〈0.01)。康复组的基本痊愈率显著高于对照组(P〈0.05),两组的总有效率、病死率差异无显著性意义(P〉0.05)。结论:脑出血患者行颅内血肿微创清除术后,早期康复治疗可以降低患者的临床神经功能缺损程度、提高日常生活能力。 相似文献

BACKGROUND: Olfactory ensheathing cell transplantation can activate axonal regeneration and enhance myelin repair, which are beneficial for treating demyelinating diseases. OBJECTIVE: To explore the effects of olfactory ensheathing cell transplantation on myelin repair, synaptophysin expression, and motor function in a rat model of experimental allergic encephalomyelitis. DESIGN, TIME AND SETTING: A randomized, controlled experiment was performed at the Laboratory of Provincial Hospital affiliated to Shandong University between August 2006 and September 2007. MATERIALS: Dibenzylamine (Hoechst 33342), luxol fast blue, and rabbit anti-rat synaptophysin antibody were provided by Sigma, USA. METHODS: Olfactory ensheatbing cells extracted from neonatal Wistar rats were cultured for 10-14 days and labeled with dibenzylamine. Spinal cord extracted from a healthy guinea pig was homogenized and equally mixed with complete Freund's adjuvant; thereafter, the mixture was intracutaneously injected into two posterior voix pedis of healthy male Wistar rats to establish models of experimental allergic encephalomyelitis. Rats were randomly divided into a control encephalomyelitis group and an olfactory ensheathing cell transplantation group, 36 rats in each group. Physiological saline (2 μ L) or an olfactory ensheathing cell suspension (2 μ L) was separately injected along lateral cerebral ventricle at day 7 post-model induction. MAIN OUTCOME MEASURES: The migration and distribution of olfactory ensheathing cells were observed under fluorescence microscopy; myelin repair was detected using hematoxylin-eosin staining and luxol fast blue staining; synaptophysin expression was measured using immunohistochemical staining; motor function was evaluated using a motor function scale. RESULTS: Olfactory ensheatbing cells could survive in vivo and migrate to the distal end of the transplant focus and spinal cord, and survived 21 days. Hematoxylin-eosin staining and luxol fast blue staining indicated that myelin in the transplantation group was intact, and the inflammatory focus gradually disappeared. Transplantation increased synaptophysin expression (P 〈 0.05 versus control) and motor function (P 〈 0.05). CONCLUSION: Olfactory ensheathing cell transplantation can promote myelin repair, increase synaptophysin protein expression, and ameliorate motor function in a rat model of experimental allergic encephalomyelitis. 相似文献

目的探讨缺氧诱导因子-1α(hypoxia inducible factor--1α,HIF-1α)、(vascular endothelialgrouth foutor,VEGF)在脑出血灶周中的表达,并探讨其表达的意义.方法应用免疫组织化学技术检测37例脑出血灶周脑组织中HIF-1α、VEGF的表达.结果37例脑出血灶周脑组织中HIF-1α阳性率为40.6%,HIF-1α在出血量>60mL时高表达,其阳性率为88.9%,与其他出血量(30~45mL,45~600mL)阳性率的表达比较有显著意义(P<0.05).HIF-1α表达与手术时间明显相关,随着手术时间的推迟而增加,差别有显著性意义(P<0.05).HIF-1与VEGF的表达呈正相关(n=15,r=0.79)(P<0.01).结论脑出血后HIF-1α的表达与出血量及手术时间相关,为判断脑出血灶周缺血半暗带的存在提供了依据;HIF-1 α与脑出血后微循环的重建有关. 相似文献
