We have demonstrated stable operation of a 2-kW Yb:YAG phase-conjugate master oscillator power amplifier (PC-MOPA) laser system with a loop phase-conjugate mirror (LPCM). This is the first demonstration of a continuous wave (CW)-input LPCM MOPA operating at a power greater than 1 kW with a nearly diffraction-limited output beam. The single-pass beam quality incident on the LPCM varied with the specific operating conditions, but it was typically ~20 times diffraction-limited (XDL). The measured beam quality with an MOPA output power of 1.65 kW was 1.3 XDL. 相似文献

The development of enabling mass spectrometry platforms for the quantification of diverse lipid species in human urine is of paramount importance for understanding metabolic homeostasis in normal and pathophysiological conditions. Urine represents a non‐invasive biofluid that can capture distinct differences in an individual's physiological status. However, currently there is a lack of quantitative workflows to engage in high throughput lipidomic analysis. This study describes the development of a MS/MSALL shotgun lipidomic workflow and a micro liquid chromatography–high resolution tandem mass spectrometry (LC–MS/MS) workflow for urine structural and mediator lipid analysis, respectively. This workflow was deployed to understand biofluid sample handling and collection, extraction efficiency, and natural human variation over time. Utilization of 0.5 mL of urine for structural lipidomic analysis resulted in reproducible quantification of more than 600 lipid molecular species from over 20 lipid classes. Analysis of 1 mL of urine routinely quantified in excess of 55 mediator lipid metabolites comprised of octadecanoids, eicosanoids, and docosanoids generated by lipoxygenase, cyclooxygenase, and cytochrome P450 activities. In summary, the high‐throughput functional lipidomics workflow described in this study demonstrates an impressive robustness and reproducibility that can be utilized for population health and precision medicine applications. 相似文献

Spectrally resolved white-light interferometry was used to measure the wavelength dependence of refractive index (i.e., dispersion) for various ocular components. Verification of the technique's efficacy was substantiated by accurate measurement of the dispersive properties of water and fused silica, which have both been well-characterized in the past by single-wavelength measurement of the refractive index. The dispersion of bovine and rabbit aqueous and vitreous humors was measured from 400 to 1100 nm. In addition, the dispersion was measured from 400 to 700 nm for aqueous and vitreous humors extracted from goat and rhesus monkey eyes. An unsuccessful attempt was also made to use the technique for dispersion measurement of bovine cornea and lens. The principles of white-light interferometry, including image analysis, measurement accuracy, and limitations of the technique, are discussed. The exotic predatory cladoceran Bythotrephes longimanus was first observed in Lake Erie on 19 September, 1985. During the early summer immediately prior to its appearance, the cladoceran community in the central basin of Lake Erie was characterized by large populations of Bosmina longirostris, Eubosmina coregoni, Daphnia mendotae, and Daphnia retrocurva, with the latter three species persisting throughout August and September, along with Diaphanosoma spp. Community composition during early summer 1986 was similar to that of the previous year, but densities of all cladocerans decreased dramatically coincident with the appearance of Bythotrephes in mid-July, and remained suppressed throughout August and September. Only D. mendotae was present in appreciable numbers during this period; from mid-July through the end of August, 86-98% of cladoceran biomass (exclusive of Bythotrephes and Leptodora) was contributed by D. mendotae. Densities of Leptodora and Bythotrephes showed a strikingly inverse relationship, both temporally and spatially, during 1986. Size frequency distributions of D. mendotae exhibited an immediate shift towards extremely large ( > 2.5 mm) individuals coincident with Bythotrephes’ appearance in 1986, suggesting an upper size limit to efficient prey utilization by Bythotrephes. These results suggest that Bythotrephes can have substantial impacts both on cladoceran community composition, and on the size distributions of individual species. 相似文献

Hypoxic regions in solid neoplasms have been associated with tumor recurrence and resistance to several cancer treatment modalities including radiation therapy. Various strategies have been designed to target these resistant cells, including the use of the bioreductive alkylating agent mitomycin C (MC), which exerts preferential cytotoxicity under hypoxic conditions in most cell lines. Analyses of the mechanism of action of MC indicate that this drug can form cross-links with DNA; it is currently thought that this bisadduct is the critical lesion responsible for inhibiting DNA synthesis. Computer-generated models suggest that the MC adduct fits snugly into the minor groove of B-DNA without imposing major distortion on the structure of the DNA molecule. To gain additional insight into the role of cross-linkage in the cytotoxicity of MC, we studied the analogue, decarbamoyl mitomycin C (DMC). The structure of DMC is identical to that of MC with the exception of the substitution of the carbamoyl group at the C-10 position by a nonalkylating hydroxyl group (-OH); this alteration would be expected to prevent DMC from forming bisadducts with DNA. In chemical systems, DMC produces only DNA monoadducts. If indeed it is the MC-DNA cross-links which are responsible for cell kill, one would predict DMC to be less cytotoxic than MC. However, tissue culture studies using DMC revealed that DMC is at least as toxic as MC to EMT6 mouse mammary tumor cells and to wild-type AA8 Chinese hamster ovary (CHO) cell lines.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

Oxalate has been implicated in the etiology of nephrocalcinosis in premature infants as well as in the formation of insoluble precipitates in total parenteral nutrition (TPN) intravenous tubing. Oxidation of ascorbate to oxalate, especially in the presence of catalysts such as copper and iron, has been implicated in formation of these precipitates. The purpose of this project was to measure oxalate formation in certain TPN components separately and in combination. Neonatal TPN solution components in combination were infused at 5 mL/h under simulated clinical conditions used in a neonatal intensive care unit. Aliquots were assayed at intervals for oxalate by capillary electrophoresis. Oxalate is present in one TPN mixture at concentrations up to 8 ppm. The addition of ascorbate to an aqueous solution of trace metals may promote oxalogenesis. 相似文献
