The interaction of carnitine with human placental brush-border membrane vesicles was investigated. Carnitine was found to associate with the membrane vesicles in a Na(+)-dependent manner. The time course of this association did not exhibit an overshoot, which is typical of a Na+ gradient-driven transport process. The absolute requirement for Na+ was noticeable whether the association of carnitine with the vesicles was measured with a short time incubation or under equilibrium conditions, indicating Na(+)-dependent binding of carnitine to the human placental brush-border membranes. The binding was saturable and was of a high-affinity type with a dissociation constant of 1.37 +/- 0.03 microM. Anions had little or no influence on the binding process. The binding process was specific for carnitine and its acyl derivatives. Betaine also competed for the binding process, but other structurally related compounds did not. Kinetic analyses revealed that Na+ increased the affinity of the binding process for carnitine and the Na+/carnitine coupling ratio for the binding process was 1. The dissociation constant for the interaction of Na+ with the binding of carnitine was 24 +/- 4 mM. This constitutes the first report on the identification of Na(+)-dependent high-affinity carnitine binding in the plasma membrane of a mammalian cell. Studies with purified rat renal brush-border membrane vesicles demonstrated the presence of Na+ gradient-driven carnitine transport but no Na(+)-dependent carnitine binding in these membrane vesicles. In contrast, purified intestinal brush-border membrane vesicles posses neither Na+ gradient-driven carnitine transport nor Na(+)-dependent carnitine binding.  相似文献

Polarization Mode Dispersion (PMD) compensation has been a matter of investigation of several papers in literature. The proposed solutions belong basically to two large families: electronic compensators and optical compensators. Both PMD compensator schemes have advantages and disadvantages: electronic PMD compensators are usually simple to include in line-terminal, potentially low-cost, very fast, and FEC compatible but their development is strongly dependent on IC technology capability that at this time allows device developments up to the 10 Gbit/s bit rates area. Furthermore, they are strongly dependent on modulation formats, and they can operate only on a single channel. Conversely, optical compensators are independent of bit rate and modulation format and potentially they can compensate more channels simultaneously; their major drawbacks are the longer response time and the complexity in the feedback signal process within the control algorithm. In this paper we consider an optical Polarization Mode Dispersion Compensator (PMDC) that is simple to realize and easy to include at limited costs in each EDFA module (distributed compensation) as well as a single-stage front-end compensator. Numerical analysis of the PMDC and experimental results confirm the utility of the PMDC proposed and its capability in compensating DGD larger than 20 ps in NRZ and RZ 40 Gbit/s optical systems.  相似文献

A new analytical method, using pressurised liquid extraction (PLE) and liquid chromatography–tandem mass spectrometry (LC–MS/MS), was developed for the simultaneous determination of bisphenol A (BPA), octylphenol (OP) and nonylphenol (NP) in powdered infant formulas (IF) and powdered skimmed milk (PM). The analytes were extracted by PLE, using this optimised conditions: ethyl acetate as solvent, 70 °C of temperature, reversed-phase silica C18 as dispersing agent and three cycles of extraction. The extracts were then injected in LC–MS/MS using a Gemini C18 column and a mixture of 5% water and 95% methanol/acetonitrile, both with 0.1% ammonia, as a mobile phase. Recoveries at different fortification levels (0.5 and 0.05 mg kg−1), were between 89% and 92% for BPA, 84 and 98% for OP, and 93% and 101% for NP. The method was applied to the analysis of samples of PM and IF, bought in Italian and Spanish markets. In positive samples, phenols concentration ranged from 0.07 to 1.29 mg kg−1 for BPA, from 0.028 to 1.55 mg kg−1 for OP and from 0.026 to 1.47 mg kg−1 for NP.  相似文献

A percentage of hemodialysis (HD) patients are resistant to recombinant human erythropoietin (rHuEPO), a phenomenon that occurs less frequently in patients dialyzed with biocompatible membranes (M) and in peritoneal dialysis. The authors evaluated the effects of paired filtration dialysis (PFD)--a dialysis technique based on the use of an emophan M in conjunction with a polysulphone M--on erythropoiesis in HD patients resistant to rHuEPO. Twelve HD patients with anemia resistant to long-term therapy with rHuEPO (200.24 U/kg body weight three times per week intravenously for 10.2 months) were studied. Patients had been treated for an average of 46.9 months with bicarbonate HD, using cuprophan M (Phase A) and, successively, for 12 months by PFD (Phase B). The following parameters were evaluated monthly: 1) hemoglobin and hematocrit values; 2) serum levels of erythropoietin (EPO); and 3) serum levels of interleukin (IL)-3, IL-6, IL-10, IL-1 beta, tumor necrosis factor alpha (TNF-alpha), and interferon gamma (IFN-gamma). At the end of Phase A and Phase B, patients underwent bone marrow biopsies to evaluate 1) bone marrow burst forming unit erythroid (BFU-E) and colony forming unit erythroid (CFU-E) proliferative capacity, and 2) bone marrow mononuclear cell EPO-receptor (EPO-R) number. During Phase B, there was a progressive rise in hematocrit and hemoglobin values, so that within the sixth month, the rHuEPO dose was reduced to 80 +/- 15 U/kg body weight three times per week. At the same time, an increase in serum IL-3, IL-6, and IL-10 levels was seen, whereas serum IL-1 beta, TNF-alpha, and IFN-gamma levels decreased. This was accompanied by a rise in BFU-E and CFU-E growth and in bone marrow mononuclear cell EPO-R number. During Phase B, after the dialysis session, serum EPO levels increased by about 30% in comparison with pre dialysis values, whereas during Phase A they decreased by about 14%. In HD patients, EPO resistance may caused either by absorption of rHuEPO to the cuprophan M, or an increased release of cytokines that inhibit erythropoiesis, such as IL-1 beta, TNF-alpha, and IFN-gamma, and to a decrease in stimulatory cytokines such as IL-3, IL-6, and IL-10. These negative phenomena are reversed by the use of biocompatible dialysis techniques such as PFD.  相似文献

Engineering Properties of Sand-Fiber Mixtures for Road Construction   总被引:4,自引:0,他引:4  
The purpose of this investigation was to identify and quantify the effect of numerous variables on the performance of fiber-stabilized sand specimens. Laboratory unconfined compression tests were conducted on sand specimens reinforced with randomly oriented discrete fibers to isolate the effect of each variable on the performance of the fiber-reinforced material. Five primary conclusions were obtained from this investigation. First, the inclusion of randomly oriented discrete fibers significantly improved the unconfined compressive strength of sands. Second, an optimum fiber length of 51 mm (2 in.) was identified for the reinforcement of sand specimens. Third, a maximum performance was achieved at a fiber dosage rate between 0.6 and 1.0% dry weight. Fourth, specimen performance was enhanced in both wet and dry of optimum conditions. Finally, the inclusion of up to 8% of silt does not affect the performance of the fiber reinforcement.  相似文献

Large eddy simulations (LES) of the flow past a wind turbine with and without tower and nacelle have been performed at 2 tip speed ratios (TSR, ), λ=3 and 6, where the latter corresponds to design conditions. The turbine model is placed in a virtual wind tunnel to reproduce the “Blind test 1” experiment performed at the Norwegian University of Science and Technology (NTNU) closed‐loop wind tunnel. The wind turbine was modeled using the actuator line model for the rotor blades and the immersed boundary method for the tower and nacelle. The aim of the paper is to highlight the impact of tower and nacelle on the turbine wake. Therefore, a second set of simulations with the rotating blades only (neglecting the tower and nacelle) has been performed as reference. Present results are compared with the experimental measurements made at NTNU and numerical simulations available in the literature. The tower and nacelle not only produce a velocity deficit in the wake but they also affect the turbulent kinetic energy and the fluxes. The wake of the tower interacts with that generated by the turbine blades promoting the breakdown of the tip vortex and increasing the mean kinetic energy flux into the wake. When tower and nacelle are modeled in the numerical simulations, results improve significantly both in the near wake and in the far wake.  相似文献

The survival of patients with glioblastoma (GBM) is poor. The main cause is the presence of glioma stem cells (GSCs), exceptionally resistant to temozolomide (TMZ) treatment. This last may be related to the heterogeneous expression of ion channels, among them TRPML2. Its mRNA expression was evaluated in two different neural stem cell (NS/PC) lines and sixteen GBM stem-like cells by qRT-PCR. The response to TMZ was evaluated in undifferentiated or differentiated GSCs, and in TRPML2-induced or silenced GSCs. The relationship between TRPML2 expression and responsiveness to TMZ treatment was evaluated by MTT assay showing that increased TRPML2 mRNA levels are associated with resistance to TMZ. This research was deepened by qRT-PCR and western blot analysis. PI3K/AKT and JAK/STAT pathways as well as ABC and SLC drug transporters were involved. Finally, the relationship between TRPML2 expression and overall survival (OS) and progression-free survival (PFS) in patient-derived GSCs was evaluated by Kaplan–Meier analysis. The expression of TRPML2 mRNA correlates with worse OS and PFS in GBM patients. Thus, the expression of TRPML2 in GSCs influences the responsiveness to TMZ in vitro and affects OS and PFS in GBM patients.  相似文献

The present study was undertaken to investigate whether less pathogenic Candida species (C. tropicalis, C. stellatoidea, C. krusei and C. glabrata) express a fibronectin receptor (FNr) antigenically related to alpha 5 beta 1 integrin, which mediates their binding to fibronectin (FN). By flow cytometric analysis, a monoclonal antibody (MAb) directed against human alpha 5 integrin subunit (clone SAM-1) and two different antisera to FNr positively stained C. tropicalis, C. stellatoidea and C. glabrata, with the greatest expression observed for C. tropicalis. No or only marginal immunoreactivity was found on C. krusei. C. tropicalis, C. stellatoidea, C. glabrata, but not C. krusei yeasts specifically adhered to FN; higher levels of adhesion were found for C. tropicalis and C. stellatoidea with respect to C. glabrata. Less pathogenic Candida spp. bound to the Arg-Gly-Asp (RGD) containing 120-kDa fragment of FN and adhesion to intact FN was markedly inhibited by Gly-Arg-Gly-Asp-Ser-Pro (GRGDSP), but not by Gly-Arg-Gly-Glu-Ser-Pro (GRGESP) peptides. In addition, anti-alpha 5 SAM-1 MAb and both anti-FNr antisera strongly blocked binding of less pathogenic Candida spp. to FN. Overall, these results indicate that less pathogenic Candida spp., including C. tropicalis, C. stellatoidea and C. glabrata, express a receptor antigenically related to alpha 5 beta 1 integrin which mediates their adhesion to FN.  相似文献
