Clasmatodendrosis is one of the irreversible astroglial degeneration, which is involved in seizure duration and its progression in the epileptic hippocampus. Although sustained heat shock protein 25 (HSP25) induction leads to this autophagic astroglial death, dysregulation of mitochondrial dynamics (aberrant mitochondrial elongation) is also involved in the pathogenesis in clasmatodendrosis. However, the underlying molecular mechanisms of accumulation of elongated mitochondria in clasmatodendritic astrocytes are elusive. In the present study, we found that clasmatodendritic astrocytes showed up-regulations of HSP25 expression, AKT serine (S) 473 and dynamin-related protein 1 (DRP1) S637 phosphorylations in the hippocampus of chronic epilepsy rats. 2-Cyano-3,12-dioxo-oleana-1,9(11)-dien-28-oic acid methyl ester (CDDO-Me; bardoxolone methyl or RTA 402) abrogated abnormal mitochondrial elongation by reducing HSP25 upregulation, AKT S473- and DRP1 S637 phosphorylations. Furthermore, HSP25 siRNA and 3-chloroacetyl-indole (3CAI, an AKT inhibitor) abolished AKT-DRP1-mediated mitochondrial elongation and attenuated clasmatodendrosis in CA1 astrocytes. These findings indicate that HSP25-AKT-mediated DRP1 S637 hyper-phosphorylation may lead to aberrant mitochondrial elongation, which may result in autophagic astroglial degeneration. Therefore, our findings suggest that the dysregulation of HSP25-AKT-DRP1-mediated mitochondrial dynamics may play an important role in clasmatodendrosis, which would have implications for the development of novel therapies against various neurological diseases related to astroglial degeneration.  相似文献

The high‐temperature sous‐vide (HTSV) method was developed to prepare carrots with a soft texture at the appropriate degree of pasteurization. The effect of heating conditions, such as temperature and time, was investigated on various package sizes. Heating temperatures of 70, 80, and 90 °C and heating times of 10 and 20 min were used to evaluate the HTSV method. A 3‐dimensional conduction model and numerical simulations were used to estimate the temperature distribution and the rate of heat transfer to samples with various geometries. Four different‐sized packages were prepared by stacking carrot sticks of identical size (9.6 × 9.6 × 90 mm) in a row. The sizes of the packages used were as follows: (1) 9.6 × 86.4 × 90, (2) 19.2 × 163.2 × 90, (3) 28.8 × 86.4 × 90, and (4) 38.4 × 86.4 × 90 mm. Although only a moderate change in color (L*, a*, and b*) was observed following HTSV cooking, there was a significant decrease in carrot hardness. The geometry of the package and the heating conditions significantly influenced the degree of pasteurization and the final texture of the carrots. Numerical simulations successfully described the effect of geometry on samples at different heating conditions.  相似文献

Doped carbon nanostructures as non-precious metal (NPM) catalysts for oxygen reduction reaction (ORR) in acid medium are mainly synthesized using 5, 10, 15, 20-tetrakis (4-methoxyphenyl)-porphyrin-Fe (III) chloride (Fe-TMPP) as doping and carbon sources. In this study, the doped carbon nanostructures used as cathode NPM catalysts for ORR are prepared using a mixture of iron phthalocyanine (FePc) and arginine as doping and carbon sources. The morphology and composition of the as-prepared samples are characterized using field-emission scanning electron microscopy, field-emission transmission electron microscopy, and energy dispersive X-ray (EDX) spectroscopy. The crystal and pore structures are analyzed using X-ray diffraction method, Raman spectroscopy, and nitrogen adsorption/desorption method. The sample prepared using a precursor mixture with a proper ratio of FePc and arginine exhibits significantly superior ORR performance, i.e. high specific activity, enhanced half-wave potential, and improved stability in an acid medium, as even compared to a commercial Pt/C. The improved ORR properties is mainly attributed to high portion of pyridinic N state with a relatively high specific surface area, which can result from the FePc precursor surrounded by the fused arginine.  相似文献

The concentration of nutrients in brown rice is mainly associated with embryo size. Various beneficial components have been purified from rice bran. Recently developed black waxy rice with a giant embryo (‘Milyang 263’, BGE), which is the ge t mutant of the GE gene, was selected and analyzed to produce high quality nutritional components. γ-Aminobutyric acid (GABA) content in BGE rice bran, one of the most important nutritional compounds in rice, showed a 10.6-fold increase (2.66±0.48 mg/g) compared to that of BGE brown rice (0.25±0.01 mg/g). In addition, brown rice and BGE rice bran showed enriched amylopectin (94.5±0.5 and 97.0±0.0%) and bioactive anthocyanin [Cy-3-G: 75.15±4.18 (brown rice), 82.97±0.81 (rice bran) and Pn-3-G: 1.52±0.50 (brown rice), 4.33±0.20 (rice bran); mg/100 g] contents. These results suggest that BGE rice bran can be used as an excellent raw material to efficiently produce high quality essential amino acids, cyanidin-3 glycoside-enriched anthocyanins, and GABA.  相似文献

Wire-woven bulk Kagome (WBK) is a new truss-type cellular metal fabricated by assembling helically-formed wires. In this work, the main design parameters governing the compressive strength of WBK were selected from various geometric and material factors. The Taguchi method was used to minimize the number of experiments required to evaluate the sensitivity of each design parameter with the specific compressive strength of WBK. As numerical experiments, finite element analyses were performed to investigate the mechanical responses of WBK with various combinations of levels for selected design parameters. We found that the slenderness ratio is the dominant and most influential design parameter characterizing the specific strength of WBK, followed by the strain-hardening exponent and the yield strength of the brazed filler metal. The height of the braze normalized by the wire diameter is the least influential design parameter. In addition, an optimized design for the highest specific strength of WBK was derived.  相似文献

Direct modulation of large-conductance Ca2+-activated K+ (maxi-K) channel by receptor-associated G protein in rabbit mesenteric arterial smooth muscle cells was studied using the outside-out patch clamp technique. Applying a beta-adrenoceptor agonist (isoproterenol) increased maxi-K channel activity by 75%, and the effect was almost completely abolished by pretreating the cells with pertussis toxin but not with cholera toxin. When the antibody against Gi protein was present in the pipette solution the stimulatory effect of isoproterenol disappeared. These results suggest that beta-adrenoceptor stimulation increases maxi-K channel activity via a membrane-delimited pathway, probably through pertussis toxin-sensitive G protein (Gi).  相似文献

Tuned Liquid damper and Tuned Liquid Column are kind of passive mechanical damper which relies on the sloshing of liquid in a rigid tank for suppressing structural vibrations. TLD and TLCD are attributable to several potential advantages —low costs; easy to install in existing structures; effective even for small-amplitude vibrations. In this paper, the shaking table experiments were conducted to investigate the characteristics of water sloshing motion in TLD (rectangular, circular) and TLCD. The parameter obtained from the experiments were wave height, base shear force and energy dissipation. The shaking table experiments show that the liquid sloshing relies on amplitude of shaking table and frequency of tank. The TLCD was more effective control vibration than TLD.  相似文献

We first investigated the effect of partial pressure of hydrogen (H2) on the performance of polymer electrolyte fuel cells (PEFCs) by controlling the ratio of hydrogen and nitrogen (N2). The cell performance with Pt/C anode was significantly decreased with reduction of the partial pressure of H2 in the presence of carbon monoxide (CO), while the performance variation was negligible in the absence of CO. Severe CO poisoning on Pt/C electrode at low partial pressure of H2 might be attributed to the hindering effect by N2 and CO. On the other hand, PtRu/C anode showed consistent power performance even at low partial pressure of H2.  相似文献
