Human leukemic cell line K562 is induced to differentiate into the megakaryocytic lineage by stimulation with 12-O-tetradecanoylphorbol-13-acetate (TPA). We demonstrate here that TPA stimulation increases tyrosine phosphorylation of an 80-kDa protein at an early stage of megakaryocytic differentiation and that this 80-kDa protein is identical with cortactin. Since tyrosine kinase Syk was activated by TPA stimulation, we examined the possibility that cortactin is a potential substrate of Syk in K562 cells. TPA-induced tyrosine phosphorylation of cortactin was decreased profoundly by overexpression of dominant-negative Syk. Furthermore, cortactin was associated with Syk even before TPA stimulation. Since cortactin was previously referred as an 80/85-kilodalton pp60src substrate, we examined the association between Src and cortactin, whereas its association could not be detected. These data suggest that Syk phosphorylates cortactin in K562 cells upon TPA treatment. 相似文献

Platelet-activating factor (PAF) is a potent inflammatory mediator which is released by various inflammatory cells and produced
by certain tissues, including the kidney. PAF has been shown to increase glomerular permeability to protein and to decrease
glomerular filtration rate (GFR) by contracting mesangium. On the basis of these observations, it has been suspected that
PAF may play a role as mediator of glomerular damage in glomerular nephritis. To examine this possibility, we studied the
effects of a specific PAF antagonist, R-75,317, on the development of an experimental model of anti-glomerular basement membrane
(anti-GBM) glomerulonephritis. Glomerulonephritis was initiated by injecting rabbit anti-rat GBM serum into rats. Proteinuria
gradually developed after serum injection, plateaued at week 2, and remained at the high level of week 2 throughout the experimental
period (6 wk). Chronic treatment with R-75,317 (10 mg/kg/day i.p.) tended to delay the onset of proteinuria and significantly
accelerated the recovery phase. Creatinine clearance (Ccr) fell to 40% at week 3. R-75,317 treatment completely prevented
this decline of Ccr. Histological changes in this model (glomerular hypertrophy, proliferation of mesangial matrix and interstitial
fibrosis) were also ameliorated by the R-75,317 treatment. The results suggest that PAF may play a role in the development
of glomerulonephritis and that PAF antagonists could be used in the treatment of human renal disease.

Acicular goethite particles were synthesized by oxidation of an alkaline suspension of ferrous hydroxide using a bubble column with draft tubes of different diameters at a constant temperature of 40°C. The oxidation rate increased with decreasing draft tube diameter. The mean size based on the major axis of acicular particle decreased with increasing oxidation rate and decreasing draft tube diameter. The major axis was found to grow preferentially compared with the minor axis. The mean size of major axis was correlated well to the modified reaction rate defined as a product of the reaction rate per unit volume of the reactor multiplied by the ratio of the column diameter to the draft tube diameter, irrespective of the draft tube diameter. 相似文献

The synthesis of magnetite nanoparticles coated with a well-defined graft polymer is reported. The magnetite nanoparticles with an initiator group for copper-mediated atom transfer radical polymerization (ATRP), 2-(4-chlorosulfonylphenyl) ethyltrichlorosilane (CTCS) chemically bound on their surfaces were prepared by the self-assembled monolayer-deposition method. The surface-initiated ATRP of methyl methacrylate (MMA) was carried out with the CTCS-coated magnetite nanoparticles in the presence of free (sacrificing) initiator, p-toluenesulfonyl chloride. Polymerization proceeded in a living fashion, exhibiting first-order kinetics of monomer consumption and a proportional relationship between molecular weight of the graft polymer and monomer conversion, thus providing well-defined, low-polydispersity graft polymers with an approximate graft density of 0.7 chains/nm2. The molecular weight and polydispersity of the graft polymer were nearly equal to those of the free polymer produced in the solution, meaning that the free polymer is a good measure of the characteristics of the graft polymer. The graft polymer possessed exceptionally high stability and remarkably improved dispersibility of the magnetite nanoparticles in organic solvent. 相似文献

Kraft lignin was pyrolyzed in molten salt media of ZnCl2-KCl mixture with tetralin vapor added in 0.4 and 4 mol% diluted with N2 in the temperature range of 400 to 700°C. The gas chromatographic analyses of pyrolysis products revealed that the yield of H2 was not increased by tetralin vapor addition. This fact implies that the hydrogen radical produced from tetralin is consumed in the formation of phenolic compounds and light liquids but not in the formation of H2. p-Cresol was the most abundant phenolic compound. The yield of total phenolic compounds with 4 mol% tetralin vapor added was increased by ca. 80% as compared to that in neat pyrolysis of Kraft lignin. 相似文献

Round bars of 13.6 mm diameter made from low alloy steel were sprayed by hard-coating Lubrotec 19985 under the following spraying
parameters: the rotation speed varied from 500 to 900 rpm, the feeding speed from 0.1 to 1.0 mm/rev, and the nozzle tip distance
from 100 to 200 mm; the sprayed bars were then subjected to bending test. Depending on the thickness of the produced coating,
the modulus of elasticity and yield bending stress of the coating were calculated by applying the bending deflection formula
and an equivalent cross-sectional area technique, respectively. The results obtained indicated that the thickness, hardness,
yield bending stress, and modulus of the elasticity of coating were varied with the variation in spraying parameters. The
optimum spraying parameter was a rotation speed of 700 r.p.m, a nozzle tip distance of 200 mm, and a feed rate of 0.1 mm/rev.
This optimum parameter produced an acceptable thickness of 1.14 mm, a hardness of 148 HV, a bending stress of 404.9 MPa, and
a modulus of elasticity of 91.47 GPa. The results obtained can also help determine the modulus of elasticity and yield bending
stress of the various coating materials when a coated round bar is subjected to the bending test.

From 1999 to 2008, he was a Professor at the Department of Mechanical Engineering at Al-Balqa’ Applied University, and at
Tafila Technical University, Jordan. Currently, he is a researcher in Canada.
Mohammed Al-Nawafleh received his B.S. and M.S degrees in Mechanical Engineering from Leningrad Ins-titute of Textile and Light Industry in 1989.
He then received his Ph.D. degree from Sankt-Petersburg State University of Technology and Design, Russia in 1993. From 1997
to 2005, Dr. Nawafleh was a Professor at the Department of Mechanical Engineering at Al-Balqa’ Applied University in Jordan.
From 2005 to 2008, he was a Professor at the Department of Mechanical Engineering at Tafila Technical University. Currently,
