The targeting of karyophilic proteins to nuclear pores is mediated via the formation of a nuclear pore-targeting complex, through the interaction of nuclear localization signal (NLS) with its NLS receptor. Recently, a novel human protein, Qip1, was identified from a yeast two-hybrid system with DNA helicase Q1. This study demonstrates that Qip1 is a novel third class of NLS receptor that efficiently recognizes the NLS of the helicase Q1. Moreover, the data obtained in this study show that the specific interaction between Qip1 and the NLS of the helicase Q1 requires its upstream sequence of the minimal essential NLS. By using purified recombinant proteins alone in the digitonin-permeabilized cell-free transport system, it was demonstrated that the two known human NLS receptors, Rch1 and NPI-1, are able to transport all the tested NLS substrates into the nucleus, while Qip1 most efficiently transports the helicase Q1-NLS substrates, which contain its upstream sequence in so far as we have examined the system. Furthermore, in HeLa cell crude cytosol, it was found that endogenous Rch1 binds to all the tested NLS substrates, while the binding of endogenous NPI-1 is restricted to only some NLSs, despite the fact that NPI-1 itself shows binding activity to a variety of NLSs. These results indicate that at least three structurally and functionally distinct NLS receptors exist in the human single cell population, and suggest that the nuclear import of karyophilic proteins may be controlled in a complex manner at the NLS recognition step by the existence of a variety of NLS receptors with various specificities to each NLS.  相似文献

Photonic networks based on wavelength division multiplexing (WDM) and optical path technologies are expected to realize flexible, transparent, and cost-effective transport networks with a large transmission capacity. This paper explores the design framework of photonic transport networks taking into consideration the operation administration and maintenance (OA&M) functions required for the successful introduction of WDM systems based on the optical path concept. From the view point of network maintenance, clear distinction is made between the optical path layer and the optical section layer to facilitate accurate and smooth failure localization. The digital multiplexing span between physical multiplexing interfaces at the end-to-end digital nodes should have the same maintenance span as the corresponding optical path. We argue that cooperative maintenance by OA&M functions at both the digital and optical layers can be a practical way of network supervision. A supervisory (SV) signal transfer method and a configuration that is suitable for the terrestrial trunk network are also indicated. As an example, a practical SV system design methodology and an actual procedure developed for a single channel optical transmission system based on optical in-line amplifiers are introduced. Furthermore, application of the developed SV system and network restoration schemes is discussed for future WDM-based photonic networks. We report a case of acute cholangitis caused by the inadvertent ligation of the right hepatic duct during a cholecystectomy performed 11 years before. When the condition of the right hepatic duct or accessory bile duct ligation persists for more than several years, resulting in atrophy of the relevant part of the liver, bile duct ligation will rarely cause severe clinical problems. However, acute cholangitis may occur, as in our patient, so long-term follow-up of the patient is warranted.  相似文献

This is the first report of glycoside hydrolase family 43 β-xylosidase from Aspergillus oryzae. To characterize this enzyme, the recombinant enzyme was expressed in Escherichia coli. Unlike known β-xylosidases from fungal origins, the enzyme did not show substrate ambiguity and was stable at alkaline pH.  相似文献

The functions of Val483, Trp534, and Met729 in Euphorbia tirucalli β-amyrin synthase were revealed by comparing the enzyme activities of site-directed mutants against that of the wild type. The Gly and Ala variants with a smaller bulk size at position 483 predominantly afforded monocyclic camelliol C, which suggested that the orientation of the (3S)-2,3-oxidosqualene substrate was not appropriately arranged in the reaction cavity as a result of the decreased bulk size, leading to failure of its normal folding into the chair–chair–chair–boat–boat conformation. The Ile variant, with a somewhat larger bulk, afforded β-amyrin as the dominant product. Intriguingly, various variants of Trp534 exhibited significantly decreased enzymatic activities and provided no aberrantly cyclized products, although the aromatic Phe and Tyr residues were incorporated and the steric sizes of the aliphatic residues were altered. Therefore, the Trp534 residue does not stabilize the transient cation through a cation–π interaction. Furthermore, the Trp residue, with the largest steric bulk among all natural amino acids, is essential for high enzymatic activity. Robust CH–π complexation between the Val483 and Trp534 residues is proposed herein. Altering the steric bulk at the Met729 position afforded the pentacyclic skeletons. Thus, Met729 is positioned at the E-ring formation site. More detailed insights into the functions of the Val483, Trp534, and Met729 residues are provided by homology modeling.  相似文献

The relationship between thermal conductivity and unidirectional cellular mullite microstructures created with the freeze casting route is discussed using actual three‐dimensional (3D) images, in which a homogenization method was conducted with microscopic models created from 3D images, the temperature variation in macroscopic models by the finite element method (FEM), and local heat conduction in microscopic models. The simulation results were consistent with the experimental results. This image‐based modeling technique was thus confirmed to be a strong tool for the prediction of various thermal properties of porous ceramics.  相似文献

This paper proposes a new estimation method to determine arbitrary circuit parameters for the harmonic analysis of a distribution network. Harmonic analysis is important in maintaining electric power quality. An interesting phenomenon, a temporary cut‐down of the harmonic voltage level at dawn, even if demand is slightly increasing at this time, is often observed in actual power systems. This paper proposes to utilize observations of this phenomenon to estimate circuit parameters. In addition, it is claimed that a detailed equivalent circuit is preferable to a conventional simplified equivalent circuit for explaining the phenomenon. This paper also discusses the usefulness of the Harmonic Characteristics Curve, which is proposed in this paper as a new concept, at the secondary winding of the distribution transformer, and the correlation of PQ, the monitored values of the active and reactive power at the secondary winding of distribution transformer. This correlation should determine the equivalent fundamental frequency admittance of the shunt capacitors installed for distribution lines. © 2014 Wiley Periodicals, Inc. The phonon thermal conductivity of misfit-layered Ca3Co4O9 has been calculated by perturbed molecular dynamics using a classical force field. Detailed numerical analyses reveal that, in spite of its smaller cross-sectional area, the CoO2 layer transports more heat than the thicker rock salt (RS) layer, although its local thermal conduction is more suppressed than in another layered cobaltite, Na x CoO2. The origins of these differences have been elucidated through careful examination of the atomic arrangements in each layer. Since thermal conduction in the RS layer can be reduced without deteriorating electronic properties for which the CoO2 layer is responsible, it is suggested that the RS layer should be modified to further suppress the overall in-plane thermal conductivity. Computational experiments with increasing number of Ca–O planes in the RS layer showed the opposite trend to what can be predicted based on the misfit between two dissimilar layers. Further analyses to reveal the origin of these unexpected results provide yet another strategy to further decrease the thermal conductivity, namely to control the dynamic interference between atoms across the interface between two layers.  相似文献

The interphase transfer behavior of poly(styrene-co-α-methyl styrene), which is known as a tackifier in rubber industry, is investigated using poly(butadiene) rubber (BR) and poly(styrene-co-butadiene) rubber (SBR). The films of pure rubbers and the blends with the tackifier were prepared by a solution-cast method using toluene as a solvent. Two sheets composed of different rubbers, in which the tackifier was mixed at least in one rubber sheet, were piled together and annealed beyond the glass transition temperature T g of the tackifier. The transfer phenomenon of the tackifier between the sheets was evaluated by the peak shift in the tensile loss modulus E″ curve, ascribed to T g of the rubbers, which was measured by the dynamic mechanical analysis. Moreover, crystallization temperature of BR was also employed as a measure of the transfer, because the tackifier retards the crystallization of BR. It is found that the tackifier moves from one rubber to another during annealing procedure to reduce severe localization. When the content is the same in each rubber, the tackifier immigrates from BR to SBR, suggesting a better miscibility with SBR. This behavior is attributed to the small difference in the solubility parameter between SBR and the tackifier as compared to that between BR and the tackifier.  相似文献
