目的:从Phellinus pini子实体中提取分离到一种水溶性多糖纽分PS1,并对其免疫活性进行初步研究。方法:先进行热水提取,DEAE-纤维素离子交换树脂,sepharose CL-4B凝胶树脂分离纯化PS1,并利用GC色谱、FT-1R光谱进行组成成分分析,然后通过刚果红吞噬实验、脾脏B淋巴细胞增殖实验及免疫器官指数评价其免疫活性。结果:PS1为一种β型杂多糖,总糖含量70.36%,蛋白质含量4.69%,分子量为230kDa。糖类构成有鼠李糖、阿拉伯糖、木糖、甘露糖、半乳糖、葡萄糖,其摩尔比为2.14:2.36:0.44:4.28:5.17:85,61。多糖PS1能显著提高正常小鼠的脾指数和胸腺指数(P〈0.01),免疫力低下小鼠的肝指数、胸腺指数也有提高,但效果不明显.PS1均能提高正常和免疫低下小鼠的巨噬细胞吞噬能力。此外。PS1能显著促进脾细胞体外增殖能力(P〈0.01)。结论:多糖PS1能显著提高机体的免疫能力。可作为一种生物效应调节剂应用于医药或保健食品领域。  相似文献

AIMS: The objective of the present study was to determine the optimal culture conditions for the production of four groups of exopolysaccharides (EPSs) in Phellinus gilvus by submerged culture and to investigate their molecular properties by multi-angle laser-light scattering (MALLS) analysis. METHODS AND RESULTS: The optimal temperature and initial pH for the production of both mycelial biomass and EPSs in P. gilvus by submerged flask cultures were found to be 30 degrees C and pH 9.0, respectively. Glucose and corn steep powder were the most suitable carbon and nitrogen source for both mycelial biomass and EPS production. Optimal medium composition was determined to be glucose 30 g l-1, corn steep powder 5 g l-1, MgSO4 1.23 g l-1, KH2PO4 0.68 g l-1, and K2HPO4 0.87 g l-1. Four groups of EPSs (Fr-I, II, III, and IV) were obtained from the culture filtrates by gel filtration chromatography on Sepharose CL-4B and characterized by size exclusion chromatography (SEC) coupled with MALLS. The weight average molar mass (Mw) of Fr-I, Fr-II, Fr-III and Fr-IV were determined to be 8.628 x 106 (+/-129 420), 1.045 x 106 (+/-19 855), 61.09 x 104 (+/-1244), and 33.55 x 104 (+/-134) g mol-1, respectively. CONCLUSIONS: Under optimal culture conditions, the maximum EPS production in a 5-l stirred fermenter indicated 5.3 g l-1 after 11 days of fermentation. The SEC/MALLS analysis revealed that Fr-I, which has extremely high molecular weight, was presumably an aggregate of complex polysaccharides forming a compact globular shape; whereas Fr-II was nearly spherical, Fr-III and Fr-IV were rod-like chains in an aqueous solution. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first report on the production of high amounts of EPSs from liquid-culture of the basidiomycete, P. gilvus. The SEC/MALLS approach used in this study could be useful in providing greater insight into the characterization of the mushroom polysaccharides without carrying out elaborate fractionation procedures prior to analysis.  相似文献

Zusammenfassung Mit Hilfe verschiedener Präparationsmethoden wurde aus dem Mycel und der Nährlösung des Basidiomyceten Phellinus igniarius eine Peroxidase (EC 1.11.1.7) gewonnen.Das Enzym ist gegenüber äußeren Einflüssen (pH, Salzkonzentration, Temperatur) bemerkenswert stabil. Sein pH-Optimum liegt im sauren Bereich. Zwei Isoenzyme wurden gefunden. Die Molmasse, der isoelektrische Punkt, die Michaelis-Menten-Konstante, die Indolessigsäure-Oxidase-Aktivität sowie verschiedene spektrale und analytische Eigenschaften dieser Peroxidase wurden bestimmt. Die Aktivität des Enzyms läßt sich durch Effektoren sowohl hemmen als auch verstärken. Es ist anzunehmen, daß das Enzym einen intra- und einen extracellulären Wirkbereich hat.
Purification and characterization of peroxidase from Phellinus igniarius
A Peroxidase (EC 1.11.1.7) of the basidiomycet Phellinus igniarius was derived from mycel and a medium containing glucose and extract of yeast by using various methods of preparation.The enzyme resists extreme conditions (pH, temperature, salt concentration). Its optimum pH for activities is in the acid range. Two isoenzymes were found. The molecular weight, isoelectric point, Michaelis-Menten constant, indolacetic acid oxidase activity and spectral and analytical properties of this peroxidase were determined. It is assumed that the enzyme has an intracellular as well as an extracellular field of activity.

Although the individual consumption of medicinal mushrooms, including Phellinus linteus (PL), Ganoderma lucidum (GL), and Inonotus obliquus (IO), is known to be neuroprotective, the associated mechanisms underlying their therapeutic synergism on focal cerebral ischemia (fCI) have yet to be elucidated. This study aimed to demonstrate the neuroprotective effects of mixed mushroom mycelia (MMM) against experimental fCI. The water-fractions, ethanolic-fractions, and ethyl acetate-fractions of the MMM (PL, GL, and IO) grown in a barley medium using solid-state fermentation techniques were prepared and their protective effects against glutamate-induced excitotoxicity were compared in PC-12 cells. After the identification of the water extracts of MMM (wMMM) as the most suitable form, which possessed the lowest toxicity and highest efficacy, further analyses for evaluating the anti-apoptotic effects of wMMM, including Hoechst 33258-based nuclear staining, fluorescence-activated cell sorting, and reactive oxygen species (ROS) detection assays, were performed. Rats were subjected to a 90 min middle cerebral artery occlusion and reperfusion, after which a wMMM treatment resulted in significant dose-dependent improvements across a number of parameters. Furthermore, measurements of intracellular ROS and levels of antioxidant enzymes revealed a wMMM-mediated ROS attenuation and antioxidant enzyme upregulation. We suggest that wMMM is neuroprotective against fCI through its anti-apoptotic and anti-oxidative effects.  相似文献

We compared the immunomodulating effects of Phellinus linteus (PL), germinated brown rice (BR) and P.␣linteus grown on germinated brown rice (PB) on IgE production in murine mesenteric lymph node (MLN) lymphocytes. All extracts decreased IgE concentrations by 43–65% compared to control mice in both serum and MLN lymphocytes. In addition, PL and PB increased the proportion of CD4+ T cells by␣9% and 12% in MLN lymphocytes. IFN-γ concentration, Th1 cytokine, was significantly increased by 44–67%, whereas IL-4 and IL-10 concentrations, Th2 cytokine, significantly decreased by 30–60% in the three treated groups compared to control group. These results suggest that PB suppresses IgE production through the modulation of Th1/Th2 balance to down-regulate Th2 response in MLN lymphocytes, even though a synergistic effect of PB was not found.  相似文献

This study presents the current knowledge on chemical composition, biological activity, and possible medicinal applications of Phellinus igniarius, Phellinus pini, Phellinus pomaceus, and Phellinus robustus. These inedible arboreal species are phytopathogens that cause the enzymatic decomposition of wood. These species belong to the medicinal mushrooms and have been known for centuries in the traditional medicine of the Far East. They have been used as an effective remedy for stomach and intestinal ailments, diarrhea, and hemorrhages. Mycochemical studies have proved the presence of polysaccharides, phenolic compounds, and terpenoids. These compounds show biological activities such as anticancer, antioxidant, antiangiogenic, and antiviral. Research studies conducted using modern analytical methods have advanced the knowledge on the potential therapeutic use of compounds isolated not only from the fruiting bodies but also from biomass obtained with in vitro biotechnological methods.  相似文献

本研究通过测定人工段木栽培3年生杨树桑黄Sanghuangporus vaninii、鲍姆桑黄S. baumii、桑树桑黄S. sanghuang及野生桑树桑黄子实体的粗蛋白质、粗脂肪、粗纤维、氨基酸、多糖、三萜、总黄酮、总酚含量和抗氧化活性(ABTS和FRAP),探究此3种栽培桑黄及野生桑树桑黄子实体的营养、药效成分及抗氧化活性差异。结果表明,供试材料在营养、药效成分及抗氧化活性上差异较大。其中,栽培鲍姆桑黄子实体粗纤维含量最低,多糖含量较高,总黄酮、总酚含量和ABTS、FRAP活性最高;栽培杨树桑黄粗纤维含量较低,粗脂肪含量最低,总黄酮、总酚、三萜含量较高,多糖含量最高;野生桑树桑黄粗蛋白、粗脂肪、总氨基酸含量最高,多糖含量较高,三萜含量最高,总黄酮、总酚含量和ABTS、FRAP活性最低;栽培桑树桑黄粗纤维含量较高,多糖、总黄酮、总酚、三萜含量和ABTS、FRAP均较低。栽培杨树桑黄、鲍姆桑黄和野生桑树桑黄在药效成分含量上各有所长。筛选到可结实、药效成分含量高的桑树桑黄菌株是可能的。桑黄优良品种选育是今后的工作重点。本研究为桑黄真菌资源开发利用提供了参考。  相似文献

‘浙黄1号’是野生桑黄通过驯化系统选育的新品种。其子实体呈扇形或马蹄形,菌盖长径6.0-15.0cm,短径3.0-5.0cm,基部厚2.0-5.0cm,边缘厚0.2-2.0cm。子实体表面呈金黄色至棕黄色,菌肉黄褐色。菌丝体生长适温25-30℃,子实体发育适温20-25℃。子实体总黄酮含量8.26%,粗多糖含量5.40%,总三萜含量0.96%。浙江省地区有春、秋两季栽培;抗逆能力较强,子实体成品率较高、整齐,商品化程度高,产量13.3g/袋(干料470g)。适宜代料栽培。  相似文献

Chemical investigation of the fungus Phellinus baumii has resulted in characterization of five previously undescribed hispidin derivatives, phellibaumins A-E (1-5), as well as two pairs of new non-equivalent epimeric benzyl dihydroflavones, methylphelligrin A (9), epi-methylphelligrin A (10), methylphelligrin B (11), and epi-methylphelligrin B (12), together with five known compounds, interfungin B (6), phelligridin H (7), phelligridimer A (8), phelligrin A (13), and epi-phelligrin A (14). Phellibaumin A (1) was a novel hispidin derivative with a unique 3,4-dihydroxybenzofuran unit. These compounds exhibited NF-κB inhibitory activity with IC50 values of 52.96 μM (1), 41.40 μM (2), 52.92 μM (5), 36.44 μM (9 and 10), and 22.46 μM (11 and 12), respectively.  相似文献

A new trimeric hispidin derivative, phellinstatin, was isolated from a culture broth of the medicinal fungus Phellinus linteus and its structure was established by various spectral analysis. Phellinstatin strongly inhibited Staphylococcus aureus enoyl-ACP reductase with an IC50 of 6 μM and also showed antibacterial activity against S. aureus and MRSA.  相似文献
