 目的 探讨NRP-1单克隆抗体(NRP-1 MAb)的特异性,以及不同剂量的NRP-1 MAb治疗乳腺癌裸鼠移植瘤的疗效。方法 Western blot和共聚焦免疫荧光法检测NRP-1 MAb是否识别MCF7细胞上NRP-1蛋白。将MCF7细胞接种于BALB/c裸鼠皮下建立乳腺癌细胞移植瘤模型,并进行瘤组织传代。传代的肿瘤体积生长至300~500 mm3时,随机分为对照组、NRP-1 MAb低剂量组、中剂量组和高剂量组,每组6只,给药7次。观察荷瘤裸鼠一般状况,测量瘤体大小及裸鼠体重。实验结束时剥离瘤体称重,提取组织蛋白,Western blot检测组织中VEGF蛋白和NRP-1蛋白的表达量。结果 NRP-1MAb成功识别MCF7细胞上的NRP-1蛋白;NRP-1 MAb能够有效抑制MCF7细胞裸鼠移植瘤的生长,低剂量组(1 mg/kg)抑瘤率为47.01%,中剂量组(5 mg/kg)抑瘤率为65.70%,高剂量组(10 mg/kg)抑瘤率为69.19%。。结论 NRP-1 MAb能够识别并有效结合MCF7细胞膜上的NRP-1蛋白,且可抑制MCF7细胞移植瘤的生长,NRP-1 MAb抑制移植瘤的增长可能与下调NRP-1和VEGF表达有关。  相似文献

目的 利用PCR—RFLP和PCR—SSCP技术分别检测犬MC1R基因中两个序列T105A和R306Ter的基因多态性,为遗传育种、培育出更加优良的实验用犬奠定基础。方法 以224只犬为实验材料,对犬MC1R基因T105A片段和R306Ter片段分别进行PCR-RFLP和PCR—SSCP分析,并且分别对具有RFLP和SSCP多态性的片段进行克隆测序,找出等位基因的差异位点。结果 经对犬MClR基因的PCR-RFLP分析,发现犬MC1R基因的T105A序列具有多态性,表现为三种基因型:AA、AB和BB。通过对具有RFLP多态性的片段进行克隆测序发现MC1R基因在编码第105位氨基酸的密码子处存在由G到A的单碱基突变,该突变导致第105位氨基酸发生由精氨酸向苏氨酸的改变,此外在第207位氨基酸的密码子处还发现由A到G的单碱基突变,并产生了一个HhαI限制性内切酶位点。同时,经过犬MC1R基因的PCR—SSCP分析,发现犬MC1R基因的R306Ter序列具有多态性,表现为三种基因型:CC、CD、DD.通过对具有SSCP多态性的片段进行了克隆测序发现,MC1R基因在编码第306位氨基酸的密码子处存在一个由CGA到TGA的终止突变,而使翻译终止。结论 利用PCR—RFLP和PCR—SSCP分析技术证实犬MC1R基因具有明显的多态性。  相似文献

We describe a simple direct extraction method for the gas-liquid chromatography determination of serum valproic acid. The working range for the assay is 2-180 mg/L and our within-run precision was 5.8 and 4.3% at the 40 and 90 mg/L concentrations respectively. Hemolyzed and lipemic sera as well as samples from patients with hyperbilirubinemia and from patients with decreased renal function were put through the assay and no interfering peaks were noted. Interference occurred when teflon-lined screw caps were used during the extraction step. The method was proven to be accurate by linear regression analysis of samples containing weighed-in amounts of valproic acid. The above assay was compared to an enzyme immunoassay technique (EMIT). The working range for the latter is 10-150 mg/L and the with-run precision was 10.8 and 5.9% and 90 mg/L concentration respectively. Samples were run by both the gas-liquid chromatograph and enzyme immunoassay methods and gave very similar results over the range 16-139 mg/L.  相似文献

[(1R)‐endo]‐(+)‐3‐bromocamphor was dehalogenated with tritium gas to [3‐3H]camphor and via [3‐3H]phenylborneol converted to [3‐3H]deramciclane isolated as the fumarate salt (specific activity 51.8 GBq/mmol). This three step synthesis from [3‐3H]camphor gave an overall yield of 22%. Benzyloxy‐acetic acid methyl ester was reduced with sodium‐borotritide to 2‐benzyloxy‐ethanol‐[1‐3H], and through a four step procedure was converted to 2‐dimethylaminoethyl‐[2‐3H] chloride. The latter was condensed with the sodium derivative of 2‐phenylborneol giving rise to [2‐dimethylamino‐[2‐3H]ethoxy]deramciclane isolated as the fumarate (specific activity 8.177 GBq/mmol). 采用~(125)I 标记小鼠抗人IL—2受体(P55)的抗Tac(CD_(25))单克隆抗体,成功地建立了人IL—2受体的免疫放射分析法,动态观察了人外周血淋巴细胞经PHA 刺激24、48和72h 后IL—2受体表达的时间曲线,通过Scatchard 作图分析表明~(125)I—抗Tac 单克隆抗体与PHA 活化的上述三个时间点的T 淋巴细胞的最大结合容量(B_(max))分别为43000位点/细胞、54000位点/细胞和61000位点/细胞。本研究为临床IL—2受体检测提供一种简便的免疫放射测定法。  相似文献

目的:探讨固本延衰方延缓亚急性衰老模型小鼠衰老的机制。方法:将60只小鼠随机分为4组,除正常对照组外均皮下注射D-半乳糖造成衰老模型,并给予不同剂量的固本延衰方治疗,分别用放免法及流式法测定脾淋巴细胞白细胞介素2及其受体水平。结果:衰老模型组IL-2及其受体水平明显低于正常对照组(P<0.05),固本延衰方各剂量组IL-2及其受体水平明显高于衰老模型组(P<0.05,P<0.01)。结论:提示固本延衰方通过提高衰老机体淋巴细胞IL-2及其受体表达水平,从而调节机体免疫功能延缓衰老。  相似文献

Orthotopic placement of in vitro propagated Dunning R3327 AT-3 tumor cells resulted in a greater percentage of tumor takes and a two-fold shift in the exponential growth curve compared to flank implantation. The orthotopic tumor appeared to disseminate preferentially to regional lymph nodes, rather than to the lungs which is characteristic of flank tumors. The results suggest an important role of stromal-epithelial interactions in the growth of this tumor. © 1995 Wiley-Liss, Inc.  相似文献

Purpose. An X-ray powder diffractometric method was developed for the simultaneous quantification of the relative amounts of the racemic compound (±) of ibuprofen (I) and S(+)-ibuprofen (II), when they occur as a mixture. Methods. The X-ray powder diffraction patterns of I and II show pronounced differences. This formed the basis for the determination of the relative amounts of I and II when they occur as a mixture. X-ray lines with d-spacings of 14.41 and 4.37 Å were unique to I and II, respectively. Mixtures containing different proportions of I and II were prepared which also contained lithium fluoride (III) as an internal standard. Results. A linear relationship was obtained when the intensity ratio (intensity of the 4.37 Å line of II/intensity of the 2.01 Å line of III) was plotted as a function of the weight fraction of II in the mixture. Similar results were obtained in the case of I. Using these standard curves, the weight fractions of I and II in 'unknown' mixtures were determined. The experimentally determined analyte concentration ranged between 98 and 104% of the true value. The relative error in the analyses of individual samples was < 10%. The minimum detectable weight fraction of I in II and II in I were 0.032 (3.2% w/w) and 0.034 (3.4% w/w), respectively. The minimum quantifiable weight fractions were 0.136 for I and 0.112 for II. Since the X-ray diffraction patterns of S(+)-ibuprofen and R(–)-ibuprofen are identical, the conclusions drawn regarding mixtures of I and II will also hold true in the quantitative analyses of mixtures of I and R(–)-ibuprofen.  相似文献
