目的:探讨异丙酚和芬太尼混合液静脉麻醉、笑气吸入麻醉及2%利多卡因宫颈局部浸润麻醉三种镇痛方法在人工流产手术中的疗效比较。方法:自2003年8月至2004年1月对236例妇科门诊早孕要求终止妊娠的妇女实施无痛人工流产术。随机分成3组。A组:异丙酚+芬太尼静脉麻醉(85例);B组:N2O(笑气)吸入麻醉(88例);C组:2%利多卡因宫颈局部浸润麻醉(63例),观察人流术中其镇痛效果并对疼痛进行分级。结果:A组85例100%可达到完全不痛(疼痛分级为0级);B组65.90%可达到完全不痛,Ⅰ级23.86%,Ⅱ级9.09%,Ⅲ级1.15%;C组0例可达到完全不痛,Ⅰ级63.49%,Ⅱ级30.16%,Ⅲ级6.35%。结论:受术者如无心肺疾患及人流禁忌证,无痛人工流产术应选择异丙酚+芬太尼静脉麻醉可达到完全无痛,痛苦最小。  相似文献

采用~(125)I 标记小鼠抗人IL—2受体(P55)的抗Tac(CD_(25))单克隆抗体,成功地建立了人IL—2受体的免疫放射分析法,动态观察了人外周血淋巴细胞经PHA 刺激24、48和72h 后IL—2受体表达的时间曲线,通过Scatchard 作图分析表明~(125)I—抗Tac 单克隆抗体与PHA 活化的上述三个时间点的T 淋巴细胞的最大结合容量(B_(max))分别为43000位点/细胞、54000位点/细胞和61000位点/细胞。本研究为临床IL—2受体检测提供一种简便的免疫放射测定法。  相似文献

Very little information is available concerning the pharmacology of type 2 astroglia. During the past decade it has become apparent that two distinct lineages of astroglial cells can be defined in vitro. These two lineages are commonly referred to as type 1 and type 2 and are distinguished from each other on the basis of their morphological features and antigenic phenotypes. In contrast to type 1 astroglia, very little is known about the pharmacology of type 2 astroglia. The lack of information concerning the responsiveness of these cells stems primarily from difficulties encountered in isolating large numbers of type 2 astroglia free of other cell types. In the present study video- and photometer-based imaging systems were used to monitor the influence of a series of neuroligands on the intracellular calcium levels of individual cerebral type 2 astroglia in order to assess their expression of calcium-mobilizing receptors. The responses of 85 immunocytochemically identified cerebral type 2 astroglia to bradykinin (BK), norepinephrine (NE), histamine (HIST), carbachol (CARB), 2-methyl-thio ATP (2MT-ATP), glutamate (GLUT), and serotonin (5-HT) were analyzed. Approximately 50% of cerebral type 2 astroglia responded to BK, NE, HIST, CARB, and 2MT-ATP whereas only 16% and 9% of the cells responded to GLUT and 5-HT, respectively. The number of neuroligands that increased calcium in individual cells ranged from 0 to 6. These responses are quite similar to those previously demonstrated in cultured cerebral type 1 astroglia. 目的用骨形成蛋白2(BMP2)干预肝癌细胞系HepG2细胞,从而观察肝癌细胞中PTEN蛋白水平的变化,观察BMP2对PTEN蛋白表达的影响,探讨BMP2与PTEN蛋白水平之间的关系,探索肝癌治疗的新途径。方法将受试对象分为3个组:对照组、BMP2100ng/ml组、BMP2300ng/ml组,作用时间为3个水平:6、12、24h。用免疫组织化学方法测定细胞中PTEN蛋白水平。结果免疫组织化学结果显示在不同浓度BMP2组中HepG2细胞中PTEN蛋白表达差异有统计学意义。不同时间组中PTEN蛋白表达差异有统计学意义。PTEN蛋白阳性表达率在300ng/ml组与100ng/ml组、对照组之间差异有统计学意义(P<0.05),在6h组与12h组、24h组之间差异有统计学意义(P<0.05)。在300ng/ml、24h组中PTEN蛋白阳性表达最高。浓度与时间之间无交互作用。结论在肝癌中BMP2可以增加PTEN蛋白水平,且呈浓度时间依赖。  相似文献

Monoclonal antibodies against human and bovine 2′:3′-cyclic nucleotide 3′-phosphodiesterase (CNPase) were generated by fusing FOX-NY myeloma cells with spleen cells from RBF/Dn mice previously immunized with the purified brain antigens. The enzyme isolated from bovine brain was quite basic, with an isoelectric point of 9.71 and both the bovine and human enzymes consisted of a closely spaced doublet at approximately 44 and 46 kDa on SDS-PAGE. Six monoclonals were identified as strongly recognizing the enzyme on both ELISA plates and on immunoblots of whole brain protein. Four monoclonals very weakly cross-reacted with guinea pig myelin basic protein. In contrast with two previous reports, some of our monoclonal antibodies did immunostain 2 or 3 protein bands in peripheral nerve, two bands closely corresponding to those immunostained in central nervous system (CNS) myelin, the Wolfgram protein fraction and in acetone powders of whole brain. Each of the 6 monoclonals reacting strongly on immunoblots recognized the enzyme in from 2 to 5 of the species examined (human, bovine, rat, mouse and rabbit). In addition, all 6 monoclonals that immunostained the enzyme in whole brain, myelin and Wolfgram protein immunoblots recognized both CNP1 (44 kDa) and CNP2 (46 kDa). The two closely spaced protein bands observed on SDS-PAGE and previously stained on immunoblots of CNS CNPase using polyvalent rabbit anti-bovine CNPase antisera, and now different monoclonal antibodies, appear to be immunologically related and to contain highly conserved sequences.  相似文献

目的探讨性别差异对脓毒症大鼠肝脏组织Toll样受体4(TLR4)和髓样分化蛋白- 2(MD-2)基因表达的影响。方法以脂多糖(LPS)按5 mg/kg体重由大鼠腹腔注射制作脓毒症动物模型,注射后2 h留取肝脏组织检测TLR4、MD-2和肿瘤坏死因子-α(TNF-α)基因表达,同时测定各组大鼠血浆中丙氨酸氨基转移酶(ALT)及雌二醇含量。结果正常雌雄性大鼠肝脏组织均可表达少量TLR4、MD-2、TNF-α基因,其中雌性组分别为0.175±0.034、0.211±0.044、0.201±0.068; 雄性组分别为0.205±0.061、0.243±0.049、0.243±0.063,两组数据差异无统计学意义(P> 0.05),但LPS刺激后雌性大鼠肝脏组织上述指标分别为0.615±0.089、0.708±0.181、0.730± 0.118,血浆中ALT含量为(81.07±10.72)U/L;雄性组分别为0.723±0.091、1.123±0.272、 0.881±0.156,ALT含量为(106.39±14.21)U/L,雌性组各项指标均明显低于雄性大鼠(P< 0.05)。相关分析表明雌性及雄性脓毒症大鼠肝脏组织TLR4及TNF-α基因表达与相应性别大鼠血浆中雌二醇含量呈显著负相关(P<0.05)。结论 LPS刺激后大鼠肝脏组织TLR4、MD-2及 TNF-α基因表达存在性别差异,内源性雌激素的作用可能导致雌性脓毒症大鼠肝脏组织损伤较雄性轻。  相似文献

AIM: To investigate the participation of adenosine receptors in the adenosine 5'-triphosphate (ATP)-induced relaxation in the corpus cavernosum penis (CCP) of rabbits. METHODS: The ATP-induced relaxation was assessed on the noradrenaline precontracted CCP of rabbits in the presence and absence of 8-(3-chlorostyryl)caffeine (CSC); an adenosine A(2A) receptor antagonist; alloxazine and MRS1754; adenosine A(2B) receptor antagonists; and ARL67156, an inhibitor of ecto-nucleoside triphosphate diphosphohydrolases. RESULTS: Adenosine and ATP relaxed the noradrenaline precontracted CCP of rabbits in a concentration-dependent manner. The adenosine- and ATP-induced relaxations were suppressed by alloxazine and MRS1754, but not by 8-(3-chlorostyryl)caffeine. ARL67156 potentiated the ATP-induced relaxation but not the adenosine-induced one. MRS1754 suppressed the ATP-induced relaxation potentiated by ARL67156. CONCLUSIONS: The above results suggest that, in the CCP of rabbits, the adenosine receptor mediating adenosine-induced relaxation is of the A(2B) receptor and the ATP directly causes relaxation through the A(2B) receptor on the CCP.  相似文献

目的:测定survivin和Bcl-2在尖锐湿疣(CA)组织中的表达并探讨两者表达的关系。方法:应用免疫组织化学链霉菌抗生物素蛋白过氧化物酶方法(SP法)分别检测60例CA组织及10例正常包皮组织中survivin、Bcl-2基因的表达。在镜下观察切片并计算阳性细胞百分比。结果:CA组织中survivin的表达率为86.67%,显著高于正常包皮的表达率20%(P<0.01)。其中棘层的表达率85.00%显著高于基底层的表达率70.00%和颗粒层的表达率66.67%(P<0.05),基底层与颗粒层的表达率之间无显著性差异(P>0.05)。Bcl-2在CA的表达率为66.67%,正常包皮组织中未见Bcl-2蛋白表达。survivin在CA中的表达与Bcl-2呈正相关(P<0.01)。结论:本实验检测出survivin基因在CA组织中有异常表达,提示survivin蛋白引起的细胞凋亡抑制在CA的发生中起到重要的作用。survivin与Bcl-2在CA的发生发展过程中起到协同作用。  相似文献

Aims To measure the prevalence of low high‐density lipoprotein (HDL)‐cholesterol (men < 1.03 mmol/l; women < 1.29 mmol/l) in European Type 2 diabetic patients receiving treatment for dyslipidaemia. Methods The pan‐European Survey of HDL‐cholesterol measured lipids and other cardiovascular risk factors in 3866 patients with Type 2 diabetes and 4436 non‐diabetic patients undergoing treatment for dyslipidaemia in 11 European countries. Results Diabetic patients were more likely to be obese or hypertensive than non‐diabetic patients. Most patients received lifestyle interventions (87%) and/or a statin (89%); treatment patterns were similar between groups. Diabetic patients had [means (SD)] lower HDL‐cholesterol [1.22 (0.37) vs. 1.35 mmol/l (0.44) vs. non‐diabetic patients, P < 0.001] and higher triglycerides [2.32 (2.10) vs. 1.85 mmol/l (1.60), P < 0.001]. More diabetic vs. non‐diabetic patients had low HDL‐cholesterol (45% vs. 30%), high triglycerides (≥ 1.7 mmol/l; 57% vs. 42%) or both (32% vs. 19%). HDL‐cholesterol < 0.9 mmol/l was observed in 18% of diabetic and 12% of non‐diabetic subjects. Differences between diabetic and non‐diabetic groups were slightly greater for women. LDL‐ and total cholesterol were lower in the diabetic group [3.02 (1.05) vs. 3.30 mmol/l (1.14) and 5.12 (1.32) vs. 5.38 mmol/l (1.34), respectively, P < 0.001 for each]. Conclusions Low HDL‐cholesterol is common in diabetes: one in two diabetic women has low HDL‐cholesterol and one diabetic man in four has very low HDL‐cholesterol. Management strategies should include correction of low HDL‐cholesterol to optimize cardiovascular risk in diabetes.  相似文献
