The gelatinisation temperatures, pasting characteristics and enzymic susceptibilities in the temperature range 48–72°C of normal, high amylose, low amylose and zero amylose barley starches were determined. Normal starches had the lowest gelatinisation temperatures, but low and zero amylose starches had the lowest pasting temperatures. Normal starches were the most readily soluble in water at 48–60°C in the presence of a mixture of α‐amylase, β‐amylase and limit dextrinase and were most readily broken down to reducing sugars by these enzymes. High amylose starch was the most resistant to enzymic hydrolysis in the temperature range 48–72°C and, hence, produced the lowest level of reducing sugars.  相似文献

Racemic amino acid esters were optically resolved via hydrolysis in organic solvents by the catalysis of an industrial alkaline protease, “Alcalase”. The products which were composed mainly of L-amino acids were insoluble and easily separated by filtration. The activity of the enzyme and enantiomeric excess of the products were significantly dependent on the nature of solvent and the water content in the reaction media. Generally, high values of enantiomeric excess were obtained at low water contents. Many natural and unnatural amino acids were resolved by this method.  相似文献

ABSTRACT: Natural carotenoids are an alternative to synthetic orange-red pigments. They are present in crustaceans as a protein-pigment complex. In order to extract this highly unstable pigment, crustacean waste must be stabilized; lactic fermentation is a simple and environmentally friendly method to achieve this goal. Shrimp wastes were inoculated with Lactobacillus bacterial cultures. Carotenoids were then extracted with an organic solvent system. Protein-pigment splitting was carried out using a mixture of 4 commercial enzymes; and the protein was separated from the pigment by ultrafiltration. Electrophoretograms showed that the pigment was attached to a 265-kDa protein. Splitting the protein-pigment complex allows studies on pigment absorption, stability and application.  相似文献

Candida rugosa lipase (CRL) and Pseudomonas fluorescein lipase (PFL) were used to hydrolyze a low-melting point butteroil fraction in the absence of orgaic solvents. CRL and PFL displayed pH activity optima of 6.7 and 7.8, and temperature-activity optima of 40–50°C and 35°C, respectively. Maximum activity for PFL was observed at 5% (v/v) water content. CRL showed higher specificity towards butyric acid relative to PFL after 3 hours of hydrolysis at 40°C (22.3 mol% vs 7.8 mol%, respectively). Both enzymes displayed, however, high specificities towards palmitic and oleic acids. CRL's inherent specificity towards butyric acid makes it an ideal choice for a biological catalyst to generate useful dairy flavor profiles.  相似文献

Dispersion–flocculation studies on a Goethite–clay system using flocculants were carried out as a function of flocculant concentrations, pH of slurry, time of agitation and dispersant dosage. Also, the effects of pH and polymer concentrations on the adsorption behaviour of the system were investigated. Results show that well flocculated goethite was preferentially obtained from 4% goethite/kaolinite clay suspensions, with 50 ppm causticised starch in the slurry at pH values of 3–11·5 and with 50 ppm polyacrylamide at pH values of 3–8. Good flocs were also obtained on flocculation of the goethite suspension with 50 ppm polyacrylamide at pH values of 5–7, while the kaolinite suspension did not respond to the same dosage of causticised starch in the same pH range. Results further reveal that for the goethite/kaolinite suspension, the best results was obtained with 50 ppm polyacrylamide at pH values of 7–10. Based on the data generated in the study, it was concluded that causticised starch is a better flocculant than polyacrylamide for goethite suspensions although polyacrylamide is an excellent flocculant for kaolinite suspensions.  相似文献

According to D. E. Broadbent's (1958) selective filter theory, people do not process unattended stimuli beyond the analysis of basic physical properties. This theory was later rejected on the basis of numerous findings that people identify irrelevant (and supposedly unattended) stimuli. A careful review of this evidence, however, reveals strong reasons to doubt that these irrelevant stimuli were in fact unattended. This review exposed a clear need for new experiments with tight control over the locus of attention. The authors present 5 such experiments using a priming paradigm. When steps were taken to ensure that irrelevant stimuli were not attended, these stimuli produced no priming effects. Hence, the authors found no evidence that unattended stimuli can be identified. An effective two-stage method has been developed for imparting antimicrobial properties to regular polyethylene terephthalate (R-PET), polyethylene glycol modified polyethylene terephthalate (PEG-M-PET), R-PET/Cotton blend (R-PET/C) and PEG-M-PET/Cotton blend (PEG-M-PET/C) fabrics. The method consists of partial hydrolysis of the fabrics to create carboxylic groups in PET macromolecules followed by subsequent reaction with dimethylalkylbenzyl ammonium chloride (DMABAC) under alkaline conditions. The reaction conditions such as pH, reaction temperature and time, carboxylic content, and DMABAC concentration were studied. Characterization of the finished fabrics was carried out through scanning electron microscopy (SEM) and Fourier transform infrared spectra (FTIR). All the modified PET fabrics showed excellent antibacterial activity towards Gram-positive (Bacillus mycoides), Gram-negative (Escherichia coli), and nonfilamentous fungus (Candida albicans). The achieved antimicrobial functions on the PET fabrics are durable in repeated laundering processes. Even after laundering 10 times the fabrics could still provide more than 85% of its antimicrobial activity against B. mycoides, E. coli, and C. albicans. © 2008 Wiley Periodicals, Inc. The relationship between facial expression and gaze processing was investigated with the Garner selective attention paradigm. In Experiment 1, participants performed expression judgments without interference from gaze, but expression interfered with gaze judgments. Experiment 2 replicated these results across different emotions. In both experiments, expression judgments occurred faster than gaze judgments, suggesting that expression was processed before gaze could interfere. In Experiments 3 and 4, the difficulty of the emotion discrimination was increased in two different ways. In both cases, gaze interfered with emotion judgments and vice versa. Furthermore, increasing the difficulty of the emotion discrimination resulted in gaze and expression interactions. Results indicate that expression and gaze interactions are modulated by discriminability. Whereas expression generally interferes with gaze judgments, gaze direction modulates expression processing only when facial emotion is difficult to discriminate.
