以2-烃基-2-羟甲基-1,3-丙二醇为原料, 经保护两个羟基, PCC氧化, 引入氰基, 脱保护后与POCl3或PSCl3环合, 成功合成了14个4-烃基-3-氰基双环笼状磷酸酯类化合物, 目标化合物的结构通过1H NMR, MS和元素分析进行了表征. 采用放射性配体受体结合实验, 测定了14个化合物抑制[3H]EBOB [4’-ethynyl-4-n-propylbicycloorthobenzoate]与家蝇及大鼠GABA受体的结合活性. 结果表明部分化合物在10-5 mol/L具有较好的抑制活性, 且在大鼠与家蝇GABA受体之间具有一定选择性.  相似文献

N,N-dimethyl-N-methacryloyloxyethyl-N-carboxyethyl ammonium (DMMCA) was graft-copolymerized onto the surface of segmented poly(ether urethane) (SPEU) and PE film. The carboxybetaine structure on SPEU and PE film surfaces was confirmed by ATR-FTIR, XPS and water contact angle measurements. Through the experiments with platelet adhesion and protein adhesion assay in vitro, the two materials studied, including poly-DMMCA gel, all show excellent nonthrombogenicity. This confirms once again that the zwitterionic molecular structure on the surfaces of materials is essential for improving their nonthrombogenicity and biocompatibility.  相似文献

采用活化酯法,将马兜铃酸A分别与牛血清蛋白(BSA)和卵清蛋白(OVA)偶联,得到免疫抗原马兜铃酸A-BSA和包被抗原马兜铃酸A-OVA.利用马兜铃酸A-BSA免疫Bal b/c小鼠,制得鼠单克隆抗体1A11,单抗效价为2×104;单抗为IgG1类,轻链为κ型;与其结构类似物马兜铃酸B、C和D的交叉反应率分别为2.8%,3.5%和31.2%.基于抗马兜铃酸A单克隆抗体的间接竞争酶联免疫分析方法(icELISA)的IC50为1.9 μg/L,检测范围为0.5~7.5 μg/L.icELISA添加回收率为86%~97%,相对标准偏差在5.2%~11.1%之间.利用所建立的icELISA测定了6个中药材和5个中成药中马兜铃酸A的含量,并用高效液相色谱法(HPLC)进行了验证,其中关木通、广防己、天仙藤、马兜铃和青木香中均检测出马兜铃酸A,而川木通和5个中成药中未检测到马兜铃酸A.结果表明: 本方法可用于中药中马兜铃酸A的快速检测.  相似文献

An in‐house flow‐injection capillary electrophoresis with capacitively coupled contactless conductivity detection method was developed for the direct measurement of colistin in pharmaceutical samples. The flow injection and capillary electrophoresis systems are connected by an acrylic interface. Capillary electrophoresis separation is achieved within 2 min using a background electrolyte solution of 5 mM 2‐morpholinoethanesulfonic acid and 5 mM histidine (pH 6). The flow‐injection section allows for convenient filling of the capillary and sample introduction without the use of a pressure/vacuum manifold. Capacitively coupled contactless conductivity detection is employed since colistin has no chromophore but is cationic at pH 6. Calibration curve is linear from 20 to 150 mg/L, with a correlation coefficient (r2) of 0.997. The limit of quantitation is 20 mg/L. The developed method provides precision, simplicity, and short analysis time.  相似文献

考察了5-(4'-氰基苯)偶氮-8-羟基喹啉(1)-镍配合物的光谱特性并将其应用于阴离子识别研究中.研究结果表明,1-Ni2+金属配合物可选择性识别不同的阴离子,并对HSO-4具有独特的光谱响应.化合物1在乙腈中为无色溶液,与Ni2+可形成稳定常数为9.2×10 6mol-1·L的1∶1型红色配合物,在此配合物中加入HSO-4后,红色的溶液逐渐褪色,而加入CH3CO-2,F-和H2PO-4却产生增色效应,Cl-和Br-加入后主体溶液无明显颜色和光谱变化,据此建立了选择性识别HSO-4新体系,并探讨其作用机理.  相似文献

用MTT法研究了14种稀土元素(La,Ce,Pr,Nd,Sm,Eu,Gd,Tb,Dy,Ho,Er,Tm,Yb和Lu)对人肝癌细胞SMMC-7721增殖的影响。他们对肝癌细胞的生长作用可分为3类。其中La^3 、Ce^3 和Eu^3 对肝癌细胞的增殖有剂量依赖性正效应,能够在一定浓度范围内刺激细胞生长;Sm^3 ,Gd^3 ,Ho^3 ,Er^3 ,Yb^3 对肝癌细胞生长的刺激作用没有剂量依赖性特征;而Pr^3 ,Nd^3 ,Tb^3 ,Dy^3 ,Tm^3 和Lu^3 则表现出对肝癌细胞的增殖具有不用程度的抑制。推测14种稀土元素作用方式的不同与他们的原子结构有一定的关系,它们对肝癌细胞的相对增殖率随着原子序数的增加呈现出一定的规律性。  相似文献

We investigated the mechanisms of DNA exit during single‐cell gel electrophoresis (the comet assay) by measuring the kinetics of the comet tail formation. In the neutral comet assay, the rate of DNA exit was found to be dependent on the topological state of DNA, which was influenced by either ethidium bromide or a low radiation dose. The results clearly show that the comet tail is formed by extended DNA loops: the loop extension, being reversible when the DNA torsional constraint remains in the loops, is favored when the constraint is relaxed. The kinetics of the comet formation in the case of a high radiation dose points out that accumulation of the single‐strand breaks causes DNA fragmentation. In contrast to the neutral comet assay, the alkaline comet assay is not related to the chromatin loops. Our results imply that the alkaline treatment induces detachment of the loops from the nuclear matrix, and the comet tail is formed by ssDNA fragments, the ends of which are pulled out from the comet head by electric force. We suggest that the kinetic approach can be considered as an important improvement of the comet assay.  相似文献

This paper presents a new simple and sensitive method for the micro-determination of protein containing SH– and –S–S– groups based on the single sweep polarographic wave of an electroactive derivative. In 0.04molL–1 Na3PO4 and 0.2% ascorbic acid solution, protein is heated in a boiling water bath for 15min, the reaction product giving a sensitive reduction wave at –0.70V (vs. SCE). The wave height is linearly proportional to the concentration of protein. The calibration curves of bovine serum albumin (BSA), human serum albumin (HSA), ovalbumin (OVA) and lysozyme (Lyso) are constructed under the optimal conditions. For BSA and HSA, the linear ranges and detection limits are 0.05–24mgL–1 and 0.02mgL–1, respectively. The method has been applied to the determination of protein in human serum samples with satisfactory results. The mechanism of the polarographic wave was also studied, and the results show that S2– ion is released from the protein molecule during the derivatization reaction, the wave being attributed to the reduction of HgS.  相似文献

Two rapid,sensitive and reliable immunoassay methods,namely competitive indirect enzyme-linked immunosorbent assay(CI- ELISA)and colloidal gold-based immunochromatographic assay(CGIA),were developed to detect ofloxacin(OFL).The linear range of the CI-ELISAwas from 0.5 to 128 ng/mL with a limit of detection(LOD)of 0.35 ng/mL.Good recoveries were obtained in analyzing simulated swine urine samples.The CGIA could accurately estimate OFL at concentrations as low as 10 ng/mL in less than 10 min,and test results were read visually without any instrument.  相似文献

S-nitrosylated proteins are biomarkers of oxidative damage in aging and Alzheimer's disease (AD). Here, we report a new method for detecting and quantifying nitrosylated proteins by capillary gel electrophoresis with laser induced fluorescence detection (CGE-LIF). Dylight 488 maleimide was used to specifically label thiol group (SH) after switching the S-nitrosothiol (S-NO) to SH in cysteine using the "fluorescence switch" assay. In vitro nitrosylation model-BSA subjected to S-nitrosoglutathione (GSNO) optimized the labeling reactions and characterized the response of the LIF detector. The method proves to be highly sensitive, detecting 1.3 picomolar (pM) concentration of nitrosothiols in nanograms of proteins, which is the lowest limit of detection of nitrosothiols reported to date. We further demonstrated the direct application of this method in monitoring protein nitrosylation damage in MQ mediated human colon adenocarcinoma cells. The nitrosothiol amounts in MQ treated and untreated cells are 14.8±0.2 and 10.4±0.5 pmol/mg of proteins, respectively. We also depicted nitrosylated protein electrophoretic profiles of brain cerebrum of 5-month-old AD transgenic (Tg) mice model. In Tg mice brain, 15.5±0.4 pmol of nitrosothiols/mg of proteins was quantified while wild type contained 11.7±0.3 pmol/mg proteins. The methodology is validated to quantify low levels of S-nitrosylated protein in complex protein mixtures from both physiological and pathological conditions.  相似文献
