A mixed gas of nitrogen, carbon dioxide, and hydrogen was discharged over 100 ml of 0.2M NaHCO3 solution in a 5 liter discharge apparatus which simulates the primitive Earth. The formation of cyanate, which is one of the possible primitive condensing agents, was demonstrated by the detection of [Cu(Py)2] (NCO)2 that was formed by the addition of copper sulfate-pyridine reagent to the solution. In a series of experiments the partial pressures of nitrogen and carbon dioxide in the starting gas were fixed at 10 cm Hg and 20 cm Hg, respectively, whereas that of hydrogen was varied between 5, 10, 15, 20, 25, and 30 cm Hg. The discharges were continued for one week. The rate of appearance of cyanate was strongly dependent upon the partial pressure of hydrogen. The maximum rate of the production of cyanate at the initial stage of the discharge was in the case of 10 cm Hg of hydrogen, in which condition the starting gas is in a predominantly oxidized state. In this case the concentration of cyanate reached about 0.012M after one day. Another discharge experiment was carried out with 0.2M phosphate solution, and the production of carbamyl phosphate was demonstrated through the formation of ATP by the incubation of the discharged solution with ADP and carbamyl phosphokinase.  相似文献

The outcome of hepatitis C virus (HCV) infection varies among individuals, but the genetic factors involved remain unknown. We conducted a population-based association study in which 238 Japanese individuals positive for anti-HCV antibody were genotyped for 269 single nucleotide polymorphisms (SNPs) in 103 candidate genes that might influence the course of infection. Altogether, 50 SNPs in 32 genes were listed. Genetic polymorphisms in IL4, IL8RB, IL10RA, PRL, ADA, NFKB1, GRAP2, CABIN1, IFNAR2, IFI27, IFI41, TNFRSF1A, ALDOB, AP1B1, SULT2B1, EGF, EGFR, TGFB1, LTBP2, and CD4 were associated with persistent viremia (P < 0.05), whereas those in IL1B, IL1RL1, IL2RB, IL12RB1, IL18R1, STAT5A, GRAP2, CABIN1, IFNAR1, Mx1, BMP8, FGL1, LTBP2, CD34, and CD80 were associated with different serum alanine aminotransferase levels in HCV carriers (P < 0.05). The sorted genes allow us to draw novel hypotheses for future studies of HCV infection to ultimately identify bona fide genes and their variations.  相似文献

MK is a gene that is expressed temporarily during the early stages of retinoic acid-induced differentiation of embryonal carcinoma (EC) cells and during the mid-gestation period of mouse embryogenesis. The 5'-regions of MK cDNAs and their mRNAs are heterogeneous; so far three kinds of MK cDNAs (MK1, MK2, and MK3) have been isolated. The MK gene was cloned from a genomic DNA library of a BALB/c mouse, and its structure was elucidated. 5'-Region sequences specific for MK1, MK2, and MK3 were arranged in the order of MK3, MK2, and MK1. Then, there was a sequence common to all MK cDNAs consisting of four exons. The results indicate that different species of MK mRNA are generated by the use of alternative promoters and different modes of splicing.  相似文献

The synthesis and properties of oligonucleotides (ONs) containing 9-(2,3,4-trihydroxybutyl)adenine, A C2 and A C3, are described. The ON containing A C2 involves the 3′ → 4′ and 3′ → 5′ phosphodiester linkages in the strand, whereas that containing A C3 possesses the 3′ → 4′ and 2′ → 5′ phosphodiester linkages. It was found that incorporation of the analogs, A C2 or A C3, into ONs significantly reduces the thermal and thermodynamic stabilities of the ON/DNA duplexes, but does not largely decrease the thermal and thermodynamic stabilities of the ON/RNA duplexes as compared with the case of the ON/DNA duplexes. It was revealed that the base recognition ability of A C2 is greater than that of A C3 in the ON/RNA duplexes.  相似文献

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been transmitted across all over the world, in contrast to the limited epidemic of genetically- and virologically-related SARS-CoV. However, the molecular basis explaining the difference in the virological characteristics among SARS-CoV-2 and SARS-CoV has been poorly defined. Here we identified that host sialoglycans play a significant role in the efficient spread of SARS-CoV-2 infection, while this was not the case with SARS-CoV. SARS-CoV-2 infection was significantly inhibited by α2-6-linked sialic acid-containing compounds, but not by α2–3 analog, in VeroE6/TMPRSS2 cells. The α2-6-linked compound bound to SARS-CoV-2 spike S1 subunit to competitively inhibit SARS-CoV-2 attachment to cells. Enzymatic removal of cell surface sialic acids impaired the interaction between SARS-CoV-2 spike and angiotensin-converting enzyme 2 (ACE2), and suppressed the efficient spread of SARS-CoV-2 infection over time, in contrast to its least effect on SARS-CoV spread. Our study provides a novel molecular basis of SARS-CoV-2 infection which illustrates the distinctive characteristics from SARS-CoV.  相似文献

Mice were exposed to 1000 R of X-rays to their trunks and sacrificed every day up to the tenth day after exposure. Cell counts were made on histological sections of the duodenum. The cell counts in the crypts were reduced to about 50% of the control value on the first day after exposure. The cell counts began to recover on the third day and an overshoot of 170% was observed on the fourth day; thereafter the crypt cell counts tended to return to the control level. The cell counts on the villi reached their minimum value on the third day after exposure. Following an overshoot on the sixth day, the villus cell counts returned to the control level by the tenth day. The above experimental results were analysed using a two-compartment model with a feedback term. A logistic proliferation was assumed for the proliferative crypt cells, while for the postmitotic villus cells the compartment was assumed to be a first in-first out type. The calculated results with this model are in general consistent with the experimental ones. The model seems to possess some essential features of the dynamics of cell renewal in the intestinal mucosa.  相似文献
