The wingless (Wnt) family of signaling ligands contributes significantly to lung development and is highly expressed in patients with usual interstitial pneumonia (UIP). We sought to define the cellular distribution of Wnt5A in the lung tissue of patients with idiopathic pulmonary fibrosis (IPF) and the signaling ligands that control its expression in human lung fibroblasts and IPF myofibroblasts. Tissue sections from 40 patients diagnosed with IPF or UIP were probed for the immunolocalization of Wnt5A. Further, isolated lung fibroblasts from normal or IPF human lungs, adenovirally transduced for the overexpression or silencing of Wnt7B or treated with TGF-β1 or its inhibitor, were analyzed for Wnt5A protein expression. Wnt5A was expressed in IPF lungs by airway and alveolar epithelium, smooth muscle cells, endothelium, and myofibroblasts of fibroblastic foci and throughout the interstitium. Forced overexpression of Wnt7B with or without TGF-β1 treatment significantly increased Wnt5A protein expression in normal human smooth muscle cells and fibroblasts but not in IPF myofibroblasts where Wnt5A was already highly expressed. The results demonstrate a wide distribution of Wnt5A expression in cells of the IPF lung and reveal that it is significantly increased by Wnt7B and TGF-β1, which, in combination, could represent key signaling pathways that modulate the pathogenesis of IPF. 相似文献

Activation of CD4(+) T cells helps establish and sustain CD8(+) T cell responses and is required for the effective clearance of acute infection. CD4-deficient mice are unable to control persistent infection and CD4(+) T cells are usually defective in chronic and persistent infections. We investigated the question of how persistent infection impacted pre-existing lymphocytic choriomeningitis virus (LCMV)-specific CD4(+) T cell responses. We identified class II-restricted epitopes from the entire set of open reading frames from LCMV Armstrong in BALB/c mice (H-2(d)) acutely infected with LCMV Armstrong. Of nine epitopes identified, six were restricted by I-A(d), one by I-E(d) and two were dually restricted by both I-A(d) and I-E(d) molecules. Additional experiments revealed that CD4(+) T cell responses specific for these epitopes were not generated following infection with the immunosuppressive clone 13 strain of LCMV. Most importantly, in peptide-immunized mice, established CD4(+) T cell responses to these LCMV CD4 epitopes as well as nonviral, OVA-specific responses were actively suppressed following infection with LCMV clone 13 and were undetectable within 12 days after infection, suggesting an active inhibition of established helper responses. To address this dysfunction, we performed transfer experiments using both the Smarta and OT-II systems. OT-II cells were not detected after clone 13 infection, indicating physical deletion, while Smarta cells proliferated but were unable to produce IFN-gamma, suggesting impairment of the production of this cytokine. Laser microsurgery and finite element modeling are used to determine the cell-level mechanics of the amnioserosa—a morphogenetically crucial epithelium on the dorsal surface of fruit fly embryos (Drosophila melanogaster). In the experiments, a tightly focused laser ablates a subcellular hole (1 μm in diameter) that passes clean through the epithelium. The surrounding cells recoil from the wound site with a large range of initial recoil velocities. These depend on the embryo's developmental stage and the subcellular wound site. The initial recoil (up to 0.1 s) is well reproduced by a base finite element model, which assumes a uniform effective viscosity inside the cells, a constant tension along each cell-cell boundary, and a large, potentially anisotropic, far-field stress—one that far exceeds the stress equivalent of the cell-edge tensions. After 0.1 s, the experimental recoils slow dramatically. This observation can be reproduced by adding viscoelastic rods along cell edges or as a fine prestressed mesh parallel to the apical and basal membranes of the cell. The marine-biodiversity assessment of New Zealand (Aotearoa as known to Māori) is confined to the 200 nautical-mile boundary of the Exclusive Economic Zone, which, at 4.2 million km2, is one of the largest in the world. It spans 30° of latitude and includes a high diversity of seafloor relief, including a trench 10 km deep. Much of this region remains unexplored biologically, especially the 50% of the EEZ deeper than 2,000 m. Knowledge of the marine biota is based on more than 200 years of marine exploration in the region. The major oceanographic data repository is the National Institute of Water and Atmospheric Research (NIWA), which is involved in several Census of Marine Life field projects and is the location of the Southwestern Pacific Regional OBIS Node; NIWA is also data manager and custodian for fisheries research data owned by the Ministry of Fisheries. Related data sources cover alien species, environmental measures, and historical information. Museum collections in New Zealand hold more than 800,000 registered lots representing several million specimens. During the past decade, 220 taxonomic specialists (85 marine) from 18 countries have been engaged in a project to review New Zealand''s entire biodiversity. The above-mentioned marine information sources, published literature, and reports were scrutinized to give the results summarized here for the first time (current to 2010), including data on endemism and invasive species. There are 17,135 living species in the EEZ. This diversity includes 4,315 known undescribed species in collections. Species diversity for the most intensively studied phylum-level taxa (Porifera, Cnidaria, Mollusca, Brachiopoda, Bryozoa, Kinorhyncha, Echinodermata, Chordata) is more or less equivalent to that in the ERMS (European Register of Marine Species) region, which is 5.5 times larger in area than the New Zealand EEZ. The implication is that, when all other New Zealand phyla are equally well studied, total marine diversity in the EEZ may be expected to equal that in the ERMS region. This equivalence invites testable hypotheses to explain it. There are 177 naturalized alien species in New Zealand coastal waters, mostly in ports and harbours. Marine-taxonomic expertise in New Zealand covers a broad number of taxa but is, proportionately, at or near its lowest level since the Second World War. Nevertheless, collections are well supported by funding and are continually added to. Threats and protection measures concerning New Zealand''s marine biodiversity are commented on, along with potential and priorities for future research. 相似文献

Capsule: Long-term trends in Merlin Falco columbarius breeding performance remained stable during a period of extensive afforestation in Ireland, where Merlin predominantly select conifer plantations for nesting.Aims: To determine breeding performance and habitat selection of Merlin in a landscape significantly altered by afforestation.Methods: We compiled data on Merlin to determine long-term trends in breeding performance and to examine habitat selection in a country with one of the fastest rates of afforestation in Europe.Results: Merlin predominantly nested in trees (99.5%; n?=?183 pairs), with a strong preference for conifer plantations, which accounted for over 12 times more nests than expected by random selection. Moors and heathland were strongly selected as land-uses adjacent to nest sites. Most nests were located within 10?m of the forest edge, and in forests aged between 31 and 40 years. Merlin showed positive selection for moors and heathland, peat bogs and natural grasslands within breeding territories, and breeding success was positively related to the proportion of these land-uses surrounding nests. Breeding was successful for 74% of nests (n?=?300), and mean productivity was 2.1 young per breeding attempt (n?=?265) between 1982 and 2014. Breeding parameters remained constant over the 33-year study period, despite an increase of more than 75% in forest cover during this time.Conclusion: Merlin breeding performance showed no long-term effects of increased afforestation. Although Merlin predominantly nested in conifer plantations, the presence of nearby open suitable foraging habitats influenced nest site selection and breeding success. The nesting preference of Merlin makes them vulnerable to disturbance from forest operations, which requires mitigation. 相似文献

Our goal was to synthesize new stereospecific benzovesamicol analogues, which could potentially be used as SPECT or PET radioligands for the vesicular acetylcholine transporter (VAChT). This paper describes the chemical synthesis, resolution and determination of binding affinity for four enantiomeric pairs of derivatives. Their intrinsic affinities were determined by competition against binding of [3H]vesamicol to human VAChT. Of the eight enantiomers, (E)-(R,R)-5-AOIBV [(R,R)-3], and (R,R)-5-FPOBV [(R,R)-4] displayed the highest binding affinities for VAChT (Kd=0.45 and 0.77 nM, respectively), which indicated that an elongation of the chain from 5-idodo as in the case of 5-iodobenzovesamicol (5-IBVM), to a 5-(E)-3-iodoallyloxy or 5-fluoropropoxy substituent, as in 5-AOIBV and 5-FPOBV, respectively, was very well tolerated at the vesamicol binding site. The enantiomer (R,R)-4-MAIBV [(R,R)-16], which retains the basic structure of (-)-5-IBVM but possess an additional aminomethyl substituent in the 4-position of the piperidine ring, displayed lower binding affinity (Kd=8.8 nM). Nevertheless, the result suggests that substitution at this position may be an interesting alternative to investigate for development of new benzovesamicol analogues. As expected, the corresponding (S,S) enantiomers displayed lower Kd values, they were approximately 10-fold lower in the case of (S,S)-5-FPOBV (Kd=8.4 nM) and (E)-(S,S)-5-AOIBV (Kd=4.3 nM). (R,R)-3, and (R,R)-4 showed the same high affinity for VAChT as (-)-5-IBVM and may be suitable as imaging agents of cholinergic nerve terminals. 相似文献

Urbanization leads to degradation in water quality and has a major effect on the biota of streams, but its effect on microbial communities is not as well understood. DNA-techniques that target functional genes are being used to examine microbial communities, but less frequently applied to freshwater aquatic systems. Our aim was to determine whether terminal restriction fragment length polymorphism and sequence analysis of polymerase chain reaction-amplified (PCR) nosZ gene sequences could be used to show if there were measurable differences in the denitrifying community in two urban streams in catchments with contrasting degrees of catchment urbanization. Community structure in the sediments and associated riparian zones were studied at the contrasting sites. We showed that the denitrifying community in the sediments and riparian soils of the two streams were significantly different. There were also significant differences between the sediment and riparian zone communities within each of the sites. Terminal restriction fragment length polymorphism analysis proved to be a valuable technique that could resolve patterns of the denitrifying community in streams of contrasting degrees of urbanization, but sequence analysis was required to confirm the identity of the amplified products. 相似文献

The concept of a varying evolutionary tempo that is regulated by energy was first suggested in the 1950s. It was based on the observation that energy‐rich habitats appear to be the centres of evolutionary change, producing novel characters more frequently and having greater speciation rates. Subsequently, a number of studies have found positive relationships between evolutionary rate and energy. Gradients of energy occur across a range of scales and these have been invoked to explain, for example, higher tropical species richness. Precipitation has also been found to influence evolutionary rate, suggesting that biologically available energy and productivity are the important variables in the relationship rather than solar energy alone. Here, we take the theoretical step of investigating at smaller scales the implications of faster evolutionary tempo where productivity is greater: first at the level of the species population and, subsequently, for the same‐guild co‐habitants in a community. To facilitate, this we begin by applying the concept of gradients of available energy, from more productive to less productive sectors within the species niche, in the context of source‐sink theory. We then propose that a species population will have its highest rate of evolution, for changes that confer a positive selection coefficient throughout the niche, in that sector where it has best adaptive fit and greatest per capita energy flux. We also posit that, where it conferred a shift in fitness, an evolutionary change at niche optimum could subsequently affect conspecific populations occupying lower energy niche sectors via the selection‐mediated dispersal of the apomorphy into the more marginal components of the niche. We then infer that this type of change in a species with a generally higher per capita energy flux might negatively affect adjacent more slowly evolving species (living in less productive peripheral niches) in situations where the increased fitness conferred by a particular apomorphy was relevant to conditions occurring beyond the limits of the progenitor niche hypervolume. We therefore suggest a directional component to evolution at the compressed scales of niche and community level microevolution whereby populations and species occupying more productive conditions have greater tempos of change with attendant enhancements of both their competitive influence and their evolutionary potential. In this manner, previously recorded macro‐evolutionary patterns indicating faster evolution with increased energy at larger scales are interpreted in the context of proposed micro‐evolutionary relationships at intraspecific and locally interspecific scales. © 2013 The Linnean Society of London, Biological Journal of the Linnean Society, 2013, 110 , 696–714. 相似文献
