Male Broad Breasted White Turkeys (BBWT) represent an experimental model of arterial hypertension characterized by high levels of circulating and tissue catecholamines. We thought interesting to evaluate the efficacy of the long term treatment with different antihypertensive drugs. 59 male BBWT were studied, divided in five groups. The first group (13 animals) was treated with placebo; the second (14 animals) with oxprenolol 4 mg/Kg/q.d.; the third (10 animals) with labetalol 25 mg/Kg/q.d.; the fourth (11 animals) with verapamil 15 mg/Kg/ q.d.; the fifth (11 animals) with captopril 8 mg/Kg/ q.d. and furosemide 2,5 mg/Kg/q.d. All drugs were given p.o, from the 8th to the 33rd week of age. Weekly or every two weeks Blood Pressure (BP) and Heart Rate (HR) were measured by an indirect method. In all animals BP progressively increased and HR progressively decreased with age. Only the labetalol-treated animals showed a significant reduction of BP and HR through the study period as compared with the placebo-treated animals. These results confirm the preminent role played by the high levels of circulating catecholamines in determining and maintaining the arterial hypertension. 相似文献

Alkalinity of the medium was shown to be the chief factor involved in the accumulation of oxalate by T. cinnabarina. Glutamate and aspartate are shown to lead to oxalate with this organism and with L. lepideus by dehydrogenation to α-ketoglutarate and oxaloacetate, respectively. Malate was also shown to be dehydrogenated. It is proposed that oxaloacetate may either undergo β-decarboxylation to yield CO2 and pyruvate, or splitting by coenzyme A to yield oxalate and acetylated coenzyme A. The reversal of this latter reaction is suggested as the explanation of the disappearance of oxalate from culture media. The reduction of resazurin by the dehydrogenase systems of the molds is inhibited by cyanide, indicating the participation of metal systems, such as the cytochromes. 相似文献

The incidence of cystic fibrosis (CF) in Saguenay-Lac-St.-Jean, a geographically isolated region of Quebec, was estimated to be 1 in 902 during the period 1975-1988. The carrier rate was calculated to be 1 in 15 inhabitants. The high incidence of CF in Saguenay-Lac-St.-Jean is probably the result of a founder effect and genetic drift for one or more mutations. Loss of the survival motor neuron gene (SMN1) is responsible for spinal muscular atrophy (SMA), the most common inherited cause of infant mortality. Even though the SMA phenotype is traditionally considered as related to spinal motor neuron loss, it remains debated whether the specific targeting of motor neurons could represent the best therapeutic option for the disease. We here investigated, using stereological quantification methods, the spinal cord and cerebral motor cortex of ∆7 SMA mice during development, to verify extent and selectivity of motor neuron loss. We found progressive post-natal loss of spinal motor neurons, already at pre-symptomatic stages, and a higher vulnerability of motor neurons innervating proximal and axial muscles. Larger motor neurons decreased in the course of disease, either for selective loss or specific developmental impairment. We also found a selective reduction of layer V pyramidal neurons associated with layer V gliosis in the cerebral motor cortex. Our data indicate that in the ∆7 SMA model SMN loss is critical for the spinal cord, particularly for specific motor neuron pools. Neuronal loss, however, is not selective for lower motor neurons. These data further suggest that SMA pathogenesis is likely more complex than previously anticipated. The better knowledge of SMA models might be instrumental in shaping better therapeutic options for affected patients. 相似文献

Calmodulin inhibits inositol 1,4,5-trisphosphate (IP3) binding to the IP3 receptor in both a Ca2+-dependent and a Ca2+-independent way. Because there are no functional data on the modulation of the IP3-induced Ca2+ release by calmodulin at various Ca2+ concentrations, we have studied how cytosolic Ca2+ and Sr2+ interfere with the effects of calmodulin on the IP3-induced Ca2+ release in permeabilized A7r5 cells. We now report that calmodulin inhibited Ca2+ release through the IP3 receptor with an IC50 of 4.6 microM if the cytosolic Ca2+ concentration was 0.3 microM or higher. This inhibition was particularly pronounced at low IP3 concentrations. In contrast, calmodulin did not affect IP3-induced Ca2+ release if the cytosolic Ca2+ concentration was below 0.3 microM. Calmodulin also inhibited Ca2+ release through the IP3 receptor in the presence of at least 10 microM Sr2+. We conclude that cytosolic Ca2+ or Sr2+ are absolutely required for the calmodulin-induced inhibition of the IP3-induced Ca2+ release and that this dependence represents the formation of the Ca2+/calmodulin or Sr2+/calmodulin complex. 相似文献
