Cellular senescence is a state of irreversible growth arrest; however, the metabolic processes of senescent cells remain active. Our previous studies have shown that radiation induces senescence of human breast cancer cells that display low expression of securin, a protein involved in control of the metaphase–anaphase transition and anaphase onset. In this study, the protein expression profile of senescent cells was resolved by two-dimensional gel electrophoresis to investigate associated metabolic alterations. We found that radiation induced the expression and activation of glyceraldehyde-3-phosphate dehydrogenase that has an important role in glycolysis. The activity of lactate dehydrogenase A, which is involved in the conversion of pyruvate to lactate, the release of lactate and the acidification of the extracellular environment, was also induced. Inhibition of glycolysis by dichloroacetate attenuated radiation-induced senescence. In addition, radiation also induced activation of the 5′-adenosine monophosphate-activated protein kinase (AMPK) and nuclear factor kappa B (NF-κB) pathways to promote senescence. We also found that radiation increased the expression of monocarboxylate transporter 1 (MCT1) that facilitates the export of lactate into the extracellular environment. Inhibition of glycolysis or the AMPK/NF-κB signalling pathways reduced MCT1 expression and rescued the acidification of the extracellular environment. Interestingly, these metabolic-altering signalling pathways were also involved in radiation-induced invasion of the surrounding, non-irradiated breast cancer and normal endothelial cells. Positive selection driving the evolution of a gene of male reproduction, Acp26Aa, of Drosophila: II. Divergence versus polymorphism 总被引:4,自引:1,他引:4
The evolution of the gene for a male ejaculatory protein, Acp26Aa, has been
shown to be driven by positive selection when nonsibling species in the
Drosophila melanogaster subgroup are compared. To know if selection has
been operating in the recent past and to understand the details of its
dynamics, we obtained DNA sequences of Acp26Aa and the nearby Acp26Ab gene
from 39 D. melanogaster chromosomes. Together with the 10 published
sequences, we analyzed 49 sequences from five populations in four
continents. The southern African population is somewhat differentiated from
all other populations, but its nucleotide diversity is lower at these two
loci. We find the following results for Acp26Aa: (1) The R: S (replacement
: silent changes) ratio is significantly higher in the between-species
comparisons than in the within-species data by the McDonald and Kreitman
test. Positive selection is probably responsible for the excess of amino
acid replacements between species. (2) However, within-species nucleotide
diversity is high. Neither the Tajima test nor the Fu and Li test indicates
a reduction in nucleotide diversity due to positive selection in the recent
past. (3) The newly derived nucleotides in D. melanogaster are at high
frequency significantly more often than predicted by the neutral
equilibrium. Since the nearby Acp26Ab gene does not show these patterns,
these observations cannot be attributed to the characteristics of this
chromosomal region. We suggest that positive selection is active, but may
be weak, for each amino acid change in the Acp26Aa gene.

Insect flight loss is a repeated phenomenon in alpine habitats, where wing reduction is thought to enhance local recruitment and increase fecundity. One predicted consequence of flight loss is reduced dispersal ability, which should lead to population genetic differentiation and perhaps ultimately to speciation. Using a dataset of 15,123 SNP loci, we present comparative analyses of fine‐scale population structure in codistributed Zelandoperla stonefly species, across three parallel altitudinal transects in New Zealand's Rock and Pillar mountain range. We find that winged populations (altitude 200–500 m; Zelandoperla decorata) show no genetic structuring within or among streams, suggesting substantial dispersal mediated by flight. By contrast, wingless populations (Zelandoperla fenestrata; altitude 200–1100 m) exhibit distinct genetic clusters associated with each stream, and additional evidence of isolation by distance within streams. Our data support the hypothesis that wing‐loss can initiate diversification in alpine insect populations over small spatial scales. The often deep phylogenetic placement of lowland Z. fenestrata within their stream‐specific clades suggests the possibility of independent alpine colonization events for each stream. Additionally, the detection of winged, interspecific hybrid individuals raises the intriguing possibility that a previously flightless lineage could reacquire flight via introgression. 相似文献

This study aimed to evaluate anti-staphylococcal properties of crude and fractionated extracts of lactic acid bacteria (LAB) isolated from local meat, dairy, and fermented products. A total of 36 LAB isolates were obtained and identified via 16S rDNA sequencing. Cell-free supernatant (CFS) of all isolates exhibiting a statistically significant inhibition against Staphylococcus aureus (ρ < 0.05), with six LAB isolates exhibiting a more prevalent inhibition. The inhibition effects of cell wall and intracellular extracts from the six prevalent isolates were evaluated. Lactobacillus plantarum USM8613 was the most prominent isolate with both CFS and cell wall extract exhibiting the most prevalent inhibition against S. aureus. Scanning electron micrographs showed alteration of S. aureus membrane morphology upon CFS treatment, suggesting an anti-staphylococcal effect via membrane destruction. Confocal laser scanning micrographs showed inhibition against biofilm formations by S. aureus in porcine skins upon CFS treatment. The CFS from L. plantarum USM8613 was separated into protein, lipid, and polysaccharide fractions for evaluation of anti-staphylococcal activity and chemical characterization. All fractions inhibited growth of S. aureus (ρ < 0.05), with protein fractions exhibiting stronger inhibition effect. Data from our present study showed that extracts from LAB could be applied as biopreservatives in the food industries and/or as an antimicrobial agent against bacterial infections for cosmeceutical and pharmaceutical uses. 相似文献

Background
Infection with H. pylori is important in the etiology of gastric cancer. Gastric cancer is infrequent in Africa, despite high frequencies of H. pylori infection, referred to as the African enigma. Variation in environmental and host factors influencing gastric cancer risk between different populations have been reported but little is known about the biological differences between gastric cancers from different geographic locations. We aim to study genomic instability patterns of gastric cancers obtained from patients from United Kingdom (UK) and South Africa (SA), in an attempt to support the African enigma hypothesis at the biological level.Methods
DNA was isolated from 67 gastric adenocarcinomas, 33 UK patients, 9 Caucasian SA patients and 25 native SA patients. Microsatellite instability and chromosomal instability were analyzed by PCR and microarray comparative genomic hybridization, respectively. Data was analyzed by supervised univariate and multivariate analyses as well as unsupervised hierarchical cluster analysis.Results
Tumors from Caucasian and native SA patients showed significantly more microsatellite instable tumors (p < 0.05). For the microsatellite stable tumors, geographical origin of the patients correlated with cluster membership, derived from unsupervised hierarchical cluster analysis (p = 0.001). Several chromosomal alterations showed significantly different frequencies in tumors from UK patients and native SA patients, but not between UK and Caucasian SA patients and between native and Caucasian SA patients.Conclusions
Gastric cancers from SA and UK patients show differences in genetic instability patterns, indicating possible different biological mechanisms in patients from different geographical origin. This is of future clinical relevance for stratification of gastric cancer therapy.100.
