The ecophysiology of cold tolerance in many terrestrial invertebrate animals is based on water and its activity at low temperatures, affecting cell, tissue and whole organism functions. The normal body water content of invertebrates varies from 40 to 90% of their live weight, which is influenced by water in their immediate environment, especially in species with a water vapour permeable cuticle. Water gain from, or loss to, the surrounding atmosphere may affect animal survival, but under sub-zero conditions body water status becomes more critical for overwinter survival in many species. Water content influences the supercooling capacity of many insects and other arthropods. Trehalose is known to maintain membrane integrity during desiccation stress in several taxa. Dehydration affects potential ice nucleators by reducing or masking their activity and a desiccation protection strategy has been detected in some species. When water crystallises to ice in an animal it greatly influences the physiology of nearby cells, even if the cells remain unfrozen. A proportion of body water remains unfrozen in many cold hardened invertebrates when they are frozen, which allows basal metabolism to continue at a low level and aids recovery to normal function when thawing occurs. About 22% of total body water remains unfrozen from calculations using differential scanning calorimetry (compared with ca 19% in food materials). The ratio of unfrozen to frozen water components in insects is 1:4 (1:6 for foods). Such unfrozen water may aid recovery of freezing tolerant species after a freezing exposure. Rapid changes in cold hardiness of some arthropods may be brought about by subtle shifts in body water management. It is recognised that cold tolerance strategies of many invertebrates are related to desiccation resistance, and possibly to mechanisms inherent in insect diapause, but the role of water is fundamental to them all. Detailed experimental studies are needed to provide information which will allow a more complete and coherent understanding of the behaviour of water in biological systems and aid the cryopreservation of a wide range of biological material.  相似文献

Osteoclasts are specialized cells derived from the monocyte/macrophage haematopoietic lineage that develop and adhere to bone matrix, then secrete acid and lytic enzymes that degrade it in a specialized, extracellular compartment. Discovery of the RANK signalling pathway in the osteoclast has provided insight into the mechanisms of osteoclastogenesis and activation of bone resorption, and how hormonal signals impact bone structure and mass. Further study of this pathway is providing the molecular basis for developing therapeutics to treat osteoporosis and other diseases of bone loss.  相似文献

F-box proteins are members of a large family that regulates the cell cycle, the immune response, signalling cascades and developmental programmes by targeting proteins, such as cyclins, cyclin-dependent kinase inhibitors, IkappaBalpha and beta-catenin, for ubiquitination (reviewed in refs 1-3). F-box proteins are the substrate-recognition components of SCF (Skp1-Cullin-F-box protein) ubiquitin-protein ligases. They bind the SCF constant catalytic core by means of the F-box motif interacting with Skp1, and they bind substrates through their variable protein-protein interaction domains. The large number of F-box proteins is thought to allow ubiquitination of numerous, diverse substrates. Most organisms have several Skp1 family members, but the function of these Skp1 homologues and the rules of recognition between different F-box and Skp1 proteins remain unknown. Here we describe the crystal structure of the human F-box protein Skp2 bound to Skp1. Skp1 recruits the F-box protein through a bipartite interface involving both the F-box and the substrate-recognition domain. The structure raises the possibility that different Skp1 family members evolved to function with different subsets of F-box proteins, and suggests that the F-box protein may not only recruit substrate, but may also position it optimally for the ubiquitination reaction.  相似文献

A reinvestigation of the type specimen of Araucarioxylon? obscurum Knowlton (1900) from the Upper Jurassic Morrison Formation in the Freezeout Hills of Wyoming has determined that this fossil should be transferred to Mesembrioxylon Seward (1919) as M. obscurum comb. nov. (Knowlton) Medlyn and Tidwell. Knowlton (1900), who was uncertain of its appropriate generic disposition, tentatively referred the wood to Araucarioxylon because it had obscure growth rings and relatively low ray height. Reexamination of Knowltons slides demonstrates that the wood has uniseriate, rarely biseriate rays, podocarpoid pitting, and diffuse axial parenchyma, none of which Knowlton mentioned. Tracheary pitting is mostly separate, round, uniseriate, occasionally biseriate and, when biseriate, opposite to subopposite. The smooth-walled crossfields exhibit 1-3 thin-bordered podocarpaceous pits per field. All of these features are present in Mesembrioxylon Seward.  相似文献

Cover and species richness of vascular and cryptogamic components of the plant community were inventoried in experimental grazing paddocks at the USDA/FS Desert Range Experimental Station, Millard County, Utah. The grazing treatments considered have been applied continuously for over 50 years. The effects of heavy (ca 17 sheep days/acre) grazing treatment applied in two different seasons (early winter versus a split between early and late winter) differed significantly between seasons. Cryptogamic cover and cryptogamic species richness both showed larger decreases under early-late as opposed to early winter only grazing. Vascular plant cover (relative to controls) was also reduced by early-late winter grazing, but not to a significant degree. Late season grazing, likewise, had no significant effect on number of vascular species per transect.  相似文献
