Experimental Study on Allogenic Decalcified Bone Matrix as Carrier for Bone Tissue Engineering

The biocompatibility and osteogenic activity of allogenic decalcified bone matrix (DBM) used as a carrier for bone tissue engineering were studied. Following the method described by Urist, allogenic DBM was made. In vitro, DBM and bone marrow stromal cell (BMSC) from rab-bits were co-cultured for 3-7 days and subjected to HE staining, and a series of histomorphological observations were performed under phase-contrast microscopy and scanning electron microscopy (SEM). In vivo the mixture of DBM/BMSC co-cultured for 3 days was planted into one side of muscules sacrospinalis of rabbits, and the DBM without BMSC was planted into other side as con-trol. Specimens were collected at postoperative week 1, 2 and 4, and subjected to HE staining, and observed under SEM. The results showed during culture in vitro, the BMSCs adherent to the wall of DBM grew, proliferated and had secretive activity. The in vivo experiment revealed that BMSCs and undifferentiated mesenchymal cells in the perivascular region invaded gradually and proliferated together in DBM/BMSC group, and colony-forming units of chondrocytes were found. Osteoblasts,trabecular bone and medullary cavity appeared. The inflammatory reaction around muscles almost disappeared at the second weeks. In pure DBM group, the similar changes appeared from the sur-face of the DBM to center, and the volume of total regenerate bones was less than the DBM/BMSC group at the same time. The results indicated that the mixture of DBM and BMSC had good bio-compatibility and ectopic induced osteogentic activty.     

An Integrated Technique for Identification of Differential Genes Expressed in Patients with Cancer

Summary: To develop a method for identification of differential gene expression between different cell populations, several convenient techniques of molecular biology, including subtractive hybridization, suppression PCR, T/A cloning and sequencing, were used to identify genes expressed differentially in CD45^- and CD45^- cells isolated from U266 cell line of multiple myeloma. Our results showed that the levels of abundant genes scale down 20 times through subtractive hybridization.Plasmid DNA from CD45^- cell clones was hybridized with forward or backward cDNA probes synthesized from CD45^- and CD45 cells, respectively. A few of differentially expressed genes reconfirmed by RT-PCR were identified from 500 expressed clones of CD45^- cells. It is concluded that a strategy for gene expression identification developed from conventional molecular biological methods can be used in different laboratories.     

国产前列腺内支架的临床应用及并发症探讨

目的探讨国产前列腺内支架治疗前列腺增生引起排尿困难的疗效及并发症.方法42例前列腺增生引起的排尿困难的患者,置入46枚前列腺内支架.支架均为国产镍钛记忆合金编织而成.结果42例患者中,36例置入支架后立即自行排尿,5例因前列腺增生过大前列腺尿道部过长或支架位置不理想,置入一个支架,排尿仍有困难,再次置入第二个支架后,立即自行排尿,但2例出现长期的尿失禁;1例伴有糖尿病的患者,置入支架1周后排尿困难未改善,而进行外科手术治疗.36例生活质量积分0～3分,29例术后随访6～21个月,国际前列腺症状评分(I-PSS)术前27.49±4.21,术后6.89±3.76(P<0.001),最大尿流率术前(1.78±3.89)ml/s,术后(13.91±3.56)ml/s(P<0.001).结论国产前列腺支架治疗前列腺增生引起的排尿困难虽然会出现一些并发症,但仍是一种安全、可靠的方法.     

SA、F/TPSA及PSAD在前列腺癌诊断中的作用

目的 :探讨血清前列腺特异抗原 (PSA) ,血清总PSA及游离PSA比值 (F/TPSA)及前列腺特异性抗原密度(PSAD)在前列腺癌诊断中的作用。方法 :对 5 1例前列腺癌患者及 14 5例良性前列腺增生症患者PSA、F/TPSA及PSAD值的差异进行分析、比较。结果 :前列腺癌组血清PSA及PSAD高于良性前列腺增生组 ;而F/TPSA值低于良性前列腺增生组 ,差异均有显著性。结论 :PSA >4ng·ml-1作为筛选前列腺癌的临界值存在一定缺陷 ;当PSA <10ng·ml-1,F/T值有助于鉴别前列腺癌和良性前列腺增生 ;而PSAD对于筛选前列腺活检病例亦有一定价值。     

Ⅰ、Ⅲ型前胶原基因第2外显子核酶对靶RNA的体外切割活性的研究

目的 体外研究锤头型α1Ⅰ型及Ⅲ型前胶原基因第2外显子片段核酶对各自靶RNA分子的切割活性及反应条件。同时观察两反义核酶对瘢痕中成纤维细胞胶原合成的影响。方法 将含α1Ⅰ型及Ⅲ型前胶原基因第2外显子片段的重组质粒(pT-Ⅰ、pT-Ⅲ)，经体外^32P标记转录后形成产物靶RNA。同时将含特异性核酶基因的重组质粒(pT-gⅠ、pT-gⅢ)进行非标记的体外转录，产物(核酶)与各自的^32P-靶RNA按不同的条件混和反应，电泳后放射自显影观察结果。将构建好的核酶以脂质体包裹后导入培养的成纤维细胞内，采用图像分析法观察核酶对成纤维细胞Ⅰ型、Ⅲ型胶原蛋白mRNA合成的影响。结果 两种核酶在37℃、42℃均能有效切割各自的靶RNA，对Mg^2 浓度的要求范围较宽(10～20mmol／L)；反应温度从65℃逐渐降至并维持在37℃的条件下核酶切割活性显提高。Ⅰ、Ⅲ型胶原蛋白mRNA的表达明显降低，胶原蛋白生成降低，胶原生成明显受抑制。结论 针对α1Ⅰ型及Ⅲ型前胶原基因第2外显子片段的核酶能有效地在体外对靶RNA进行切割并能有效地抑制瘢痕中成纤维细胞胶原的合成。     

流行性出血热82例误诊分析

目的 :探讨流行性出血热 (EHF)误诊情况 ,提高临床医师对本病的早期诊断水平。方法 :将我院 1982年 1月～ 1991年 12月和 1992年 1月～ 2 0 0 1年 12月各 10年收治的出血热和误诊情况进行回顾性分析。结果 :前 10年共收治EHF 4 82例 ,其中误诊 6 0例 ,后 10年收治 5 2例 ,误诊 2 2例 ,误诊率分别为 12 %和 4 3% ,而误诊疾病竟达2 2种之多。结论 :在任何季节里对有发热、出血的病人我们应高度警惕有可能是EHF ,及时作相关检查。     

脂肪抽吸法副乳腺矫治术

目的：介绍将脂肪抽吸术应用于矫治副乳腺的8例临床实践经验。方法：根据患者不同情况分别采用单纯脂肪抽吸和脂肪抽吸 皮肤切除术两种手术方法治疗副乳腺。结果：所有患者完全吸除副乳腺及局部增生的脂肪组织，效果良好，无并发症发生。结论：脂肪抽吸法不仅可以应用小切口微创治疗副乳腺，而且很好的解决了副乳腺局部脂肪组织增生的问题，提供了副乳腺矫治的一种较好的新方法。
