Methane is an end product of anaerobic degradation of organic compounds in fresh water environments such as lake sediments and the intestinal tract of animals. Methanogenic archaea produce methane from carbon dioxide and molecular hydrogen, acetate and C1 compounds such as methanol in an energy gaining process. The methanogenic pathway utilizing carbon dioxide and molecular hydrogen involves ten methanogen specific enzymes, which catalyze unique reactions using novel coenzymes. These enzymes have been purified and biochemically characterized. The genes encoding the enzymes have been cloned and sequenced. Recently, crystal structures of five methanogenic enzymes: formylmethanofuran : tetrahydromethanopterin formyltransferase, methenyltetrahydromethanopterin cyclohydrolase, methylenetetrahydromethanopterin reductase, F420H2:NADP oxidoreductase and methyl-coenzyme M reductase were reported. In this review, we describe the pathway utilizing carbon dioxide and molecular hydrogen and the catalytic mechanisms of the enzymes based on their crystal structures.  相似文献

Bangladesh is a potential site of implementing renewable energy system to reduce the severe power crisis throughout the year. According to this, Chittagong is the southeastern part of Bangladesh is also a potential site for implementing renewable energy system such as grid-connected photovoltaic (PV) system. Financial viability and green-house gas emission reduction of solar PV as an electricity generation source are assessed for 500 kW grid connected solar PV system at University of Chittagong, Chittagong. Homer simulation software and monthly average solar radiation data from NASA is used for this task. In the proposed system monthly electricity generation varies between 82.65 MW h and 60.3 MW h throughout the year with a mean value of 68.25 MW h depending on the monthly highest and lowest solar radiation data. It is found that per unit electricity production cost is US$ 0.20 based on project lifetime 25 years. The IRR, equity payback and benefit-cost ratio shows favorable condition for development of the proposed solar PV system in this site. A minimum 664 tones of green-house gas emissions can be reduced annually utilizing the proposed system.  相似文献

By sulfurization of E---B evaporated precursors, CZTS(Cu2ZnSnS4) films could be prepared successfully. This semiconductor does not consist of any rare-metal such as In. The X-ray diffraction pattern of CZTS thin films showed that these films had a stannite structure. This study estimated the optical band gap energy as 1.45 eV. The optical absorption coefficient was in the order of 104cm−1. The resistivity was in the the order of 104 Ω cm and the conduction type was p-type. Fabricated solar cells, Al/ZnO/CdS/CZTS/Mo/Soda Lime Glass, showed an open-circuit voltage up to 400 mV.  相似文献

A simple magnetic circuit of salient-pole synchronous machines is presented. A method for calculating steady-state electrical performances using the circuit is described. Magnetic saturation including the cross-magnetizing phenomenon is considered in the method. The saturated reactances, field currents, and load angles are calculated easily from the method without much computational effort as compared with the finite element method. Despite the simplicity of the magnetic circuit, the calculated reactances at various load conditions using the method are close to the measured and FEM values.  相似文献

In this work we present the first magnetoresistance measurements on multilayered vertical Co(~6?nm)/Cu(~6?nm) and slanted Co(x?nm)/Cu(x?nm) (with x≈6, 11, and 16?nm) nanocolumns grown by oblique angle vapour deposition. The measurements are performed at room temperature on the as-deposited nanocolumn samples using a scanning tunnelling microscope to establish electronic contact with a small number of nanocolumns while an electromagnet generates a time varying (0.1?Hz) magnetic field in the plane of the substrate. The samples show a giant magnetoresistance (GMR) response ranging from 0.2 to 2%, with the higher GMR values observed for the thinner layers. For the slanted nanocolumns, we observed anisotropy in the GMR with respect to the relative orientation (parallel or perpendicular) between the incident vapour flux and the magnetic field applied in the substrate plane. We explain the anisotropy by noting that the column axis is the magnetic easy axis, so the magnetization reversal occurs more easily when the magnetic field is applied along the incident flux direction (i.e., nearly along the column axis) than when the field is applied perpendicular to the incident flux direction.  相似文献

Effects of oil-droplet size, the weight ratio of oil to wall material and the storage temperature on the oxidation of methyl linoleate microencapsulated with maltodextrin by spray-drying were examined. The oxidation of methyl linoleate was more retarded for the microcapsules prepared from the emulsion having smaller oil droplets. The oxidation was more suppressed for the microcapsules having a lower weight ratio of oil to wall material. The fraction of unoxidized methyl linoleate leveled off after 10- to 15-days storage. The level, Y(infinity), depended on the weight ratio. The dependence of Y(infinity) on the weight ratio was analyzed based on the percolation theory, and the three-dimensional model of the theory was suitable to express the dependence. The effect of the storage temperature on the oxidation of microencapsulated methyl linoleate was also examined, and the activation energy was evaluated. The value of the energy suggested that the oxidation itself was a rate-limiting step for the oxidation of methyl linoleate encapsulated with maltodextrin.  相似文献

This study investigated how ferulic acid (FA) affects the formation of certain Maillard reaction products (MRPs), i.e., early MRPs, fluorescent and non-fluorescent advanced glycation end products (AGEs), and melanoidins in model systems. Glycation mixtures were prepared containing soy glycinin or bovine serum albumin (final concentration 10 mg/ml) and fructose (222 mM) in 0.2% KOH in the presence or absence of FA (12.95 mM) and incubated at 60 °C for 60 min. The extent of the MR was estimated by analysis of free amino groups, the incorporation of sugar into the protein backbone as well as the formation of N?-(carboxymethyl)lysine (CML), fluorescent AGEs (λexc = 337 nm, λem = 350–550 nm) and melanoidins (absorbance at 420 nm). Formation of CML and fluorescent AGEs was reduced by nearly 90% by the addition of FA while early MRPs and melanoidins were inhibited to a lesser extent (∼10% and 28%, respectively) compared to AGE formation. A controlled formation of early MRPs was achieved by use of FA, and it is a new finding. To the best of our knowledge, for the first time the use of FA as a reliable means of obtaining novel glycoprotein preparations containing low amounts of AGEs, with the potential to be used as functional food ingredients, is proposed.  相似文献

Crystal growth and magnetic behavior of iron oxide nanoparticles assembled with biomolecules have been investigated. The nanoparticles assembled with trypsin molecules exhibit superparamagnetism at room temperature with blocking temperature ($sim$80 K) significantly lower than those without trypsin ($sim$140 K). This is attributed to reduced magnetostatic couplings between particles due to increased distance between particles separated by trypsin molecules. Moreover, the synthesized nanoparticle–biomolecule assemblies consist of a unique one-dimensional self-assembled arrays of nanoparticles found by structural analysis using transmission electron microscopy. The moirÉ fringes observed from the particle arrays indicate that the particles are aligned with slight misorientation of their crystallographic axes. Such an unusual formation of nanoparticle arrays may be relevant to specific ligand sites in trypsin molecules and the magnetostatic interparticle couplings.  相似文献

In the modern baking industry, high-sucrose-tolerant (HS) and maltose-utilizing (LS) yeast were developed using breeding techniques and are now used commercially. Sugar utilization and high-sucrose tolerance differ significantly between HS and LS yeasts. We analysed the gene expression profiles of HS and LS yeasts under different sucrose conditions in order to determine their basic physiology. Two-way hierarchical clustering was performed to obtain the overall patterns of gene expression. The clustering clearly showed that the gene expression patterns of LS yeast differed from those of HS yeast. Quality threshold clustering was used to identify the gene clusters containing upregulated genes (cluster 1) and downregulated genes (cluster 2) under high-sucrose conditions. Clusters 1 and 2 contained numerous genes involved in carbon and nitrogen metabolism, respectively. The expression level of the genes involved in the metabolism of glycerol and trehalose, which are known to be osmoprotectants, in LS yeast was higher than that in HS yeast under sucrose concentrations of 5-40%. No clear correlation was found between the expression level of the genes involved in the biosynthesis of the osmoprotectants and the intracellular contents of the osmoprotectants. The present gene expression data were compared with data previously reported in a comprehensive analysis of a gene deletion strain collection. Welch's t-test for this comparison showed that the relative growth rates of the deletion strains whose deletion occurred in genes belonging to cluster 1 were significantly higher than the average growth rates of all deletion strains.  相似文献
