Aspiration biopsy of tumors and transplanted organs is a popular atraumatic method for diagnosis and also for the management of patients. Apart from the cytological analyses, it is clear that large aggregates of cells are obtained and these may be examined by electron microscopy. A study was carried out with dogs to assess the yield of intact glomeruli and tubules, resulting from a standard fine-needle aspiration procedure. Employing a 20-gauge needle, yields of approximately ten intact glomeruli were obtained with a tissue processing protocol that utilized filtration through a nylon mesh and backwashing, to resuspend the trapped aggregates. Processing for electron microscopy was carried out in microcentrifuge tubes. The ultrastructural preservation was excellent and the extension of these studies to investigations of not only renal transplantation, but also for diagnosis in human renal diseases of a broad spectrum, is suggested.  相似文献

Tyrosine hydroxylase (TyrOH) catalyzes the conversion of tyrosine to L-DOPA, the rate-limiting step in the biosynthesis of the catecholamines dopamine, adrenaline, and noradrenaline. TyrOH is highly homologous in terms of both protein sequence and catalytic mechanism to phenylalanine hydroxylase (PheOH) and tryptophan hydroxylase (TrpOH). The crystal structure of the catalytic and tetramerization domains of TyrOH reveals a novel alpha-helical basket holding the catalytic iron and a 40 A long anti-parallel coiled coil which forms the core of the tetramer. The catalytic iron is located 10 A below the enzyme surface in a 17 A deep active site pocket and is coordinated by the conserved residues His 331, His 336 and Glu 376. The structure provides a rationale for the effect of point mutations in TyrOH that cause L-DOPA responsive parkinsonism and Segawa's syndrome. The location of 112 different point mutations in PheOH that lead to phenylketonuria (PKU) are predicted based on the TyrOH structure.  相似文献

The surface and interface properties of Pd0.9Cr0.1/SiC Schottky diode gas sensors both before and after annealing are investigated using Auger electron spectroscopy (AES), scanning electron microscopy (SEM), and energy dispersive spectroscopy (EDS). At room temperature the alloy reacted with SiC and formed PdxSi only in a very narrow interfacial region. After annealing for 250 h at 425°C, the surface of the Schottky contact area has much less silicon and carbon contamination than that found on the surface of an annealed Pd/SiC structure. Palladium silicides (PdxSi) formed at a broadened interface after annealing, but a significant layer of alloy film is still free of silicon and carbon. The chromium concentration with respect to palladium is quite uniform down to the deep interface region. A stable catalytic surface and a clean layer of Pd0.9Cr0.1 film are likely responsible for significantly improved device sensitivity.  相似文献

The 2-process theory of semantic priming (J. H. Neely, 1977; M. I. Posner and C. R. Snyder, 1975) was used to determine the maintenance of automatic processes after severe closed head injury (CHI) and to determine whether processes that demand attention suffer a deficit. Ss with severe CHI (N?=?18,?>?2 yrs postinjury) and 18 matched control Ss completed a lexical decision task in which a category prime was followed by a target. Automatic and attentional priming were determined by orthogonally varying prime–target relatedness, expectancy, and stimulus onset asynchrony. Although the CHI Ss had slower reaction times (RTs) overall, there were no significant group differences in the magnitude of either the automatic or attentional component of semantic priming. The present results indicate the integrity of semantic processes and normal semantic priming in long-term patients with severe CHI. Abstract: Facility location and customer allocation problems are pervasive in industry and in the private sector. In this paper, we outline a new approach to solving these problems that avoids the need to resort to integer programming techniques. The approach exploits the mathematical structure of the problem as well as characteristics frequently found in applied location/allocation problems. Using this approach, large scale problems are readily solvable on personal computers. We illustrate the approach on a small example and provide computational results based on a real-world application of the technique.
