Multicopper oxidases (MCOs) are encoded in the genomes of Eukarya, Bacteria, and Archea. These proteins are unique in that they contain at least four Cu atom prosthetic groups organized into one each of the three spectral classifications of copper sites in biology: type 1 (T1), type 2 (T2), and binuclear type 3 (T3), where the T2 and T3 sites form a trinuclear Cu cluster. With these four redox-active copper sites, the multicopper oxidases catalyze the four-electron (4e(-)) reduction of dioxygen to 2H2O, an activity that they alone share with the terminal heme-containing oxidases. Most MCOs exhibit broad specificity towards organic reductants, while a relatively small number of family members exhibit equally robust activity towards metal ions like Fe(II), Cu(I), and Mn(II) and, thus, are considered metallo-oxidases. This Account analyzes the structure-activity features of multicopper oxidases that determine their relative substrate specificity. Since the substrate oxidation step involves an outer-sphere electron transfer from the reductant to the T1Cu site in the protein, the concepts of Marcus theory are applied to unravel the origin of the substrate specificity of the multicopper ferroxidases.  相似文献

Coho salmon (Oncorhynchus kisutch) is a fatty fish species whose farming production has greatly increased in recent years. Lipid damage produced during Coho salmon chilled storage was studied for up to 24 d. Lipid hydrolysis (free fatty acids, FFA) and oxidation (conjugated dienes; peroxide value, PV; thiobarbituric acid index, TBA‐i; fluorescent compounds formation, FR; browning development) were determined and compared to lipid composition (polyene index, PI; astaxanthin, AX) changes and sensory assessment (rancid odour development) results. Most lipid damage indices developed slowly during storage; thus, values obtained for FFA, PV, TBA‐i and FR were in all cases under 1.5 g/100 g, 4.0 meq oxygen/kg lipid, 0.40 mg malondialdehyde/kg muscle and 0.40, respectively. Odour assessment showed a significant (p <0.05) rancidity development at day 10, when compared to starting fish material; then, non‐acceptable values were obtained at days 19 and 24. The PI analysis showed not many differences during the storage time, with the lowest mean value at day 19. AX analysis indicated a relatively high content in the white muscle, which was maintained till the end of the experiment. A low oxidation development is concluded for Coho salmon lipids when compared to other fatty fish species under the same chilling conditions. AX was found to contribute to the oxidation stability of Coho salmon lipids, due to its free radical scavenger properties.  相似文献

Brazilian redspotted shrimp (Farfantepenaeus paulensis) waste is an important source of carotenoids such as astaxanthin and lipids with a high ω−3 fatty acids content, mainly docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). In order to establish an efficient and environmental friendly recovery process, the lipids and astaxanthin were extracted from the freeze-dried redspotted shrimp waste (including head, tail and shell) using supercritical carbon dioxide. The effects of the extraction conditions of pressure (200-400 bar) and temperature (40-60 °C) on the global yield (X0), astaxanthin extraction yield and astaxanthin concentration in the extract were evaluated. It was found that the pressure and temperature showed a very low significant effect on the lipid extraction yield using supercritical CO2. In comparison with lipid extraction by solvents, maximum efficiency of supercritical fluid extraction achieved 64% of hexane extraction yield. On the other hand, temperature and pressure had significant effects on astaxanthin extraction yield. Thegreatest amount of extract was obtained at 43 °C and 370 bar, with 39% of recovery.  相似文献

The purpose of this paper is to study the thermal and mechanical behaviour in machining of aluminium alloys (Al 7075-0) using PCD (polycrystalline diamond) and K10 (cemented carbide) tools and to make a comparison between the performances of both tools. The study was made using a commercial finite element software. This software has a user friendly interface and can output several results including cutting forces, temperature, pressure, von Mises stress, maximum shear stress, plastic strain, and plastic strain rate which were the objectives of this study. By analysing the simulations, it was concluded that the polycrystalline tool has a superior performance in terms of cutting and feed forces and temperature when compared to the cemented carbide tool.  相似文献

We consider the problem of scheduling on uniform machines which may not start processing at the same time with the purpose of minimizing the maximum completion time. We propose using a variant of the MULTIFIT algorithm, LMULTIFIT, which generates schedules which end within 1.382 times the optimal maximum completion time for the general problem, and within \(\sqrt{6}/2\) times the optimal maximum completion time for problem instances with two machines. Both developments represent improvements over previous results. We prove that LMULTIFIT worst-case bounds for scheduling on simultaneous uniform machines are also LMULTIFIT worst-case approximation bounds for scheduling on nonsimultaneous uniform machines and show that worst-case approximation bounds of MULTIFIT variants for simultaneous uniform machines from previous literature also apply to LMULTIFIT. We also comment on how a PTAS for scheduling on a constant number of uniform machines with fixed jobs can be used to obtain a PTAS for scheduling on a constant number of uniform nonsimultaneous parallel machines.  相似文献

Activated carbon honeycomb-monoliths with different textural properties were prepared by chemical activation of African palm shells with H(3)PO(4), ZnCl(2) and CaCl(2) aqueous solutions of various concentrations. The adsorbents obtained were characterized by N(2) adsorption at 77 K, and their carbon dioxide adsorption capacities were measured at 273 K and 1 Bar in volumetric adsorption equipment. The experimental adsorption isotherms were fitted to Langmuir and Tóth models, and a better fit was observed to Tóth equation with a correlation coefficient of 0.999. The maximum experimental values for adsorption capacity at the highest pressure (2.627-5.756 mmol·g(-1)) are between the calculated data in the two models.  相似文献

The effect of highly hydrophobic emulsifiers, the palmitic sucrose ester P‐170 (hydrophilic/lipophilic balance (HLB) = 1.0), the stearic sucrose ester S‐170 (HLB = 1.0), the polyglycerol ester decaglycerol decastearate DAS 7S (HLB = 3.7) and the polyglycerol ester decaglycerol dodecabehenate DDB 750 (HLB = 2.6), on the nucleation of a high melting point milk fat fraction (HMF) and its blends with sunflower oil (SFO) was investigated by polarized laser light turbidimetry, X‐ray diffractometry and polarized light microscopy (PLM). Addition of polyglycerol esters accelerated nucleation, giving shorter induction times for the same supercooling. On the contrary, sucrose esters inhibited nucleation since induction times were elongated in all conditions selected. Addition of emulsifiers modified the polymorphic behavior in the blends with SFO. The β' form was promoted especially with the addition of S‐170. DAS 7S and DDB 750 promoted crystallization. PLM images showed many small crystals that did not appear in HMF images. Addition of P‐170 and S‐170 delayed nucleation and inhibited crystal growth. Crystals were notoriously smaller than the ones that appeared in HMF images. The Fisher–Turnbull model was used to calculate activation free energies of nucleation. In all cases, sucrose esters elevated the energy barrier for nucleation. Polyglycerol esters, however, if they had an effect on the energy barrier, lowered the values.  相似文献

Photocyclovoltammetric experiments with films of poly(3-methylthiophene) under polychromatic light irradiation show a p-type semiconductor behavior in the reduced state, with a flat band potential of 0.18 V. The photocurrent depends on film thickness and surface morphology. Films with different thickness were characterized by scanning electron microscopy, indicating a globular morphology with globule sizes changing according to the charge density used during polymer deposition. The highest photocurrent is observed for the lowest globule diameter. These films were also irradiated with monochromatic light from the electrolyte side and from the substrate side. A higher photocurrent and a photocurrent spectrum matching the absorption spectrum of the reduced form of the polymer is observed on irradiation from the electrolyte side of a 0.7 μm thick film. In contrast, for irradiation from the electrode side, the photocurrent is lower and the spectrum shows a peak at lower energy. These results were interpreted in terms of a delocalized space charge zone, different kinetics for charge transfer and mass transport across the solvent swollen polymer film and different depth of penetration of the light as a function of wavelength.  相似文献

Some diamond tools use iron in their composition, and it is known that iron is a strong catalyst for the graphitization of diamonds. This graphitization occurs mainly during the processing of composite materials – conventional sintering or hot pressing, and during cutting operations. This work studies the effect of TiC coating on diamond, on structure, microstructure and mechanical properties of the processed composites. Samples were prepared by mixing powders of Fe (?40 μm) and diamond (?425 μm), and subsequent hot pressing at 35 MPa/900 °C, during 3 minutes. Microstructural aspects were observed by scanning electron microscopy (SEM) and iron diffusion on diamond was studied by energy dispersive spectrometry (EDS). Structural analysis was performed by X‐ray diffraction and Raman spectroscopy. Compressive tests were carried out, as well as the wear resistance of the diamond composites. The importance of employing coated diamonds is stablished. It was observed that iron has not activated the graphitization of diamond crystals.  相似文献
