The performance of the ISO method for the ‘Enumeration of the Specially Thermoresistant Spores of Thermophilic Bacteria in Dried Milk’ (ISO/TS27265; 2009) was compared with a more practical method. Both were tested for predictability of spoilage of UHT treated reconstituted milk. The data show that heating for 30 min at 100 °C has the same predictive value as heating for 30 min at 106 °C, provided that specifications are increased 1 log10 and the use of TSA as a cultivation medium is recommended over PCMA. Predictability of spoilage using classical plating methods is furthermore discussed in relation to variation in spore heat resistance of spores commonly present in the dairy industry.  相似文献

For the enumeration of Listeria monocytogenes in cold-smoked salmon, a sensitive enumeration method, based on membrane filtration followed by transfer of the filter on a selective medium has been recently developed (Gnanou Besse et al., 2004, A contribution to the improvement of L. monocytogenes enumeration in cold-smoked salmon. International Journal of Food Microbiology, 91, 119-127). The aim of the study was to assess the performance of this enumeration method through an inter-laboratory study, using cold-smoked salmon artificially contaminated at 2 different levels (approximately 0.6 and 1.6 log10 CFU g(-1)). A reproducibility standard deviation of 0.23 log10 CFU g(-1)and 0.15 log10 CFU g(-1) was obtained for the method respectively at the lower level and the higher level. Under certain conditions, the uncertainty of measurement can be derived from the method reproducibility standard deviation and was calculated to be 0.46 log10 CFU g(-1) for the lower contamination level and 0.30 log10 CFU g(-1) for the higher contamination level. These values can be considered as satisfactory for such low contamination levels.  相似文献

A maximum likelihood framework for the probabilistic assessment of cyclically induced reconsolidation settlements of saturated cohesionless soil sites is described. For this purpose, over 200 case history sites were carefully studied. After screening for data quality and completeness, the resulting database is composed of 49 high-quality, cyclically induced ground settlement case histories from seven different earthquakes. For these case history sites, settlement predictions by currently available methods of Tokimatsu and Seed (1984), Ishihara and Yoshimine (1992), Shamoto et al. (1998), and Wu and Seed (2004) are presented comparatively, along with the predictions of the proposed probabilistic model. As an integral part of the proposed model, the volumetric strain correlation presented in the companion paper is used. The accuracy of the mean predictions as well as their uncertainty is assessed by both linear regression and maximum likelihood methodologies. The analyses results revealed that (1) the predictions of Shamoto et al. and Tokimatsu and Seed are smaller than the actual settlements and need to be calibrated by a factor of 1.93 and 1.45, respectively; and (2) Ishihara and Yoshimine, and Wu and Seed predictions are higher than the actual settlements and need to be calibrated by a factor of 0.90 and 0.98, respectively. The Wu and Seed procedure produced the most unbiased estimates of mean settlements [i.e., their calibration coefficient (0.98) is the closest to unity], but the uncertainty (scatter) of their predictions remains high as revealed by the second to last smaller R2 value, or relatively higher standard deviation (σε) of the model error. In addition to superior model predictions, the main advantage of the proposed methodology is the probabilistic nature of the calibration scheme, which enables incorporation of the model uncertainty into mean settlement predictions. To illustrate the potential use of the proposed model, the probability of cyclically induced reconsolidation settlement of a site after a scenario earthquake to be less than a threshold settlement level is assessed.  相似文献

Beta thalassemia major (βT) is a hereditary anemia characterized by transfusion-dependency, lifelong requirement of chelation, and organ dysfunction. MicroRNA (miRNA) can be packed into extracellular vesicles (EVs) that carry them to target cells. We explored EV-miRNA in βT and their pathophysiologic role. Circulating EVs were isolated from 35 βT-patients and 15 controls. EV miRNA was evaluated by nano-string technology and real-time quantitative polymerase chain reaction (RT-qPCR). We explored effects of EVs on cell culture proliferation, apoptosis, and signal transduction. Higher amounts of small EV (exosomes) were found in patients than in controls. The expression of 21 miRNA was > two-fold higher, and of 17 miRNA < three-fold lower in βT-EVs than control-EVs. RT-qPCR confirmed differential expression of six miRNAs in βT, particularly miR-144-3p, a regulator of erythropoiesis. Exposure of endothelial, liver Huh7, and pancreatic 1.1B4 cells to βT-EVs significantly reduced cell viability and increased cell apoptosis. βT-EV-induced endothelial cell apoptosis involved the MAPK/JNK signal-transduction pathway. In contrast, splenectomized βT-EVs induced proliferation of bone marrow mesenchymal stem cells (BM-MSC). In summary, the miR-144-3p was strongly increased; βT-EVs induced apoptosis and decreased endothelial, pancreatic, and liver cell survival while supporting BM-MSC proliferation. These mechanisms may contribute to βT organ dysfunction and complications.  相似文献

A number of investigations have demonstrated that zeolite NaA could be synthesized using Si, extracted from rice husk ash; however, experiments on direct extraction of Si from rice husk (RH) are scarce. The main objective of the present study was to explore the possibility to synthesize high-quality zeolite NaA from RH and waste aluminium cans (as a source of Al), applying different procedures for the preparation of initial hydrogel and a unified procedure for crystallization of zeolite NaA. Products were characterized by SEM–EDX and XRD analyses. The investigation demonstrated that Si could be extracted directly from RH, avoiding the process of RH burning. Practically complete dissolution of Si from RH was achieved by alkali treatment (with 10 % NaOH for 7 h) at boiling temperature and atmospheric pressure, i.e. using refluxing system instead of autoclave for the preparation of Si-gel. Zeolite NaA samples synthesized from such Si-gels were pure, highly crystalline and white. Furthermore, it was found that the direct dissolution of Al in Si-gel did not affect the quality of the final product. Although this investigation was not focused on the mechanism of zeolite NaA crystallization, the results obtained indicated clearly that the history of Si-gel preparation played an important role in the nucleation and growth of zeolite NaA crystals and influenced their yield, size, and shape. Therefore, the optimization of Si-gel preparation procedure has to be considered as essential not only for the economy of the synthesis of NaA from RH, but also for the quality of the final product.  相似文献

Dialkyl imidazoline, alkyl pyridine and alkyl quinoline containing salts have been synthesized and used as new surfactants for cationic exchange of layered silicates, such as montmorillonite (MMT) in order to evaluate their role in the formation of intercalated MMT clays. The new salts have been characterized by FTIR, 1H, 13C‐NMR spectra. These compounds have been intercalated into the layers of MMT via a solution dispersion technique and the extent of the interlayer space expansion was confirmed by using wide angle X‐ray diffraction and FTIR spectra. Thermogravimetry analysis shows that these salts can be used in place of ammonium salts to produce organophilic clays with higher thermal stability than commercial alkyl ammonium‐treated MMT. Moreover, the results also show that the imidazolium‐modified clays have a greater thermal stability compared to the quinolinium and the pyridinium‐modified clay. © 2012 Wiley Periodicals, Inc. J. This paper describes studies on the reduction of birnessite thin layers electrodeposited onto a cheap transparent semiconductor substrate, tin dioxide (SnO2), in neutral sulphate solutions. A coupled approach based on electrochemical measurements (cyclic voltammetry and chronoamperometry) and X-ray diffraction (XRD) characterisation allowed us to give information about reduction mechanism of birnessite in presence or absence of Mn(II) in solution. In absence of Mn(II), birnessite is reduced only at low potential (E = −0.6 V) into β-MnOOH (feitkneichtite), if Qreduction is lower than Qsynthesis, and in amorphous Mn(II) compound, if Qreduction is equal to Qsynthesis. In presence of Mn(II) in solution, hausmannite (Mn3O4) was detected, and even at high potentials (E = 0.15 V). These results signify that a complex between Mn(III), coming from the reduction of Mn(IV) species, and Mn(II) present in solution, can be formed near the surface and leads to the formation of Mn3O4, a very resistive compound. These results are important for studies devoted to environmental applications, chemical sensors and also for energy storage.  相似文献

Adolescent idiopathic scoliosis (AIS) is a common orthopedic disorder of unknown etiology and pathogenesis. Melatonin and melatonin pathway dysfunction has been widely suspected to play an important role in the pathogenesis. Many different types of animal models have been developed to induce experimental scoliosis mimicking the pathoanatomical features of idiopathic scoliosis in human. The scoliosis deformity was believed to be induced by pinealectomy and mediated through the resulting melatonin-deficiency. However, the lack of upright mechanical spinal loading and inherent rotational instability of the curvature render the similarity of these models to the human counterparts questionable. Different concerns have been raised challenging the scientific validity and limitations of each model. The objectives of this review follow the logical need to re-examine and compare the relevance and appropriateness of each of the animal models that have been used for studying the etiopathogenesis of adolescent idiopathic scoliosis in human in the past 15 to 20 years.  相似文献

The Padé approximation problem in operator theory and its solution will be repeated briefly together with some properties of the Padé approximants. Effective methods for the solution of differential and integral equations are of great practical importance and there is a vast literature devoted to this subject.

Here a few methods, resulting from the use of Padé approximants in operator theory, are introduced and illustrated by means of some typical examples (initial value problems, boundary value problems, partial differential equations, nonlinear integral equations).

Among the new methods is an iterative scheme which we will call Halley's method and which proves to be very useful in the neighbourhood of singularities.

Well known methods such as Newton's and Chebyshev's method prove to be special cases of the class of iterative procedures.  相似文献
